Description: WIPI1 Antibody: WIPI1 (WD repeat domain, phosphoinositide interacting-1), also known as WIPI1, ATG18 or WIPI49, is thought to play a role in autophagy and may regulate protein trafficking in certain recycling pathways. It contains three WD repeats and has a 7-bladed propeller structure with a conserved motif that facilitates its interaction with other proteins. WIPI1 localizes to cytoplasmic vesicles, endosomes, clathrin-coated vesicles and the trans-Golgi network. It is ubiquitously expressed with highest expression in heart, testis, placenta, pancreas and skeletal muscle. WIPI1 is upregulated in a variety of tumors, suggesting a role in carcinogenesis.

Description: The Sec1/Munc18 (SM) family of proteins regulate intracellular trafficking through interactions with individual SNARE (soluble N-ethylmeleimide-sensitive factor attachment protein receptor) proteins and assembled SNARE complexes. The SM protein VPS45 (Vacuolar protein sorting-associated protein 45) has a calculated molecular mass of 65 kDa and shares 38% identity with yeast Vps45 which is required for the formation of yeast endosomal SNARE complexes and is thus essential for traffic through the endosomal system. VPS45 may play a role in vesicle-mediated protein trafficking from the Golgi stack through the trans-Golgi network.

Description: Complexins are soluble proteins that regulate the activity of soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) complexes necessary for vesicle fusion. Neuronal specific complexin 1 (CPLX1) has inhibitory and stimulatory effects on exocytosis by clamping trans-SNARE complexes in a prefusion state and promoting conformational changes to facilitate membrane fusion following cell stimulation. Complexin2 (CPLX2) is a pre-synaptic protein believed to regulate neurotransmitter release from pre-synaptic terminals, it is downregulated in schizophrenic patients suffering from depression, animal models of depression and neurological disorders such as Huntington's disease in which depression is a major symptom.

Description: Fat Free Antibody: Fat Free, also known as ANG2, was identified though a yeast two-hybrid screen using VP53. It is thought to be a structural and functional homolog of the yeast protein VPS51, a protein that together with VPS52, VPS53, and VPS54 form the Golgi-associated retrograde protein (GARP) complex that mediates the tethering and fusion of endosome-derived transport carriers to the trans-Golgi network (TGN). Similar to depletion of any of the VPS52, VPS53, or VPS54, depletion of Fat Free using RNAi impairs protein retrieval to the TGN indicating that Fat Free is missing component of the GARP complex in most eukaryotes.

Description: Fusion between membranes is mediated by specific SNARE (soluble N-ethylmeleimide-sensitive factor attachment protein receptor) complexes. Two human SNARE proteins, VTI1A and VTI1B, are homologous to the yeast Q-SNARE Vtilp which is part of several SNARE complexes in different transport steps . VTI1A is involved in transport between the endosome and the trans-Golgi network. The SNARE complex of STX10, STX16, VTI1A, and VAMP3 is required for MPR (mannose 6-phosphate receptors) transport from endosomes to the Golgi after delivering lysosomal enzymes to the endocytic pathway . Transport from early/recycling endosomes to the TGN is mediated by the SNARE complex of STX6, STX16, VTI1A and VAMP4 .

Description: This gene encodes a member of the sorting nexin family. Members of this family contain a phox domain, which is a phosphoinositide binding domain, and are involved in intracellular trafficking. Overexpression of this gene results in a decrease in the processing of insulin and hepatocyte growth factor receptors to their mature subunits. This decrease is caused by the mislocalization of furin, the endoprotease responsible for cleavage of insulin and hepatocyte growth factor receptors. This protein is involved in endosomal trafficking from the plasma membrane to recycling endosomes or the trans-Golgi network. Alternative splicing results in multiple transcript variants. Read-through transcription also exists between this gene and the upstream ADP-ribosylation factor-like 2 gene.

Description: This antibody is predicted to react with bovine, mouse, porcine and rat based on sequence homology. This gene belongs to a group of vacuolar protein sorting genes. The encoded protein is a component of a large multimeric complex, termed the retromer complex, involved in retrograde transport of proteins from endosomes to the trans-Golgi network. The close structural similarity between the yeast and human proteins that make up this complex suggests a similarity in function. Expression studies in yeast and mammalian cells indicate that this protein interacts directly with VPS35, which serves as the core of the retromer complex. Alternative splicing results in multiple transcript variants encoding different isoforms.

Description: TIP47 Antibody: Tail-interacting protein (TIP47) is a cytosolic protein essential for the transport of mannose-6-phosphate receptors (MPRs) from endosomes to the trans-Golgi compartments in cells. TIP47 is recruited from the cytoplasm to the late endosomal surface by the Ras-associated protein Rab9 GTPase, enabling it to bind more efficiently to MPR cytoplasmic domains. Recently, it was shown that TIP47 regulates the expression of Rab9, as expression of siRNA to TIP47 in transfected cells dramatically decreased the half-life of the Rab9 protein in addition to stabilizing the subcellular localization of Rab9. At least two isoforms of TIP47 are known to exist.

Description: Nuclear receptor that binds DNA as a monomer to ROR response elements (RORE) containing a single core motif half-site 5'-AGGTCA-3' preceded by a short A-T-rich sequence. Considered to have intrinsic transcriptional activity, have some natural ligands such as all-trans retinoic acid (ATRA) and other retinoids which act as inverse agonists repressing the transcriptional activity. Required for normal postnatal development of rod and cone photoreceptor cells. Modulates rod photoreceptors differentiation at least by inducing the transcription factor NRL-mediated pathway. In cone photoreceptor cells, regulates transcription of OPN1SW. Involved in the regulation of the period length and stability of the circadian rhythm. May control cytoarchitectural patterning of neocortical neurons during development. May act in a dose-dependent manner to regulate barrel formation upon innervation of layer IV neurons by thalamocortical axons. May play a role in the suppression of osteoblastic differentiation through the inhibition of RUNX2 transcriptional activity (By similarity)

Description: IGSF11 is also known as BTIGSF (brain and testis-specific immunoglobulin superfamily protein) or VSIG3 (V-set and immunoglobulin domain-containing protein 3) and is a 431 amino acid protein that is expressed as three isoforms. IGSF11 is highly expressed in testis and ovary and is also expressed in brain, kidney and skeletal muscle, localized to the cellular membrane as a single-pass membrane protein. IGSF11 is an immunoglobulin with V-type and C2-type domains that function in molecular recognition. When IGSF11 is in the trans position, it plays an important role in cell-cell adhesion via both homophilic and heterophilic interactions with other molecules. These cell–cell interactions are also thought to be important for neuronal cell interactions, such as neuron–neuron or neuron–glia interactions, which are important for the development and function of the central nervous system. In addition, IGSF11 might also be involved interactions between Sertoli cells and spermatocytes, which are important associations during spermatogenesis. The IGSF11 gene is commonly upregulated in gastric cancer and IGSF11 is highly expressed in many types of human tumors, indicating that it may be useful as a target for immunotherapy.
