Description: This antibody is predicted to react with mouse and rat based on sequence homology. RB1 likely acts as a regulator of other genes. It forms a complex with adenovirus E1A and with SV40 large T antigen, acts as a tumor suppressor, and may bind and modulate functionally certain cellular proteins with which T and E1A compete for pocket binding. RB1 is a potent inhibitor of E2F-mediated trans-activation, and also recruits and targets histone methyltransferase SUV39H1 leading to epigenetic transcriptional repression. This protein inhibits the intrinsic kinase activity of TAF1. Defects in RB1 are the cause of childhood cancer retinoblastoma (RB), a congenital malignant tumor that arises from the nuclear layers of the retina. Defects in RB1 are also a cause of bladder cancer and osteogenic sarcoma.

Description: TCC52 Antibody: Cancer-testis (CT) antigens are a type of antigen whose expression is highly restricted, primarily in normal testis, but aberrantly in different tumor types. Their immunogenicity in cancer patients makes them prime targets for immunotherapy of human tumors. One such CT antigen is TCC52, a novel centrosomal protein. TCC52 is a WD repeat-containing protein that interacts with the COP9 signalosome, a macromolecular complex that interacts with cullin-RING E3 ligases and regulates their activity by hydrolyzing cullin-Nedd8 conjugates. The normal biological function of this protein is not known at this time.

Description: Nanos1 Antibody: Nanos1 is one of three known mammalian homologs to the Drosophila gene nanos. Nanos1 is an RNA-binding protein containing a zinc-finger motif and is expressed in the developing nervous system and continues in the adult brain. Interestingly, unlike mice deficient in either nanos2 or nanos3, mice lacking the nanos1 gene develop normally with no sign of abnormalities. Recently it has been found that expression of nanos1 mRNA is down-regulated by E-cadherin in a human breast cancer cell line and the amino-terminal domain on Nanos1 interacts with the E-cadherin-binding protein p120ctn. Furthermore, overexpression of Nanos1 in human colorectal DLD1 cancer cells functionally abolished cell-cell adhesion, allowing the cancer cells to develop strong migratory and invasive properties. These results suggest that targeting Nanos1 might prove an effective strategy in the treatment of E-cadherin-negative tumors.

Description: Wild-type p53 induced phosphatase 1 (WIP1) / protein phosphatase magnesium-dependent 1 delta (PPM1D), a protein identified in the p53 DNA response pathway, is a member of the PP2C family of serine/threonine protein phosphatases which selectively inactivates p38 MAPK and dephosphorylates the ATM/ATR targets, Chk1 and p53. Studies have shown that it is overexpressed in human cancers and is involved in the regulation of multiple DNA damage signaling pathways. PPM1D deletion results in a reduction of T and B cell function and compromised cell division, rendering cells resistant to becoming cancerous and slowing tumor development.

Description: CCDC12, also known as FLJ39430, FLJ40801 or MGC23918, is a 166 amino acid protein encoded by a gene mapping to human chromosome 3. Chromosome 3 is made up of about 214 million bases encoding over 1,100 genes. Notably, there is a chemokine receptor gene cluster and a variety of human cancer related loci on chromosome 3. Particular regions of the chromosome 3 short arm are deleted in many types of cancer cells. Key tumor suppressing genes on chromosome 3 encode apoptosis mediator RASSF1, cell migration regulator HYAL1 and angiogenesis suppressor SEMA3B. Marfan Syndrome, porphyria, von Hippel-Lindau syndrome, osteogenesis imperfecta and Charcot-Marie-Tooth Disease are a few of the numerous genetic diseases associated with chromosome 3.

Description: This antibody recognizes a 66 kDa protein, which is identified as Tripartite motif-containing protein 29 (TRIM29). It interacts with the intermediate filament protein vimentin, a substrate for the PKC family of protein kinases, and with hPKCI-1, an inhibitor of the PKCs. TRIM29 protein contains both zinc finger and leucine zipper motifs, suggesting that the it may form homodimers and possibly associate with DNA. High expression of TRIM29 has been reported in gastric cancer and pancreatic cancer, and correlates with enhanced tumor growth and lymph node metastasis. TRIM29 is also able to distinguish lung squamous cell carcinoma from lung adenocarcinoma with ~90% positive accuracy, when used in a panel with TTF-1, p63, CK5/6, and Napsin-A antibodies.

