Description: Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the g phosphate of ATP, onto an acceptor amino acid in a substrate protein. By this basic mechanism, protein kinases mediate most of the signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement and cell movement, apoptosis, and differentiation. With more than 500 gene products, the protein kinase family is one of the largest families of proteins in eukaryotes. The family has been classified in 8 major groups based on sequence comparison of their tyrosine (PTK) or serine/threonine (STK) kinase catalytic domains. The AGC kinase group consists of 63 kinases including the cyclic nucleotide-regulated protein kinase (PKA & PKG) family, the diacylglycerol-activated/phospholipid-dependent protein kinase C (PKC) family, the related to PKA and PKC (RAC/Akt) protein kinase family, the kinases that phosphorylate G protein-coupled receptors family (ARK), and the kinases that phosphorylate ribosomal protein S6 family (RSK). Description: Also known as dioxopiperazines, piperazine-2,5-diones or DKPs. Diketopiperazines may occur as by-products during peptide synthesis or during the degradation of peptides. These cyclic dipeptides have been detected as taste-modulating compounds in food, they often show biological activity. DKPs are valuable chiral synthons, employed e.g. in Schöllkopf's versatile bislactim ether approach. They also have found use as catalysts for enantioselective synthesis, e.g. in the asymmetric Strecker reaction. See also the TRH metabolite cyclo(-His-Pro), G-1745, and cyclo(-Asp-Phe), G-1695, the major degradation product of aspartame.

Description: PDE9A catalyzes the hydrolysis of cAMP and cGMP to their corresponding monophosphates. The protein plays a role in signal transduction by regulating the intracellular concentration of these cyclic nucleotides. Multiple transcript variants encoding several different isoforms have been found for this gene. PDE9A is widely expressed in various tissues, including spleen, small intestine and brian.

Description: SERBP1 (Plasminogen activator inhibitor 1 RNA-binding protein) may play a role in the regulation of mRNA stability. SERBP1 binds to the 3’-most 134 nt of the SERPINE1/PAI1 mRNA, a region which confers cyclic nucleotide regulation of message decay. SERBP1 interacts with CHD3 and is localized in cytoplasmic and nuclear. It is expressed at high level in the heart, skeletal muscle and kidney, and at low levels in placenta, liver and brain.

Description: MRP4 transports cyclic nucleotides and anti-retroviral compounds and may be an organic anion pump relevant to cellular detoxification. It is widely expressed, with particularly high levels in prostate, but is barely detectable in liver. It belongs to the ATP binding cassette (ABC) transporter family. MRP4 confers resistance to topotecan and acts as a double agent in protecting the brain from chemotherapy.

Description: Cyclic GMP (cGMP), which has been shown to be present at levels typically 10-100 fold lower than cAMP, is formed by the action of the enzyme guanylate cyclase on GTP. Some hormones, such as Acetylcholine, Insulin, and Oxytocin, as well as certain other chemicals like Serotonin and Histamine cause an increase in cGMP levels. Stimulators of guanylate cyclase such as the vasodilators nitroprusside, nitroglycerin, sodium nitrate, and nitric oxide (NO) also stimulate cGMP levels.

Description: Prepro-atrial natriuretic factor (26-55) (human) (NT-pro-ANP (1-30)) is one of four peptide hormones derived from the atrial natriuretic prohormone. It is also known as long acting natriuretic peptide. The peptide has potent vasodilatory properties equal to atrial natriuretic factor. Vasodilation is associated with a 4-5 fold increase in cyclic GMP levels due to activation of particulate guanylate cyclase activity. Like prepro-atrial natriuretic factor (56-92) and kaliuretic peptide it is present in higher concentration in individuals with congestive heart failure. It causes the maximal increase in prostaglandin E2 (PGE2) levels of the three peptides. The known increase in PGE2 in chronic heart failure may be in part secondary to these peptides.
