Description: CD273 (B7-DC; PD-L2) is a type I surface molecule with homology to CD80, CD86, CD274. It is expressed primarily by dendritic cells and provides a stimulatory signal to CD279 (PD-1; programmed death molecule) which has an important immunoregulatory role by downregulating the T cell response. CD273 binds to CD279 (PD-1) with a 2-6 fold higher affinity than CD274.

Description: TIP60 is the catalytic subunit of the NuA4 histone acetyltransferase complex which is involved in transcriptional activation of selected genes principally by acetylation of nucleosomal histones H4 and H2A. This modification may both alter nucleosome-DNA interactions and promote interaction of the modified histones with other proteins which positively regulate transcription. This complex is required for the activation of transcriptional programs associated with oncogene and proto-oncogene mediated growth induction, tumor suppressor mediated growth arrest and replicative senescence, apoptosis and DNA repair. NuA4 may also play a direct role in DNA repair when recruited to sites of DNA damage. TIP60 directly acetylates and activates ataxia-telangiectasia mutated (ATM). In case of HIV-1 infection, interaction with the viral Tat protein leads to KAT5 polyubiquitination and targets it to degradation.

Description: Recombinant human soluble vascular endothelial growth factor receptor-1 (sVEGFR-1) is the naturally occurring form and is a glycosylated monomeric protein. The biological function of sVEGFR-1 seems to be an endogenous regulator of angiogenesis, binding VEGF with the same affinity as the full-length receptor. VEGFR-1 is a tyrosine-protein kinase that acts as a cell-surface receptor for VEGFA, VEGFB and PGF, and plays an essential role in the development of embryonic vasculature, the regulation of angiogenesis, cell survival, cell migration, macrophage function, chemotaxis and cancer cell invasion. It may play an essential role as a negative regulator of embryonic angiogenesis by inhibiting excessive proliferation of endothelial cells. It can promote endothelial cell proliferation, survival and angiogenesis in adulthood. Its function in promoting cell proliferation seems to be cell-type specific. Promotes PGF-mediated proliferation of endothelial cells, proliferation of some types of cancer cells, but does not promote proliferation of normal fibroblasts (in vitro). It has a very high affinity for VEGFA and relatively low protein kinase activity. It may function as a negative regulator of VEGFA signaling by limiting the amount of free VEGFA and preventing its binding to KDR. Modulates KDR signaling by forming heterodimers with KDR. Ligand binding leads to the activation of several signaling cascades. Activation of phospholipase C-gamma (PLCG) leads to the production of the cellular signaling molecules diacylglycerol and inositol 1,4,5-trisphosphate and the activation of protein kinase C. Mediates phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase, leading to activation of phosphatidylinositol kinase and the downstream signaling pathway. Mediates activation of MAPK1/ERK2, MAPK3/ERK1 and the MAP kinase signaling pathway, as well as of the AKT1 signaling pathway. Phosphorylates SRC and YES1 and may also phosphorylate CBL.

Description: Bim belongs to the Bcl-2 protein family. Bcl-2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. This protein contains a Bcl-2 homology domain 3 (BH3). It has been shown to interact with other members of the Bcl-2 protein family (BCL2, BCL2L1/BCL-X(L) and MCL1) and to act as an apoptotic activator, inducing apoptosis and anoikis. Several variants have been identified. The isoforms vary in cytotoxicity with isoform BimS being the most potent and isoform BimEL being the least potent.

Description: LYVE-1 has been identified as a major receptor for HA (extracellular matrix glycosaminoglycan hyaluronan) on the lymph vessel wall. Like CD44, the LYVE-1 molecule binds both soluble and immobilized HA. However, unlike CD44, the LYVE-1 molecule co-localizes with HA on the luminal face of the lymph vessel wall and is completely absent from blood vessels. Hence, LYVE-1 is the first lymph-specific HA receptor to be characterized and is a uniquely powerful marker for lymph vessels themselves.

Description: Irisin is a recently described exercise and PGC1-alpha-induced myokine secreted by skeletal muscle in mice and humans. Irisin has been shown to induce browning of white adipose tissue, but this role has been questioned and a new function in the control of mass and strength of cortical bone has been reported. The membrane fibronectin type III domain containing protein 5 (FNDC5) can be cleaved by an unknown protease to release the Irisin protein (aa 32-143 for human) and potentially also the FNDC5 (extracellular domain, aa 32-153 for human).

