Description: The Notch ligand delta-like protein 4 (DLL4) is expressed highly and selectively within the arterial endothelium and has been shown to function as a ligand for Notch1 and Notch4. It is induced by VEGF as a negative feedback regulator and acts to prevent overexuberant angiogenic sprouting, promoting the timely formation of a well differentiated vascular network. DLL4-Notch1 signaling regulates the formation of appropriate numbers of tip cells to control vessel sprouting and branching in the mouse retina.

Description: Mouse vascular endothelial growth factor 164 (VEGF 164) is produced as a homodimer. It binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin. It is a specific mitogen for vascular endothelial cells and a strong angiogenic factor in vivo. In addition to its action as a mitogen it is a potent vascular permeability factor (VPF) in vivo and is also a chemoattractant for monocytes and endothelial cells. It induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and induces permeabilization of blood vessels. It may play a role in increasing vascular permeability during lactation, when increased transport of molecules from the blood is required for efficient milk protein synthesis.

Description: Thymidineglycol (TG) is one of the major oxidation products of DNA. Thymidine (T) can be damaged by oxidative stress such as radiation and energy metabolism. Two different pathways to form TG have been suggested. Deoxythymidine in DNA is directly oxidised by hydroxy radical, to form TG. TG can be also formed through an intermediate thymidine chlorohydrin, which is derived from hypochlorous acid (HOCl) from neutrophil myeloperoxidase. Thymidineglycol is derived from DNA, not from RNA. TG is the oxidative stress marker secific for DNA damage. In comparison with 8-hydroxy-2'-deoxyguanosine (8-OHdG), which is the best known DNA oxidation product it was shown that in a lipopolysaccharide (LPS)-treated mouse liver, 8-OHdG can be stained within 24 hours after LPS treatment. TG can be detected within 6 hours, and remains at least for 72 hours.

Description: Apaf-1 is a cytoplasmic protein that forms one of the central hubs in the apoptosis regulatory network. It contains (from the N terminal) a caspase recruitment domain (CARD), an ATPase domain (NB-ARC), few short helical domains and then several copies of the WD40 repeat domain. Upon binding cytochrome c and dATP, this protein forms an oligomeric apoptosome, mediates the cytochrome c-dependent autocatalytic activation of pro-caspase-9 (Apaf-3), leading to the activation of caspase-3 and apoptosis.

Description: HMGB1 was originally discovered as an essential DNA-binding protein for regulating p53, NF-kappaB and other important proteins. It is secreted from activated dentric cells, macrophage and nectrotic cells, and acts as a ligand for RAGE, TLR-2 and TLR-4 expressed on surrounding cells. As a result, HMGB1 activates Rac, CDC42 and NF-kappaB inducing localized innate immunity of damaged tissue, tissue regeneration by recruitment of stem cells and hemostasis by induction of tissue factor expression. HMGB1 is also causative agent of various diseases as it causes localized inflammation such as arteriosclerosis, chronic rheumatoid arthritis and nephritis.

Description: Progranulin (PGRN) is a widely expressed pluripotent growth factor which plays a role in processes such as development, wound repair and inflammation by activating signaling cascades that control cell cycle progression and cell motility. Its function in the central nervous system is of interest, as mutations in the PGRN gene were found in cases of frontotemporal degeneration (FTLD). In addition, PGRN has also been linked to tumorigenesis. Progranulin is a biomarker for FTLD, other types of Alzheimer‘s Disease (AD) and potentially for MCI (Mild Cognitive Impairment). Additionally, PGRN is described as a new ligand of TNF receptors and a potential therapeutic against inflammatory disease like arthritis.

