Description: The MTS510 monoclonal antibody specifically reacts with the Toll-Like Receptor 4 (TLR-4) and MD-2 (LY96) molecular complex. The complex of TLR-4, MD-2, and CD14 regulates the innate immune system recognition of bacterial lipopolysaccharides (LPS) and is expressed on the surface of thioglycollate-elicited macrophages. The epitope that binds the MTS510 antibody is lost after LPS stimulation and the antibody can be used to co-immunoprecipitates MD-2 and TLR4.

Description: The genus Salmonella is a member of the family Enterobacteriaceae. The genus is composed of Gram-negative bacilli that are facultative and flagellated (motile). Salmonellae possess 3 major antigens; the "H" or flagellar antigen (phase 1 & 2), the "O" or somatic antigen (part of the LPS moiety) and the "Vi" or capsular antigen (referred to as "K" in other Enterobacteriaceae). Salmonellae also possess the LPS endotoxin characteristic of Gram-negative bacteria. This LPS is composed of an "O" polysaccharide ("O" antigen) an "R" core and the endotoxic inner "Lipid A". Endotoxins evoke fever and can activate complement, kinin and clotting factors. Until recently the most common cause of food poisoning by Salmonella species was due to S. Typhimurium. As its name suggests, it causes a typhoid-like disease in mice. In humans S. Typhimurium does not cause as severe disease as S. Typhi, and is not normally fatal. The disease is characterized by diarrhea, abdominal cramps, vomiting and nausea, and generally lasts up to 7 days.

Description: The Sa2-8 monoclonal antibody specifically reacts with mouse CD14, a cell surface anchored glycoprotein that is primarily expressed on macrophages and is secreted in certain conditions. CD14 associates with LPS and LBP to form a receptor complex, which signals specifically in response to bacterial lipopolysaccharide (LPS) binding. The Sa2-8 antibody exhibits a weak antagonistic activate in relation to TNF alpha production and NF-kappa B activation.

Description: Interleukin-1 (IL-1) and lipopolysaccharide (LPS) induces cellular response through IL-1 receptor (IL-1R) and Toll like receptors (TLR). IL-1 receptor-associated kinase (IRAK and IRAK2) mediates the activation of NF-betaB by IL-1/Toll receptors, which is a pivotal transcription factor mediating inflammatory and immune response. A novel member in the IRAK/Pelle family was recently identified and designated IRAK-M. IRAKs associate with IL-1/Toll receptors after IL-1 or LPS stimulation and the dominant negative mutants of IRAKs inhibit IL-1 or LPS induced NF-betaB activation. Members in IRAK/Pelle family play a central role in IL-1R/TLR mediated inflammatory responses to cytokine IL-1 and LPS.

Description: MD-2 Antibody: MD-2 is a member of the Toll/interleukin-1 receptor (TIR) family, a group of proteins that include the Toll-like receptors (TLRs). TLRs are signaling molecules that recognize different pathogen-associated molecular patterns (PAMPs) and serve as an important link between the innate and adaptive immune responses. TLR4, the major signaling receptor for lipopolysaccharide (LPS), requires the binding of MD-2 to its extracellular region for maximal response to LPS. The specificity of this response is determined by the species of MD-2; e.g., human MD-2 can cause mouse TLR4 to react to LPS analogs that are normally antagonistic to human but not mouse TLR4.

Description: Antimicrobial peptides are produced by plants and most organisms throughout the animal kingdom including humans. Antimicrobial peptides protect against a broad range of infectious agents, as bacteria, fungi, and viruses. The amphibian skin is an especially rich source of antimicrobial peptides. See also the product families: Hepcidins LL-37 and Fragments Tuftsin and Analogs (subfamily).

Description: AG-126 prevents lipopolysaccharide-induced lethal toxicity (septic shock) in mice. Administered at 1.5 mg/mouse, it reduced LPS-induced lethal toxicity from 95% to 10%. AG-126 greatly reduces LPS-induced tyrosine phosphorylation of p42MAPK in murine peritoneal macrophages at 50 µM. EGFR, Her-2/neu receptor or PDGF-R are not inhibited at 100 µM. Inhibits tyrosine kinase-dependent B cell receptor signaling.

Description: Triggering Receptor Expressed on Myeloid Cells 1 (TREM-1) is a transmembrane protein with a single Ig-like domain. TREM-1 associates with the adapter protein, DAP12, to deliver an activating signal. TREM-1 is expressed on blood neutrophils and monocytes, and the expression is up-regulated by bacterial LPS. TREM-1 is expressed at high levels on neutrophils of patients with microbial sepsis and in mice with a TREM-1/Fc fusion protein protected mice against LPS-induced shock. Human TREM-1 shares 42% sequence homology with mouse TREM-1.

Description: CD14 is a cell surface anchored glycoprotein that is expressed predominantly by monocytes and tissue macrophages. CD14 associates with MD-2 (LY-96) and TLR4 to form a receptor complex, which signals specifically in response to bacterial lipopolysaccharide (LPS) binding. The CD14/MD-2/TLR4 receptor complex signals via MyD88, TIRAP, and TRAF6, and ultimately activates NF-kappa-β. CD14 also exists in a soluble form, designated as sCD14, which is capable of specifically binding LPS in the extracellular space. Recombinant sCD14 is a 331 amino acid glycoprotein comprising the extracellular portion of the CD14 receptor.

Description: Molecular chaperone that promotes the maturation, structural maintenance and proper regulation of specific target proteins involved for instance in cell cycle control and signal transduction. Undergoes a functional cycle that is linked to its ATPase activity. This cycle probably induces conformational changes in the client proteins, thereby causing their activation. Interacts dynamically with various co-chaperones that modulate its substrate recognition, ATPase cycle and chaperone function. Binds bacterial lipopolysaccharide (LPS) et mediates LPS-induced inflammatory response, including TNF secretion by monocytes.

Description: Activation of cells by LPS is mediated by the Toll-like receptor 4 (TLR4), a member of the highly conserved protein family of TLRs, which are specialised in the recognition of microbial components. In mice, defects in TLR4 result in LPS unresponsiveness. For optimal interaction with LPS, TLR4 requires association with myeloid differentiation protein 2 (MD-2). According to current consensus activation of TLR4 is preceded by the transfer of LPS to membrane-bound (m) or soluble (s) CD14 by LPS-binding protein (LBP). This mechanism is believed to be generally true for LPS signaling. Re-form LPS and lipid A, but not S-form LPS, are capable of inducing TNF-alpha responses also in the absence of CD14. LPS, synthesized by most wild-type (WT) Gram-negative bacteria (S-form LPS), consists of three regions, the O-polysaccharide chain, which is made up of repeating oligosaccharide units, the core oligosaccharide and the lipid A, which harbors the endotoxic activity of the entire molecule. R-form LPS synthesized by the so-called rough (R) mutants of Gram-negative bacteria lacks the O-specific chain. Furthermore, the core-oligosaccharide may be present in different degrees of completion, depending on the class (Ra to Re) to which the mutant belongs. Monophosphoryl Lipid A (MPLA) represents a detoxified derivative of Lipid A and constitutes an important adjuvant in prophylactic and therapeutic vaccines.