Description: Angiopoietin-1 (Ang-1) and Angiopoietin-2 (Ang-2) are closely related secreted ligands which bind with similar affinity to Tie-2. Tie-2 and angiopoietins have been shown to play critical roles in embryogenic angiogenesis and in maintaining the integrity of the adult vasculature. Ang-1 activates Tie-2 signaling on endothelial cells to promote chemotaxis, cell survival, cell sprouting, vessel growth and stabilization. Ang-2 has been identified as a secreted protein ligand of Tie-2 and has alternatively been reported to be an antagonist for Ang-1 induced Tie-2 signaling as well as an agonist for Tie-2 signaling, depending on the cell context. anti-Angiopoietin-2, mAb (rec.) (blocking) (Angy-2-1) is an antibody developed by antibody phage display technology using a human naive antibody gene library. These libraries consist of scFv (single chain fragment variable) composed of VH (variable domain of the human immunoglobulin heavy chain) and VL (variable domain of the human immunoglobulin light chain) connected by a polypeptide linker. The antibody fragments are displayed on the surface of filamentous bacteriophage (M13). This scFv was selected by affinity selection on antigen in a process termed panning. Multiple rounds of panning are performed to enrich for antigen-specific scFv-phage. Monoclonal antibodies are subsequently identified by screening after each round of selection. The selected monoclonal scFv is cloned into an appropriate vector containing a Fc portion of interest and then produced in mammalian cells to generate an IgG like scFv-Fc fusion protein.

Description: Inflammatory caspases (also known as group I caspases) are encoded by three main genes in humans, caspase-1, caspase-4 and caspase-5 and three main genes in mouse, caspase-1, caspase-11 and caspase-12. Caspase-11 is the murine orthologue of human caspase-4 and -5. Murine caspase-11 is a poorly characterized member of the caspase-1 subfamily. Caspase-11-deficient embryonic fibroblasts are resistant to apoptosis induced by ectopic expression of caspase-1, suggesting that caspase-11 is an upstream activator of caspase-1. Unlike caspase-1, the expression of caspase-11 is LPS-inducible and it is reasonable to postulate that other members of the family are regulated at the transcriptional or translational level by extracellular stimuli. Recently caspase-11 has been shown to be required for activation of the Nlrp3 inflammasome upon E. coli, V. cholerae and C. rodentium infection. The upstream activators of caspase-11 are TLR4 and TRIF, that modulate enteropathogen-induced inflammasome activation by promoting caspase-11 expression and activation. Formation of ASC foci (specks), a measure of NLRP3/ASC complex formation, requires caspase-11 but not caspase-1 indicating that caspase-11 acts upstream of the NLRP3/ASC complex.

Description: The most prominent members of the interleukin-1 (IL-1) superfamily are IL-1alpha and IL-1beta. They lack a signal peptide and are secreted by an unconventional, endoplasmic reticulum-Golgi-independent mechanism. IL-1alpha was reported to be more widely and constitutively expressed and has intracellular functions, but also acts locally in a membrane-bound form by activating IL-1R1. Additionally, passive release of IL-1alpha upon cell death can trigger a sterile inflammatory response to dying cells. The cleavage of IL-1alpha is not mediated by caspase-1 and is not required for binding to IL-1R1. Recently it has been observed that all activators of the inflammasome NLRP3/NALP3 induce the simultaneous secretion of IL-1alpha and IL-1beta. Although most activators fully rely on the inflammasome for IL-1alpha secretion, some induce the processing and secretion of IL-1alpha in an inflammasome-independent manner.

Description: The transmembrane and secreted protein delta-like 1 homolog (DLK1; Protein delta homolog 1; Preadipocyte factor 1; Pref-1) belongs to the EGF-like family. It can be cleaved by a tumor necrosis factor-alpha converting enzyme (TACE) to release a soluble form. DLK1 plays important roles in regulating cell differentiation such as adipogenesis and osteogenesis. Membrane-bound DLK1 promotes hypertrophic myotube formation, whereas soluble DLK1 inhibits myotube formation. The soluble form of DLK1 inhibits preadipocyte differentiation, while membrane DLK1 represses preadipocyte proliferation. DLK1 is also associated with tumor invasion.

Description: Reactive oxygen species (ROS) are involved in lipid peroxidation inside the living bodies, and membrane lipids are one of the major targets of ROS. During the peroxidation process, a variery of aldehydes are formed. 4-hydroxy-2-nonenal (4-HNE) is an alpha, beta unsaturated aldehyde that can be formed by peroxidation of omega-6 unsaturated fatty acids such as linoleic acid and arachidonic acid. Especially, 4-HNE formed inside living body is reported to originate from phospholipid-bound arachidonic acid. 4-HNE may be one of the most reliable biomarker of lipid peroxidation.
