Description: ACO2, also referred to as aconitate hydratase, citrate hydrolyase or aconitase, is an iron-sulfur hydrolyase that catalyzes the non-limiting interconversion of citrate and isocitrate in the tricarboxylic acid cycle. It is expressed in the mitochondria and maintains a citrate:isocitrate ratio of approximately 10:1. ACO2 contains a redox-sensitive iron-sulfur cluster that exists in two states: active (Fe4S4) and inactive (Fe3S4). ACO2 activity is dependent on the state of this cluster as well as the presence of two conserved cysteine residues. In normal prostate epithelial cells ACO2 activity is prevented due to the high levels of zinc inhibiting the enzyme. In these citrate-producing epithelial cells citrate oxidation is impaired allowing citrate to accumulate and exhibit a citrate:isocitrate ratio of approximately 30:1. In malignant prostate cells zinc is unable to accumulate, therefore ACO2 activity resumes and citrate is oxidized.

Description: ACO2(aconitate hydratase, mitochondrial) is also named as citrate hydro-lyase and belongs to the aconitase/IPM isomerase family. It plays a key function in cellular energy production, and loss of its activity has a major impact on cellular and organismal survival. Western blot shows two bands of 83 kDa and 40 kDa. The 40 kDa fragment decreases with age and oxidative stress, whereas other fragmentation products with molecular weights between 40 and 83 kDa increased with age and MnSOD(mitochondrial manganese superoxide dismutase) deficiency. Defects in ACO2 are the cause of infantile cerebellar-retinal degeneration (ICRD).

Description: ACO2 belongs to the aconitase/IPM isomerase family. It is an enzyme that catalyzes the interconversion of citrate to isocitrate via cis-aconitate in the second step of the TCA cycle. This protein is encoded in the nucleus and functions in the mitochondrion. It was found to be one of the mitochondrial matrix proteins that are preferentially degraded by the serine protease 15 (PRSS15), also known as Lon protease, after oxidative modification. The protein encoded by this gene belongs to the aconitase/IPM isomerase family. Description: Iron metabolism is essential for sustaining mammalian homeostasis. Iron uptake and distribution is a highly regulated process in mammalian cells that is monitored by two iron sensing proteins; iron regulatory protein-1 and -2 (IRP-1 and -2), also known as iron responsive element-binding protein-1 and -2 (IREBP-1 and -2) or aconitase 1 and 2. IRP-1 and IRP-2 are important soluble regulatory factors that mediate iron uptake and storage in mammalian cells. They are capable of either repressing translation or enhancing mRNA stability by associating with stem-loop motifs known as iron-responsive elements (IREs). IRPs respond to stress mediators, iron concentration and signaling factors, including nitrogen monoxide, cytokines and hydrogen peroxide.