Description: The protein encoded by this gene is part of a complex of proteins that constitute adherens junctions (AJs). AJs are necessary for the creation and maintenance of epithelial cell layers by regulating cell growth and adhesion between cells. The encoded protein also anchors the actin cytoskeleton and may be responsible for transmitting the contact inhibition signal that causes cells to stop dividing once the epithelial sheet is complete. Finally, this protein binds to the product of the APC gene, which is mutated in adenomatous polyposis of the colon. Mutations in this gene are a cause of colorectal cancer (CRC), pilomatrixoma (PTR), medulloblastoma (MDB), and ovarian cancer. Three transcript variants encoding the same protein have been found for this gene.[provided by RefSeq, Oct 2009].

Description: MINA Antibody: MINA is nuclear localized, myc-inducible protein that is thought to play a role in mammalian cell proliferation. Treatment of cancer cells lines such as the colon cancer cell line SW680 with siRNA against MINA inhibits cell growth, demonstrating that MINA may be a potential therapeutic target. MINA regulates several genes related to cell adhesion and metabolism that have also been shown to be regulated by c-Myc, but also regulates other genes whose expression are not modulated by c-Myc such as EGFR, IL-6 and HGF. MINA has also been found to act as a repressor to IL-4 expression in T cells, indicating that it may also play a role in T cell differentiation and genetic variation in T helper type 2 bias.

Description: ZBTB7A Antibody: The ZBTB family of proteins is comprised of diverse zinc finger proteins that also contain a BTB (BR-C, ttk and bab) domain. ZBTB7A, also known as LRF, was initially identified as a potential target of the LAZ-3/BCL-6 oncogene. Later reports demonstrated that it could bind to the wild-type IST (inducer of short transcripts) elements of HIV-1 long terminal repeats (LTRs) and stimulate Tat activity on the HIV-1 LTR. More recently it ZBTB7A has been recognized as a proto-oncogene whose overexpression contributes to malignancy in breast cancer and functions as a co-repressor of the androgen receptor in prostate cancer cells.

Description: Cytochrome c oxidase (COX) is the terminal component of the mitochondrial respiratory chain, catalyzes the electron transfer from reduced cytochrome c to oxygen. The mitochondrially encoded subunits function in electron transfer, and the nuclear encoded subunits may function in the regulation and assembly of the complex. COX7A2L (cytochrome c oxidase subunit 7A-related protein), also known as COX7AR or COX7RP, is an inner mitochondrial membrane protein. This gene is expressed in all tissues, and upregulated in a breast cancer cell line after estrogen treatment. Recently it was found that DNA mythylation of COX7A2L had a significant association with therapy response in patients with breast cancer. In Alzheimer’s disease the expression of COX7A2L was reduced.

Description: SCUBE3 Antibody: SCUBE3 is a member of a family of secreted glycoproteins that contain N-terminal EGF-like repeats and C-terminal cysteine-rich motifs and CUB domain and is highly expressed in primary osteoblasts and bones, and to a lesser extent in heart. Other studies have shown that overexpression of SCUBE3 in mice induced cardiac hypertrophy, suggesting that it may also play a role in the regulation of cardiac growth. SCUBE3 has been shown to be an endogenous TGF-beta receptor ligand and is thought to promote lung cancer cell mobility and invasiveness. In lung cancer cells, the secreted SCUBE3 protein was cleaved by MMP2 and MMP9, allowing the activation of the TGF-beta receptor, the increase of Smad2/3 transcriptional activity and the upregulation of expression of proteins such as TGF-beta1, VEGF, Snail, and Slug.

Description: Members of the dickkopf-related protein family (DKK-1, -2, -3, and -4) are secreted proteins with two cysteine-rich domains separated by a linker region. And DKK3 has been proposed as tumour suppressor gene and a marker for tumour blood vessels. DKK3 is the only DKK family member abundantly expressed in normal lung, but silenced by promoter hypermethylation in a large fraction of lung cancer cell lines and lung tumors. Downregulation of DKK3 was correlated with tumor progression and expression of nuclear beta-catenin in lung tumors. Ectopic expression of DKK3 in lung cancer cells with DKK3 hypermethylation induced apoptosis and inhibited TCF-4 activity as well as nuclear accumulation of beta-catenin and expression of TCF-4 targets c-Myc and cyclin D1. DKK3 modulates FGF and Activin/Nodal signaling to regulate mesoderm induction during early Xenopus development, was reported.
