Description: Hsp40/Hdj1 is a cytosolic co-chaperone belonging to the class DnaJ, named after its homolog in E. coli. Hsp40 functions in protein folding by binding nascent peptides and unfolded substrates and facilitating substrate interaction with Hsp70. Binding of Hsp40 to Hsp70 increases Hsp70 ATP hydrolysis and substrate binding, which is reversed by the action of nucleotide exchange factors (GrpE in E. coli). Repeated cycles of Hsp40/Hsp70 peptide binding and ATP hydrolysis prevent premature folding and aggregation, promoting protein maturation throughout the cell.

Description: Nuclear factor kappa-B (NFk) B) functions as a sequence specific transcriptional activator that binds to the intronic enhancer of kappa light chain gene in B lymphocytes. NF-kB is a heterodimer consisting of a 50 kDa DNA binding subunit (p50) and a 65 kDa transactivation subunit (p65/RelA) (ref. 3), both of which exhibit sequence homology to the protooncogene c-Rel. p50 has an isoform called p49/p52, and both proteins derive from the amino-terminal of precursor protein p105 and p100, respectively. The p50/p65 heterodimer remains in inactive form in the cytosol as a complex with its inhibitor, IkB. Upon stimulation of cells by a variety of stimuli, including lipopolysaccharide (LPS), pro-inflammatory cytokines like interleukin-1 and tumor necrosis factor, and viral infection, IkB becomes phosphorylated and degraded by the proteosome. The active NF-kB heterodimer translocates into the nucleus and induces gene expression.

Description: Osteopontin (OPN) is an acidic extracellular matrix cell adhesion protein that is relatively abundant not only in bone matrix, plasma, urine, and milk, but is also found in malignant and atherogenic tissues. Phosphorylation, glycosylation and calcium modifications allow intact and fragmented Osteopontin to direct a variety of diverse responses including tissue remodeling, inflammation and cell survival. Plasma Osteopontin has been shown to provide prognostic information in breast, prostate, colon, and lung cancers as well as metastatic carcinomas. The notable presence of Osteopontin in a variety of tumors is strongly correlated to pathological stage, suggesting its critical role in tumor invasiveness, progression and metastasis. In addition, Osteopontin inhibits inducible nitric oxide synthase activity, thereby protecting tumor cells from NO-mediated macrophage cytotoxic attack. Osteopontin is found in atherosclerotic plaques and may drive a number of diabetic vascular pathologies.

Description: α-Actinin-4 is an actin-bundling protein of ~100kDa that is associated with cell motility, endocytosis, and cancer invasion. The α-actinin family comprises two non-muscle isoforms (α-actinin-1 and -4) and two skeletal muscle isoforms (α-actinin-2 and -3), with α-actinin-2 being also expressed in cardiac muscle. While α-actinin-4 is almost ubiquitously expressed, particularly high concentrations are found in glomeruli. On the subcellular level it is associated with actin stress fibers, but in certain cells it also localizes to the nucleus. Mutations in the α-actinin-4 gene cause an autosomal-dominant form of familial focal segmental glomerulosclerosis (FSGS), which is thought to result from a defect in glomerular podocyte function. A point mutation in the α-actinin-4 gene was found to generate an antigenic peptide that is recognized by autologous cytolytic T lymphocytes (CTL) on a human lung carcinoma. α-actinin-4 interacts with a variety of proteins, including the ring finger protein BERP, the PDZ-LIM protein CLP-36, the hemidesmosomal and cell-cell contact protein BP180, and the tight junction protein MAGI-1. Moreover, α-actinin-4 forms a ternary complex with Ca2+/ Calmodulin-dependent protein kinase II and Densin-180, a protein of postsynaptic densities in CNS neurons. Ca2+-dependent association of α-actinin-4 with E3KARP is required for Ca2+-dependent inhibition of the Na+/ H+exchanger 3 (NHE3).

Description: VASP (vasodilator stimulated phosphoprotein) is a proline-rich protein substrate of cAMP- and cGMP-dependent protein kinases. Phosphorylation of VASP at Ser-157 causes a mobility shift in SDS gel electrophoresis from 46 to 50 kDa, which has been used as a convenient marker to monitor cyclic nucleotide-dependent protein kinase activity. VASP is the founding member of the Ena-VASP protein family, comprising the Drosophila protein Enabled (Ena), its mouse homologue Mena (mammalian Enabled), and mouse EVL (Ena-VASP-like protein). With these proteins VASP shares a conserved overall domain organization:
a) the conserved N-terminal Ena-VASP homology domain 1 (EVH1), which mediates binding to a proline-rich motif
b) a more divergent proline-rich central domain (which is responsible for profilin binding)
c) a conserved C-terminal EVH2 domain.

VASP is expressed in a variety of mammalian cell types and tissues. In cultured cells, VASP is associated with focal adhesions, cell-cell contacts, microfilaments, and highly dynamic membrane regions. From in vitro binding data VASP has been suggested to link profilin to zyxin, vinculin, and the Listeria spp. surface protein ActA, respectively. Functional evidence indicates that VASP is a crucial factor involved in the enhancement of actin filament formation and the actin-dependent motility of intracellular bacterial pathogens.
