Description: Cyclophilin C is an enzyme (EC 5.2.1.8) found in both prokaryotes and eukaryotes that interconverts the cis and trans isomers of peptide bonds with the amino acid proline. Proline has an unusually conformationally restrained peptide bond due to its cyclic structure with its side chain bonded to its secondary amine nitrogen. Most amino acids have a strong energetic preference for the trans peptide bond conformation due to steric hindrance, but prolines unusual structure stabilizes the cis form so that both isomers are populated under biologically relevant conditions. Proteins with prolyl isomerase activity include cyclophilin, FKBPs, and parvulin, although larger proteins can also contain prolyl isomerase domains.

Description: Alpha-methyacyl-CoA racemase (AMCR), is an enzyme invloved in beta oxidation of branched chain fatty acids and bile salt intermediates, and has recently been identified as a neomarker for prostate cancer, where it is over expressed. Several different isoforms have been reported that are produced either by extensinve alternatve splicing of 5 exons or by use of alternate initiation codons. At least 2 different transcripts, each derived from the 5 exons, have been reported, AMCR I and AMCR II. AMCR I is the most abundant form and enclodes for a 382 amino acid protein (42kDa). The other isoform AMCR II exhibits significnat homolgy to fumerate hydratase and encoes a 288 amino acid protein (32 kDa). Several other variants of IA and IIA isoforms are charcterized recently. The variants lack exon 3 and are designated as IB and IIB. In prostate tumor tissues that over express AMCR, both the A and B forms are over-expressed.

Description: AMACR (alpha-methylacyl-CoA racemase) has been recently described as prostate cancer-specific gene that encodes a protein involved in the beta-oxidation of branched chain fatty acids. Expression of AMACR protein is found in prostatic adenocarcinoma but not in benign prostatic tissue. It stains premalignant lesions of prostate: high-grade prostatic intraepithelial neoplasia (PIN) and atypical adenomatous hyperplasia. AMACR can be used as a positive marker for PIN. Defects in AMACR are the cause of congenital bile acid synthesis defect type 4 (CBAS4); also known as cholestasis, intrahepatic, with defective conversion of trihydroxycoprostanic acid to cholic acid or trihydroxycoprostanic acid in bile. Clinical features include neonatal jaundice, intrahepatic cholestasis, bile duct deficiency and absence of cholic acid from bile.

Description: Processes such as transcription, repair and replication that require efficient DNA recognition are dependent on modulation of chromatin structure. Chromatin relaxation is a critical event that occurs during DNA repair and is associated with the negatively charged polymer of adenosine 5'-diphosphate (ADP)-ribose (PAR). PAR is synthesized from nicotinamide adenine dinucleotide (NAD+) by the poly(ADP-ribose) polymerase protein family (PARPs), of which PARP-1 (and to a lesser extent PARP-2) respond to DNA-strand breaks. PARP-1 is selectively activated by DNA strand breaks to catalyze the addition of long branched chains of PAR to a variety of nuclear proteins, most notably PARP itself. The amount of PAR formed in living cells with DNA damage is commensurate with the extent of the damage. Under DNA damage conditions, PAR undergoes a rapid turnover, with a half-life in the range of minutes, as PAR is rapidly hydrolyzed and converted to free ADP-ribose by the enzyme poly(ADP-ribose)glycohydrolase (PARG). PAR regulates not only cell survival and cell-death programmes, but also an increasing number of other biological functions with which novel members of the PARP family have been associated. These include transcriptional regulation, cell division, intracellular trafficking, inflammation and energy metabolism.
