Description: TRIM5 alpha Antibody: TRIM5 is a member of a broad family of otherwise unrelated proteins defined by the presence of a tripartite motif containing a RING domain, a B-box type 1, and a B-box type 2, followed by a coiled-coil region. TRIM5 has six alternately spliced isoforms, the longest of which is the alpha variant which also contains a carboxy-terminal B30.2 (SPRY) domain. Expression of TRIM5 alpha variants from humans, rhesus monkeys, and African green monkeys enabled resistance to infection by various retroviruses including HIV-1, albeit at differing efficiencies. All TRIM5 alpha variants could inhibit at least two different retroviruses, but not from those viruses isolated from the same species, suggesting that TRIM5 alpha acts as a natural barrier to cross-species retrovirus transmission.

Description: Lano Antibody: Lano is a member of the LAP (leucine-rich repeats and PDZ) family of proteins that also includes Densin-180, Erbin, and hScribble. The LAP proteins generally contain multiple leucine-rich repeat (LRR) domains which serve to target them to the basolateral membrane of epithelial cells. Lano is unique in that it alone does not possess one or more PDZ (PSD95/DLG/ZO-1) domains as do the other members of the LAP family. However, it can bind to the PDZ domain of Erbin in addition to those of membrane-associated and guanylate kinase (MAGUK) proteins which regulate adhesion and plasticity at cell junctions. It has been suggested that it is through these interaction that these LAP proteins participate in the maintenance of proper embryonic development and integrity of epithelial tissues.

Description: ZFX Antibody: The X-linked zinc finger protein ZFX is a member of the Kruppel C2H2-type zinc-finger protein family. The full-length protein contains an acidic transcriptional activation domain (AD), a nuclear localization sequence (NLS) and a DNA binding domain (DBD) consisting of 13 C2H2-type zinc fingers. Studies in mouse embryonic and adult hematopoietic stem cells showed that this gene was required as a transcriptional regulator for self-renewal of both stem cell types, but it was dispensable for growth and differentiation of their progeny.

Description: The angiopoietin-related protein 3 (ANGPTL3) is a member of a family of secreted proteins that function in angiogenesis. ANGPTL3 is processed into an N-terminal coiled-coil domain-containing chain and a C-terminal fibrinogen chain. The N-terminal chain is important for lipid metablism, while the C-terminal chain may be involved in angiogenesis. Recent experiments have shown that a deficiency of ANGPTL3 is associated with increased insulin sensitivity, lipoprotein lipase activity, and decreased serum free fatty acids, suggesting that ANGPTL3 may also play a role in modulating glucose metabolism.

Description: THEM4 Antibody: THEM4, also known as CTMP, binds specifically to the carboxy-terminal regulatory domain of PKB/Akt at the plasma membrane and acts as a negative regulator, reversing the phenotype of v-Akt-transformed cells. Hypermethylation of the THEM4 promoter and transcriptional downregulation of the gene has been reported in multiple glioblastomas, suggesting that epigenetic regulation of THEM4 may play a role in the progression of this cancer. Bioinformatic analysis, confirmed by in vitro testing, indicates that THEM4 is a broad-range, high activity acyl-CoA thioesterase. Recent reports have also indicated that TMEM4 is a mitochondrial protein whose overexpression is associated with an increase in mitochondrial membrane depolarization and caspase-3 and PARP cleavage, suggesting that THEM4 is involved in the apoptotic program.

Description: Syntaphilin Antibody: Syntaphilin was initially identified in a yeast two-hybrid screen with the carboxy terminal region of Syntaxin-1 as bait. Syntaxin-1 is a key component of the synaptic vesicle docking machinery that forms the SNARE complex with synaptobrevin and SNAP-25. Syntaphilin competes with SNAP-25 for binding to syntaxin-1 and inhibits the formation of the SNARE complex, thereby potentially regulating synaptic vesicle exocytosis. Syntaphilin also binds dynamin-1 and inhibits dynamin-dependent endocytosis. Mice lacking syntaphilin show an increased level of mitochondrial motility and a reduced density of axonal mitochondria. This correlates with an enhanced short-term facilitation and significant impairments in motor ability, suggesting syntaphilin plays a major role in presynaptic function. Multiple isoforms are known to exist.

Description: StrepII-tag Antibody: Streptavidin is a tetrameric protein purified from Streptomyces avidinii. It has wide use in numerous molecular biological protocols dues to its strong affinity for biotin. The original Strep-tag is a nine amino acid peptide with high specificity and affinity towards streptavidin. The addition of this tag to the C-terminus of recombinant proteins allowed the simple purification of protein by use of affinity columns. It was re-engineered to allow it to also placed at the N-terminus of recombinant proteins.

Description: PRR5 Antibody: Proline-rich protein 5 (PRR5), also known as Protor-1, is a 388 amino acid protein in Protor family, is thought to act as a tumor suppressor in breast and colorectal tumorigenesis. PRR5 is widely expressed and possesses two RICTOR interaction sites and a C-terminal Proline rich region. It promotes Rapamycin complex 2 (mTORC2) activity. There are four isoforms of PRR5 that are produced as a result of alternative splicing events and these isoforms play an important role in the modulation of platelet-derived growth factor signaling and in the regulation of PDGFR-beta expression.

