Description: NAIP Antibody: Neuronal apoptosis inhibitor protein (NAIP) was the first human inhibitor of apoptosis protein (IAP) identified and was discovered by its association with the neurodegenerative disorder spinal muscular atrophy. Members of the IAP family contain one to three copies of an approximately 70 amino acid motif termed baculovirus IAP repeat (BIR); these BIRs promote protein-protein interactions with various caspases such as caspase-3, -7, and -9 as well as members of the TRAF family of signal molecules. Unlike other IAPs however, NAIP requires ATP to bind caspase-9 and is not inhibited by the IAP-inhibiting molecule Smac/DIABLO, suggesting that NAIP is unique among the IAPs in its regulation of its activity. Finally, although only one human NAIP protein has been identified, other shorter NAIP mRNA transcripts have been reported.

Description: p16-INK4a is a nuclear protein that regulates the cell cycle by inhibiting cyclin dependent kinase-4 (CDK4) and CDK6. p16-INK4a inhibits CDK activity by binding to the CDK molecules in a manner that interferes with their ability to interact with cyclin D. This activity has the effect of suppressing tumor formation and growth, and of inducing replicative senescence in various normal cells, including stem cells. The expression of p16-INK4a steadily increases with age and tends to accumulate in stem cell compartments. The deletion, rearrangement, or mutation of the p16-INK4a gene is frequently found in melanomas as well as in certain other types of cancer. P16-INK4a and other transcription factors have been introduced into cells by DNA transfection, viral infection, or microinjection. Protein transduction using TAT fusion proteins represents an alternative methodology for introducing transcription factors and other nuclear proteins into primary as well as transformed cells. Recombinant human p16-INK4a-TAT expressed in E. coli is a 18 kDa protein containing 167 amino-acid residues, including the 156 residues of full-length p16-INK4a and a 13-residue C-terminal TAT peptide (GGYGRKKRRQRRR).

Description: Macrophage migration inhibitory factor (MIF) is a pro-inflammatory lymphokine that functions during cell-mediated immmunity. MIF promotes fibroblast migration by inducing interleukin 1 (IL-1), interleukin 8 (IL-8), and matrix metalloproteinase (MMP) expression. In interferon-gamma-activated macrophages, MIF stimulates nitric oxide (NO) production and tumor necrosis factor-alpha (TNF-α) secretion.

Description: TAB1 Antibody: TAB1 was identified as a regulator of the MAP kinase kinase kinase TAK1/MAP3K7, which is known to mediate various intracellular signaling pathways, such as those induced by TGF-beta and members of the Toll-IL-1R (TIR) superfamily, thus acting as an intermediate in both proliferative and innate and adaptive immune responses. This protein, together with either TAB2 or TAB3, activates TAK1 kinase in response to upstream signals. It has been shown that the C-terminal portion of TAB1 is sufficient for binding and activation of TAK1, while a portion of the N-terminus acts as a dominant-negative inhibitor of TGF-beta, demonstrating how this protein can function as a mediator between TGF-beta receptors and TAK1.

Description: AIM Antibody: Apoptosis inhibitor of macrophages (AIM) is a member of the scavenger receptor cysteine-rich domain superfamily (SRCR-SF) initially identified as an inducible cell surface ligand of CD5. It was shown that AIM functions in the thymus as the inducer of resistance to apoptosis within CD4+/CD8+ thymocytes and as the supporter of the viability of these cells before thymic selection. AIM was also shown to support macrophage survival and enhance their phagocytic function. More recent experiments using recombinant AIM significantly inhibited apoptosis of NKT and T cells obtained from C. parvum-stimulated livers in vitro, suggesting that AIM functions to induce resistance to apoptosis in these cells and supports host defense against inflammation during infection.

Description: IL-15 Antibody: Interleukin 15 (IL-15) is a cytokine that regulates T and natural killer cell activation and proliferation. This cytokine and IL-2 share many biological activities as both have been found to bind common hematopoietin receptor subunits, and may compete for the same receptor, and thus negatively regulate each other's activity. The number of CD8+ memory cells is shown to be controlled by a balance between IL-15 and IL-2. This cytokine induces the activation of JAK kinases, as well as the phosphorylation and activation of transcription activators STAT3, STAT5, and STAT6. In mouse, studies suggest that IL-15 may increase the expression of apoptosis inhibitor Bcl-xL, possibly through the transcription activation activity of STAT6, and thus prevent apoptosis.

Description: UNG1 Antibody: The human uracil-DNA glycosylase (UNG) gene encodes both mitochondrial (UNG1) and nuclear (UNG2) forms through differentially regulated promotes and alternative splicing. While UNG2 is the major enzyme in the base excision repair pathway that removes uracil residues from nuclear DNA that arise through either misincorporation during replication or cytosine deamination, inhibition of UNG1 by uracil glycosylase inhibitor did not lead to increased levels of spontaneous or induced mitochondrial DNA mutations. However, decreased levels of UNG activity and increased oxidative damage to mitochondrial DNA were seen in older mice, suggesting that mitochondrial DNA repair mechanisms may be involved in various neurodegenerative disorders in an age-dependent manner. This UNG1 antibody will not cross-react with UNG2.
