Catalog Number: (CAPI22360)
Supplier: Thermo Scientific
Description: Thermo Scientific Pierce SMCC is a hetero-bifunctional crosslinker that contain N-hydroxysuccinimide (NHS) ester and maleimide groups that allow covalent conjugation of amine- and sulfhydryl-containing molecules. NHS esters react with primary amines at pH 7–9 to form amide bonds, while maleimides react with sulfhydryl groups at pH 6.5–7.5 to form stable thioether bonds. In aqueous solutions, NHS ester hydrolytic degradation is a competing reaction whose rate increases with pH. The maleimide group is more stable than the NHS-ester group, but will slowly hydrolyze and lose its reaction specificity for sulfhydryls at pH values > 7.5. For these reasons, conjugations with these crosslinkers are usually performed at pH 7.2–7.5, with the NHS ester (amine-targeted) reacted before or simultaneous with the maleimide (sulfhydryl-targeted) reaction.

Supplier: Anaspec Inc
Description: Tetramethylrhodamine (TMR) is one of the fluorophores most often used for preparing peptide, protein, nucleotide and nucleic acid conjugates, especially fluorescent antibodies and avidin derivatives used in immunochemistry. TMR dyes have been widely used as acceptors for FAM fluorophores in a variety of FRET studies. The succinimidyl esters of 5-TAMRA, 6-TAMRA or the mixed isomers are the primary labeling reagents.

Supplier: Thermo Scientific
Description: Thermo Scientific Pierce LC-SDA (NHS-LC-Diazirine) combines proven NHS-ester and diazirine-based photoreaction chemistries with conjugate amine-containing molecules with nearly any other functional group via long-wave UV-light activation. A 12.5Å spacer arm separates the two photoreactive groups.

Supplier: Thermo Scientific
Description: Thermo Scientific Pierce DSP (Lomant's Reagent) is a water-insoluble, homo-bifunctional N-hydroxysuccimide ester (NHS ester) crosslinker that is thiol-cleavable, primary amine-reactive, and useful for many applications. DSP contains an amine-reactive NHS ester at each end of an 8-carbon spacer arm. NHS esters react with primary amines at pH 7–9 to form stable amide bonds and releasing N-hydroxy-succinimide. Proteins, including antibodies, generally have several primary amines in the side chain of lysine (K) residues and the N-terminus of each polypeptide that are available as targets for NHS ester crosslinking reagents. DSP is non-sulfonated and insoluble in water, so it must first be dissolved in an organic solvent and then added to the aqueous reaction mixture. Because DSP does not possess a charged group, it is lipophilic and membrane-permeable and so useful for intracellular and intramembrane conjugation. A sulfonated analog of DSP (DTTSP) is water soluble. DSS, the non-cleavable analog of the DSP crosslinker is also available for applications that require a stable spacer arm that cannot be cleaved in the presence of reducing agents.

Catalog Number: (89139-516)
Supplier: Biotium
Description: 5-(and-6)-Carboxyrhodamine 110, SE (5-(and-6)-Carboxyrhodamine 110, succinimidyl ester, mixed isomers), also known as Carboxyrhodamine 110-NHS, is a superior green fluorescent dye that readily reacts with primary or secondary amines. For many applications, the dye is preferred over 5-(and-6)-carboxyfluorescein succinimidyl ester (5(6)-FAM) or FITC because of its exceptional photostability and fluorescence insensitivity to pH (4-9).

Description: 5-FAM SE is an amine-reactive form of 5-carboxyfluorescein single isomer.

Supplier: Anaspec Inc
Description: Although FITC reagents have been more often used to prepare fluoresceinated bioconjugates, the low stability of FITC bioconjugates makes some researchers use amine-reactive succinimidyl esters of carboxyfluorescein (commonly called FAM) in bioconjugations. FAM reagents give carboxamides that are more resistant to hydrolysis. We have shown that FAM reagents require less stringent reaction conditions and give better conjugation yields, and the resulted conjugates have superior stability. We noted that FITC labeled peptides tend to deteriorate more quickly than the corresponding FAM conjugates.
