Description: CD274, also known as B7-H1 or programmed death ligand 1 (PD-L1), is a 40 kD type I transmembrane protein and a member of the B7 family within the immunoglobulin receptor superfamily. Programmed death-1 ligand-1 (PD-L1, CD274, B7-H1) has been identified as the ligand for the immunoinhibitory receptor programmed death-1(PD1/PDCD1) and has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. By binding to PD1 on activated T-cells and B-cells, PD-L1 may inhibit ongoing T-cell responses by inducing apoptosis and arresting cell-cycle progression. Accordingly, it leads to growth of immunogenic tumor growth by increasing apoptosis of antigen specific T cells and may contribute to immune evasion by cancers. PD-L1 thus is regarded as promising therapeutic target for human autoimmune disease and malignant cancers.

Description: Glycosylation of proteins affects cell-cell interaction, interactions with the matrix, and the functions of intracellular molecules. ST6GALNAC1 transfers a sialic acid, N-acetylneuraminic acid (NeuAc), in an alpha-2,6 linkage to O-linked GalNAc residues. The cancer-associated sialyl-Tn (sTn) antigen is formed by ST6GALNAC1-catalyzed sialylation of GalNAc residues on mucins.Glycosylation of proteins affects cell-cell interaction, interactions with the matrix, and the functions of intracellular molecules. Description: Epidermal Growth Factor (EGF) exerts its effects by binding to the EGF Receptor, a protein kinase that initiates the intracellular signaling (1). EGF is widely distributed in tissues like the kidney, cerebrum, prostate, and salivary glands. EGF acts as a potent factor in promoting cell division, and the phosphorylated receptor recruit adapter proteins like GRB2, which then activate complex downstream signaling cascades. EGF stimulates the proliferation differentiation, and survival of epithelial and epidermal cells. EGF possesses three intramolecular disulfide bonds and forms a strong attachment to the epidermal growth factor receptor (EGFR). Activation of EGFR initiates diverse cellular pathways in response to toxic environmental stimuli, or to EGF binding to the receptor, the EGFR forms homo- or heterodimers with other family members. (2, 3). EGF activates at least four major downstream signaling pathways, including the RAS-RAF-MEK-ERK, PI3 kinase-AKT, PLC gamma-PKC, and STAT modules. Additionally, research suggests that EGF may play a significant role in activating the NF-kappa-B signaling cascade (4). Mutations in the EGF gene are responsible for hypomagnesemia type 4, and its dysregulation has been linked to the development and progression of certain cancers (5).

Description: The protein encoded by the TRIM68 gene contains a RING finger domain, a motif present in a variety of functionally distinct proteins and known to be involved in protein-protein and protein-DNA interactions. TRIM68 is expressed in many cancer cell lines. Its expression in normal tissues, however, was found to be restricted to prostate. This gene was also found to be differentially expressed in androgen-dependent versus androgen-independent prostate cancer cells.

Description: The XPA (xeroderma pigmentosumgroup A) protein specifically recognizes the UV-orchemically damaged DNA lesions, and triggers thenucleotide excision repair process. XPA binds to thereplication protein A (RPA) or the excision repaircross complementing 1 protein (ERCC 1). In the absence of nucleotide excision repair persisting(unrepaired) DNA lesions (adducts) may lead to the accumulation of gene mutations and ultimately to cancer. Xeroderma pigmentosum patients have a >2000 fold increased risk to develop skin cancer atsun-exposed areas.

Description: FAM120A may participate in mRNA transport in the cytoplasm. FAM120A is the critical component of the oxidative stress-induced survival signaling. It activates src family kinases and acts as a scaffolding protein enabling src family kinases to phosphorylate and activate PI3-kinase. FAM120A binds RNA and promotes the secretion of IGF-II. It may play a pivotal role in the progression of scirrhous-type gastric cancer by supporting cancer cell survival in environments with various oxidative stresses.

Description: Hexokinases phosphorylate glucose to produce glucose-6-phosphate, thus committing glucose to the glycolytic pathway. HK2 (hexokinase 2) is the predominant form found in skeletal muscle. It localizes to the outer membrane of mitochondria. Expression of this protein is insulin-responsive, and studies in rat suggest that it is involved in the increased rate of glycolysis seen in rapidly growing cancer cells. Description: CCDC36 , also known as CT74 (cancer/testis antigen 74), is a 584 amino acid protein that contains coiled-coil domains and is expressed as three alternatively spliced isoforms. The gene encoding CCDC36 maps to human chromosome 3, which houses over 1,100 genes, including a chemokine receptor (CKR) gene cluster and a variety of human cancer-related gene loci. Key tumor suppressing genes on chromosome 3 include those that encode the apoptosis mediator RASSF1, the cell migration regulator HYAL1 and the angiogenesis suppressor SEMA3B. Marfan Syndrome, porphyria, von Hippel-Lindau syndrome, osteogenesis imperfecta and Charcot-Marie-Tooth Disease are a few of the numerous genetic diseases associated with chromosome 3.

Description: ATP11A Antibody: ATP11A is a widely expressed integral membrane ATPase. ATP11A is probably phosphorylated in its intermediate state and likely drives the transport of ions such as calcium or other molecules across membranes. While the exact molecule ATP11A transports is unknown, increased expression of ATP11A mRNA has been observed in murine leukemia cells made resistant to anti-cancer drugs such as farnesyltransferase inhibitors (FTIs). Furthermore, overexpression of ATP11A provided protection against the FTI SCH66336 while knockdown of ATP11A via siRNA made cells more sensitive to this drug. Other reports suggest that elevated levels of ATP11A mRNA may also be a predictive marker of metastasis in colorectal cancer patients.
