Description: Sialidase-1 belongs to the N-acetyl-a neuraminidase family. Sialidase-1 is expressed in many tissues; it is highly expressed in the pancreas, and weakly expressed in the brain. Sialidase-1 is a lysosomal enzyme, which cleaves terminal sialic acid residues from substrates such as glycoproteins and glycolipids. Deficiencies in the human enzyme Sialidase-1 leads to sialidosis, a rare lysosomal storage disease. Sialidase-1 has been shown to interact with Cathepsin A (protective protein), beta -galactosidase and N-acetylgalactosamine-6-sulfate sulfatase in a multienzyme complex.

Description: Fibroblast growth factor 2(FGF2) is a secreted protein and belongs to the heparin-binding growth factors family. FGF2 is produced by epithelial, tumor and other cell types. It involved in developmental processes and regulates differentiation, proliferation, and migration, FGF2 is a critical factor for growing embryonic stem cells in culture without inducing differentiation. FGF2 has a high affinity for heparan sulfate and binding is a step in the FGF basic activation of FGFR tyrosine kinase.

Description: Growth factor active in angiogenesis, vasculogenesis and endothelial cell growth. Induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and induces permeabilization of blood vessels. Binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin. NRP1/Neuropilin-1 binds isoforms VEGF-165 and VEGF-145. Isoform VEGF165B binds to KDR but does not activate downstream signaling pathways, does not activate angiogenesis and inhibits tumor growth.

Description: NAP2 is a platelet-derived growth factor that belongs to the CXC chemokine family. This growth factor is a potent chemoattractant and activator of neutrophils. It has been shown to stimulate various cellular processes including DNA synthesis, mitosis, glycolysis, intracellular cAMP accumulation, prostaglandin E2 secretion, and sythesis of hyaluronic acid and sulfated glycosaminoglycan. It also stimulates the formation and secretion of plasminogen activator by synovial cells.

Description: Probably plays a role in cartilage scaffolding. May act by antagonizing TGF-beta1 (TGFB1) and IGF1 functions. Has the ability to suppress IGF1-induced proliferation and sulfated proteoglycan synthesis, and inhibits ligand-induced IGF1R autophosphorylation. May inhibit TGFB1-mediated induction of cartilage matrix genes via its interaction with TGFB1. Overexpression may lead to impair chondrocyte growth and matrix repair and indirectly promote inorganic pyrophosphate (PPi) supersaturation in aging and osteoarthritis cartilage.

Description: SPOCK3,also called Testican3,is a secreted protein and make up a family of extracellular heparan/chondroitin sulfate proteoglycans. It contains 1 Kazal-like domain and 1 thyroglobulin type-1 domain and mainly expressed in brain. SPOCK3 contain inhibitory regions in several domains targeted to different classes of protease, and in some cases may act as protease inhibitors. In addition to their presence in testis, testicans are enriched in brain and have been shown to regulate neuronal attachment and outgrowth.

Description: Sulfotransferase enzymes catalyze the sulfate conjugation of many hormones, neurotransmitters, drugs, and xenobiotic compounds. These cytosolic enzymes are different in their tissue distributions and substrate specificities. The gene structure (number and length of exons) is similar among family members. This gene encodes one of two phenol sulfotransferases with thermostable enzyme activity. Multiple alternatively spliced variants that encode two isoforms have been identified for this gene.

Description: This gene encodes a protein with 13 putative coiled-coil domains, a region with homology to SMC chromosome segregation ATPases, six KID motifs, three tropomyosin homology domains and an ATP/GTP binding site motif A. The protein is localized to the centrosome and cilia and has sites for N-glycosylation, tyrosine sulfation, phosphorylation, N-myristoylation, and amidation. Mutations in this gene have been associated with Joubert syndrome and nephronophthisis and the presence of antibodies against this protein is associated with several forms of cancer. Description: Glypican-1 is a cell membrane protein and belongs to the glypican family. The protein may act as a catalyst in increasing the rate of conversion of prion protein PRPN(C) to PRNP(Sc) via associating (via the heparan sulfate side chains) with both forms of PRPN, targeting them to lipid rafts and facilitating their interaction. It is required for proper skeletal muscle differentiation by sequestering FGF2 in lipid rafts preventing its binding to receptors (FGFRs) and inhibiting the FGF-mediated signaling.

Description: CDO1 (cysteine dioxygenase, type I) belongs to the cysteine dioxygenase family and is involved in organosulfur biosynthesis. CDO1 functions as a dioxygenase that uses iron and zinc as cofactors to catalyze the conversion of L-cysteine and oxygen to 3-sulfinoalanine. CDO1 exists as a monomer and is expressed at high levels in liver and placenta and at lower levels in brain, pancreas and heart. CDO1 is involved in pyruvate-, sulfate- and taurine-related metabolic pathways and is a crucial regulator of cysteine concentrations within the cell.