Description: FBXL16 Antibody: FBXL16 (F-box and leucine-rich repeat protein 16), a member of the F-box protein family, is a probable substrate-recognition component of the SCF (SKP1-CUL1-F-box protein)-type E3 ubiquitin ligase complex. It is characterized by an approximately 40-amino acid F-box motif. F-box proteins interact with SKP1 through the F-box, and they interact with ubiquitination targets through other protein interaction domains. FBXL16 is a novel E2F1-regulated gene in crucial cell cycle regulation pathways.

Description: WDR18 Antibody: WD repeat domain 18 (WDR18) is a member of the WD repeat protein family, which is involved in a variety of cellular processes, including cell cycle progression, signal transduction, apoptosis, and gene regulation. WD repeats are minimally conserved regions of approximately 40 amino acids that commonly form a tertiary propeller structure. WDR18 is a 432 amino acid protein that contains six WD repeats and is encoded by a gene located on human chromosome 19. WDR18 may play a role during development. The complex PELP1-TEX10-WDR18 is required for the maturation and nucleolar release of the large ribosomal subunit.

Description: IRAK-4 Antibody: Interleukin-1 (IL-1) and lipopolysaccharide (LPS) induces cellular responses through IL-1 receptor (IL-1R) and Toll-like receptors (TLR). IL-1R-associated kinases (IRAK, IRAK2, and IRAK-M) regulate the activation of NF-kappa B and MAP kinase (MAPK) by IL-1R/TLR. A novel member in the IRAK/Pelle family was recently identified and designated IRAK-4. Overexpression of IRAK-4 activates NF-kappa B and MAPK pathways. IRAK-4 interacts with and phosphorylates IRAK-1. IRAK-4-deficient animals are completely resistant to the challenge with LPS. Animals and humans lacking IRAK-4 are impaired in their responses to viral and bacterial challenges. Members in IRAK/Pelle family play a central role in IL-1R/TLR mediated inflammatory responses and in innate immunity.

Description: Trail Antibody: Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand in the TNF family through their death domain containing receptors, TNFR1 and Fas. A novel member in the TNF family was recently identified and designated TRAIL (for TNF-related apoptosis-inducing ligand) and Apo-2L (for Apo-2 ligand)1, TRAIL is a type II membrane protein and expressed in a variety of human tissues. Two novel death domain containing receptors DR4 and DR5 have been identified as the receptor for TRAIL3-6. Like TNF and Fas ligand, TRAIL induces apoptosis and NF-kappa B activation in many tissues and cells.

Description: DRAK2 Antibody: Apoptosis is mediated by death domain containing adapter molecules and a caspase family of proteases. Certain serine/threonine protein kinases, such as ASK-1 and RIP, are mediators of apoptosis. Two novel serine/threonine kinases that induce apoptosis were recently identified and designated DRAK1 and DRAK2 (for DAP kinase-related apoptosis-inducing protein kinases). DRAKs contain an N-terminal kinase domain and a C-terminal regulation domain. Overexpression of DRAK2 induces apoptosis. DRAKs have high sequence homology to DAP and ZIP kinases, and they represent a novel family of serine/threonine kinases, which mediates apoptosis through their catalytic activities. DRAK2 is located in nucleus and the messenger RNA was ubiquitously expressed in human tissues.

Description: ADAM10 Antibody: Proinflammatory cytokine tumor necrosis factor-alpha (TNF-alpha ) contributes to a variety of inflammatory responses and programmed cell death. Notch receptor and its ligand participate in cell fate decisions during vertebrate development and are associated with several human disorders, including a T-cell lymphoma. TNF-alpha , notch and its ligand delta are all membrane-bond molecules, which are cleaved by proteases to release mature proteins or functional receptor. ADAM10, a metalloprotease-disintegrin in the family of mammalian ADAM (for a disintegrin and metalloprotease), was recently identified to cleave TNF-alpha , notch and its ligand delta. The genes encoding human, mouse, and bovine ADAM10 were recently cloned and designated ADAM 10, kuzbanian (KUZ), and MADM, respectively. ADAM10 mRNA is expressed in a variety of human and bovine tissues.

Description: ICAD Antibody: Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. These death signals finally cause the degradation of chromosomal DNA by activated DNase. A human DNA fragmentation factor (DFF) was identified recently which was cleaved by caspase-3 during apoptosis. Mouse homologue of human DFF was identified as a DNase inhibitor designated ICAD, for inhibitor of caspase-activated DNase. Upon cleavage of DFF/ICAD, a caspase activated deoxyribonuclease (CAD) is released and activated and eventually causes the degradation of DNA in the nuclei. Therefore, the cleavage of CAD inhibitor molecule DFF/ICAD, which causes DNase activation and DNA degradation, is the hallmark of apoptotic cell death.

Description: BAFF Receptor Antibody: Members in the TNF superfamily regulate immune responses and induce apoptosis. A novel member in the TNF family was recently identified by several groups and designated BAFF, BLyS, TALL-1, THANK, and zTNF4. BAFF/BLyS was characterized as a B cell activator since it induced B cell proliferation and immunoglobulin secretion. Two receptors, TACI and BCMA, for BAFF were originally identified. A third receptor was identified recently and designated BAFF-R and BR3 for BLyS receptor 3. Unlike BCMA and TACI, which bind to BAFF and April, BAFF-R/BR3 is specific for BAFF and plays a predominant role in BAFF induced B cell development and survival. BAFF and its receptors are involved in B cell associated autoimmune diseases, and activate NF-kappa B and c-jun N-terminal kinase.
