Description: The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules. The TRL family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition. Ten human homologs of TLRs (TLR1-10) have been described. TLR8 gene contains three exons, two of which have coding function. TLR8 cDNA codes for a protein of approximate molecular weight of 120kDa.

Description: Leukotrienes are metabolic products of arachidonic acid formed by the action of 5-lipoxygenase (5-LO), with the principal products being leukotrienes C4 (LTC4) and B4 (LTB4). LTB4 is a dihydroxy-leukotriene produced in a variety of cells including eosinophils, neutrophils, platelets, and vascular cells, and is a potent leukocyte chemoattractant known to regulate leukocyte activation, cytokine secretion, inflammatory gene transcription, and IgE synthesis. As such, LTB4 has been implicated in a number of disease pathologies including atherosclerosis, asthma, psoriasis, and inflammatory bowel disease. The many actions of LTB4 are associated with two identified mammalian receptors, BLT1 and BLT2. BLT1 is a high-affinity LTB4 receptor expressed on peripheral leukocytes, while BLT2 is a more ubiquitously expressed low affinity LTB4 receptor whose function is not well understood. The BLT1 receptor is transiently expressed by antigen-primed T-cells in humans, and its expression is known to play a critical role in collagen-induced arthritis in mice.

Description: Fibronectins are high molecular weight, disulphide-linked, dimeric cell adhesion glycoproteins found in basement membranes and in the interstitial connective tissue matrix. A single fibronectin gene is subject to alternative splicing in a cell-type-, development- and age-regulated manner which gives rise to multiple molecular forms. In addition to their prominent role in adhesion, fibronectins have been reported to mediate various aspects of cellular interaction, including migration during development and wound-healing, haemostasis, and the regulation of cell growth and differentiation. Cellular fibronectins (cFn) are found in low amounts in normal human plasma and tissues, but they are abundant in the plasma of carcinoma patients and in the stroma of various carcinomas. In contrast, a soluble form of fibronectin produced by hepatocytes is readily detectable in plasma and becomes deposited in pericellular matrices and within tissues. This form of fibronectin, referred to as ‘plasma fibronectin’ (pFn), differs from cFn by the absence of an amino acid sequence, known as extra domain A1.

Description: Bad is an important regulatory protein involved in the control of cell survival and apoptosis, and is a member of the BH3-only family of proteins such as Bim and Bid that share homology with Bcl-2 only in the BH3 domain. Phosphorylated Bad is sequestered in the cytosol by 14-3-3 proteins under normal conditions, but is dephosphorylated and activated following apoptotic signals. Apoptotic stimuli induce Bad dephosphorylation by a number of phosphatases (e.g., calcineurin) and recruitment to the mitochondria, where Bad heterodimerizes with and inhibits prosurvival proteins of the Bcl family. In contrast, survival signals lead to phosphorylation of Bad at multiple serine residues, including phosphorylation at Ser112 and Ser136 which induce association with 14-3-3 proteins. Phosphorylation at Ser112 and Ser136 is required for a third phosphorylation event at Ser155, which induces dissociation of Bad/Bcl heterodimers to promote cell survival. This antibody specifically binds the Bad protein, but does not recognize Bad/Bcl-2 heterodimers in immunoprecipitation assays.
