Description: CD8a (clone SK1) is a mouse monoclonal antibody derived from the CD8 alpha chain. This antibody belongs to the Biotium Choice list of select antibodies that have been validated and optimized in-house for optimal performance. The antibody is available conjugated to CF® Dyes and Astral™ tandem dyes. They are supplied in PBS, 0.1% BSA, 0.05% azide.

CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 beta chain (CD8 alpha/beta) or as a homodimer (CD8 alpha/alpha). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. CD8 is also found in littoral cell angioma of the spleen.

Description: This MAb specifically precipitates heterogeneous material of high MW, identified as perlecan, a major heparan-sulfate proteoglycan (HSPG) within all basement membranes and cell surfaces. It does not cross-react with laminin, fibronectin, or dermatran sulfate proteoglycan. Because of perlecan s strategic location and ability to store and protect growth factors, it has been strongly implicated in the control of tumor cell growth and metastatic behavior. Perlecan possesses angiogenic and growth-promoting attributes primarily by acting as a co-receptor for basic fibroblast growth factor (FGF-2). Suppression of perlecan causes substantial inhibition of neoplastic growth and neovascularization. Thus, perlecan is a potent inducer of neoplasm growth and angiogenesis in vivo and therapeutic interventions targeting this key modulator of tumor progression may improve neoplastic treatment.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®405S is a blue fluorescent dye (Ex/Em 404/431 nm) with superior brightness compared to other blue dyes; it is also compatible with super-resolution imaging by SIM. Note: Conjugates of blue fluorescent dyes are not recommended for detecting low abundance targets, because blue dyes have lower fluorescence and can give higher non-specific background than other dye colors.

PI4KII phosphorylates PtdIns at the D-4 position, an essential step in the biosynthesis of PtdInsPs. PI4K II is activated by detergent and inhibited by adenosine. Overexpression of PI4KII in COS-7 cells increases synthesis of PtdIns4P. Some cells overexpressing PI4KII have scattered or no perinuclear Golgi. Knockdown of PI4KII by RNA interference (RNAi) does not disrupt the Golgi, and some cells show expanded Golgi. RNAi reduces the Golgi level of PtdIns4P and blocks the association between AP1 and the trans-Golgi network. PI4KII RNAi had little effect on intra-Golgi trafficking, but it inhibited export to plasma membrane export by 35%. It has been proposed that PI4KII generates PtdIns4P-rich domains within the Golgi that specify docking of the AP1 coat machinery.

Description: CD40 is a 45-50 kDa type I membrane protein and a member of the tumor necrosis factor (TNF) receptor superfamily. CD40 is also known as pb50 or CDW40.  CD40 is expressed primarily by professional antigen-presenting cells and it is also expressed on non-immune cell types. The CD40 expressed by professional antigen-presenting cells plays a critical role in co-stimulation and antigen-presenting cell activation in T cell-dependent immune responses. Signals generated through CD40 in B cells are anti-apoptotic. These signals are also required for T cell-dependent B cell activation and proliferation, isotype switching, up-regulation of costimulatory receptors, germinal center formation and memory generation. CD40 signal transduction is initiated by binding trimeric CD40 ligand (CD40L) which is found on the surface of activated T cells.  CD40 engages several signalling pathways in B cells and these include NF-kB, the mitogen-activated protein kinases p38 and c-Jun N-terminal kinase. The 62 amino acid cytoplasmic domain (CD40c) of CD40 contains two linear TNF-receptor associated factor (TRAF) binding domains. The membrane proximal site of CD40c binds TRAF6 and the membrane distal site binds TRAF1, 2 and 3. CD40c is thought to mediate the CD40-dependant signalling pathways by recruitment of TRAF to this multimerized domain. CD40 ligation has also been shown to stimulate iNOS expression, and therefore NO production, by activating NF-kB in IFN-g treated microglial cells. NO derived from microglia has been implicated in the damage of myelin-producing oligodendrocytes in demyelinating disorders like multiple sclerosis (MS) and neuronal death during Alzheimer’s disease and brain trauma. Increased CD40-CD40 ligation has been implicated in the pathogenesis of several autoimmune inflammatory diseases, such as MS, arthritis and insulin-dependant diabetes.
