Description: The Hsp70 family of heat shock protiens contains multiple homologs ranging in size from 66-78 kDa, and are the eukaryotic equivalents of the bacterial DnaK. The most studied Hsp70 members include the cytosolic stress-induced Hsp70 (Hsp72), the constitutive cytosolic Hsc70 (Hsp73), and the ER-localized BiP (Grp78). Hsp70 family members contain highly conserved N-terminal ATP-ase and C-terminal protein binding domains. Binding of peptide to Hsp70 is assisted by Hsp40, and stimulates the inherent ATPase activity of Hsp70, facilitating ATP hydrolysis and enhanced peptide binding. Hsp70 nucleotide exchange and substrate binding coordinates the folding of newly synthesized proteins, the re-folding of misfolded or denatured proteins, coordinates trafficking of proteins across cellular membranes, inhibits protein aggregation, and targets the degradation of proteins via the proteasomal pathway.

Description: The rate limiting enzyme in heme catabolism, heme oxygenase, was first described in 1968 and was thought to be a species of cytochrome P450. However, subsequent work showed the enzyme to be a protein distinct from the P450 family and capable of catalysing the degradation of heme to biliverdin and iron, with the concomitant release of carbon monoxide. This enzyme activity is highly conserved throughout the Animal Kingdom and has been identified in algae and plants as well as in all higher species. Heme oxygenase-1 (HO-1) was first purified to homogeneity from rat liver and pig spleen and shown to have a molecular weight of 32kDa. Increased expression of HO-1 can be induced by a large number of structurally unrelated pharmacological and other agents, including cytokines and oxidants, as well as by heat shock and other forms of cellular stress. Baseline levels of HO-1 expression are high in certain tissues, including spleen and liver, but enriched enzyme preparations are recommended for experimental analysis. Recent data have demonstrated a cytoprotective role for HO-1, preventing cell death by modulating intracellular iron levels.

Description: The β-amyloid peptide (β A4), proteolytically released from the amyloid precursor protein (APP), provides the principal component of senile plaques in Alzheimer's disease. Description: Endothelins are vasoactive peptides that exist in three forms (ET-1, ET-2, and ET-3), and function in the maintenance of vascular tone via two identified seven transmembrane-spanning domain-containing G protein-coupled receptors (GPCR), endothelin A (ETA) and endothelin B (ETB). The human ETA receptor is primarily expressed in vascular smooth muscle cells, including those of the heart (also in myocytes), lung, and brain, and its stimulation generally results in vasoconstriction. Activation of ETA receptors can be stimulated by binding of ET-1 and ET-2, but not ET-3. triggering the release of vasorelaxive factors such as nitric oxide (NO) and prostanoids from endothelial cells. Human ETB receptors are primarily expressed in endothelial cells lining the vessel walls of the lungs, heart, and brain, and in contrast to ETA receptors, stimulation generally results in transient vasodilation. Activation of ETB receptors can be stimulated by binding of all three endothelins, triggering the release of vasorelaxive factors such as nitric oxide (NO) and prostanoids from endothelial cells. Both ETA and ETB receptors signal primarily via coupling to Gq/11 alpha subunits, although Gi/o and Gs coupling are also established mechanisms of endothelin signaling, resulting in stimulation of phospholipases C, A2, and D.