Description: Ubiquitin is a 76 amino acid (aa) protein that is ubiquitously expressed in all eukaryotic organisms. ubiquitin is highly conserved with 96% aa sequence identity shared between human and yeast ubiquitin, and 100% aa sequence identity shared between human and mouse ubiquitin. In mammals, four ubiquitin genes encode for two ubiquitin-ribosomal fusion proteins and two poly-ubiquitin proteins. Cleavage of the ubiquitin precursors by deubiquitinating enzymes gives rise to identical ubiquitin monomers each with a predicted molecular weight of 8.6 kDa. Conjugation of ubiquitin to target proteins involves the formation of an isopeptide bond between the C-terminal glycine residue of ubiquitin and a lysine residue in the target protein. This process of conjugation, referred to as ubiquitination or ubiquitylation, is a multi-step process that requires three enzymes: a ubiquitin-activating (E1) enzyme, a ubiquitin-conjugating (E2) enzyme, and a ubiquitin ligase (E3). ubiquitination is classically recognized as a mechanism to target proteins for degradation and as a result, ubiquitin was originally named ATP-dependent Proteolysis Factor 1 (APF-1). In addition to protein degradation, ubiquitination has been shown to mediate a variety of biological processes such as signal transduction, endocytosis, and post-endocytic sorting.

Description: PARP-1 (ARTD1) is involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks. PARP-1 positively regulates the transcription of MTUS1 and negatively regulates the transcription of MTUS2/TIP150. It forms a complex with EEF1A1 and TXK that acts as a T-helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFN-gamma to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production.

Description: Cold-inducible RNA-binding protein (CIRP) is from the family of cold shock proteins that plays a protective role in the genotoxic and cold stress response. CIRBP functions as an RNA chaperone to facilitate translation and to stabilize transcripts of genes involved in cell survival. CIRP (human) is a 172-aa nuclear protein consisting of one amino-terminal consensus sequence RNA-binding domain and one carboxyl-terminal glycine-rich domain. When overexpressed, CIRP promotes assembly of stress granules (SGs). CIRP is constitutively expressed at low levels in various tissues becoming up-regulated during mild hypothermia as well as exposure to UV irradiation and hypoxia. CIRP is also found extracellularly, where it acts as a proinflammatory mediator causing deleterious effects during hemorrhagic and septic shock.

Description: Angiopoietin-1 (Ang-1) and Angiopoietin-2 (Ang-2) are closely related secreted ligands which bind with similar affinity to Tie-2. Tie-2 and angiopoietins have been shown to play critical roles in embryogenic angiogenesis and in maintaining the integrity of the adult vasculature. Ang-1 activates Tie-2 signaling on endothelial cells to promote chemotaxis, cell survival, cell sprouting, vessel growth and stabilization. Ang-2 has been identified as a secreted protein ligand of Tie-2 and has alternatively been reported to be an antagonist for Ang-1 induced Tie-2 signaling as well as an agonist for Tie-2 signaling, depending on the cell context.

Description: Netrin-1 controls guidance of CNS commissural axons and peripheral motor axons. Its association with either DCC or some UNC5 receptors will lead to axon attraction or repulsion, respectively. It also serve as a survival factor via its association with its receptors which prevent the initiation of apoptosis. Netrin-1 is also Involved in tumorigenesis by regulating apoptosis. Netrin-1 promotes atherosclerosis by retaining macrophages in the artery wall. Netrin-1 also governs induced pluripotent stem cell (iPS) formation and improves reprogramming efficiency of human and mouse somatic cells by limiting apoptosis mediated by Netrin-1 receptors DCC or UNC5b.

Description: Interleukin-23 receptor (IL-23R), is a member of the type I cytokine receptor family, type 2 subfamily. It is expressed by monocytes, Th1, Th0, NK and dendritic cells. Isoform 1 is specifically expressed in NK cells. IL-23 functions in innate and adaptive immunity and may participate in acute response to infection in peripheral tissues. IL-23 may be responsible for autoimmune inflammatory diseases and be important for tumorigenesis. IL-23R associates with IL-12RB1 to form the interleukin-23 receptor. It binds IL-23 and mediates T cells, NK cells and possibly certain macrophage/myeloid cells stimulation probably through activation of the Jak-Stat signaling cascade. Genetic variations in IL-23R are associated with inflammatory bowel disease type 17 (IBD17). IBD17 is a chronic, relapsing inflammation of the gastrointestinal tract with a complex etiology. Genetic variations in IL-23R also can cause susceptibility to psoriasis type 7.
