Supplier: BIOGEMS INTERNATIONAL INC.
Description: The 61D3 monoclonal antibody specifically reacts with CD14, a cell surface anchored glycoprotein that is primarily expressed on monocytes, inerfollicular macrophages, and a subset of dendritic cells. CD14 associates with MD-2 (LY-96) and TLR4 to form a receptor complex, which signals specifically in response to bacterial lipopolysaccharide (LPS) binding. The CD14/MD-2/TLR4 receptor complex signals via MyD88, TIRAP, and TRAF6, and ultimately activates NF-kappaB.
BG Violet 450 conjugate is an alternative to the Pacific Blue, eFluor 450, or BD Horizon V450 dyes. It is excited by the violet (405 nm) laser, with a peak emission of 450nm.

Description: Collectin-11 is a secreted protein that belongs to the COLEC10/COLEC11 family. Collectin-11 contains one C-type lectin domain and one collagen-like domain. Collectins play important roles in the innate immune system by binding to carbohydrate antigens on microorganisms, facilitating their recognition and removal. Collectin-11 binds to various sugars including fucose and mannose, but does not bind to glucose, N-acetylglucosamine and N-acetylgalactosamine. It has a higher affinity for fucose compared to mannose. Collectin-11 binds lipopolysaccharides (LPS). It also involved in fundamental development serving as a guidance cue for neural crest cell migration. Defects in Collectin-11 are the cause of 3MC syndrome type 2 (3MC2).

Description: IRGM belongs to a family of interferon-gamma inducible GTPases, belonging to the GTPase superfamily, and are selectively induced by IFN-gamma or bacterial lipopolysaccharide (LPS) stimulation. IRGM is primarily expressed in all cells derived from B-cell lineages, and is highly expressed in macrophages following IFN-gamma stimulation. IRGM has been shown to be required for host defences against a broad range of intracellular pathogens. Specifically, IRGM deficient mice show defects with intracellular microbial killing, phagosome maturation and autophagy. More recently IRGM has been shown to inhibit baseline hematopoietic proliferation and is required for a normal hematopoietic stem cell response to chemical and infectious stimuli.

Description: LRRFIP2 Antibody: The leucine-rich repeat FLI-I-interacting protein 2 (LRRFIP2), like the related protein LRRFIP1, was identified in a yeast two-hybrid system through binding to the LRR domain of human FLI. It can activate the Wnt signaling pathway in cultured cells and is thought to be a component of the Wnt signaling pathway that modulates Wnt signaling through interactions with Disheveled to increase the cellular levels and transcription activity of beta-catenin. LRRFIP2 has recently been characterized as a positive regulator of the TLR4 signaling pathway for activating NF-kappa B during the early host response to LPS stimulation through binding to the TLR adaptor protein MyD88, and that this interaction with MyD88 is governed by phosphorylation of specific residues in LRRFIP2.

Description: MD-1 Antibody: Toll-like receptors (TLRs) are evolutionarily conserved pattern-recognition molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. The signaling of these TLRs is kept under tight control by the expression of endogenous inhibiting proteins such as RP105, a recently identified homolog of TLR4. This protein, in association with MD-1, interacts with and inhibits the TLR4/MD-2 signaling pathway. While MD-2 can directly bind to LPS, one of the activating molecules of TLR4, the function of MD-1 is less well-known. It has been suggested however, that the RP105/MD-1 complex influences antibody production mediated by both TLR4/MD-2 and TLR2 receptor complexes.

Description: PD-1 Antibody: Cell-mediated immune responses are initiated by T lymphocytes that are themselves stimulated by cognate peptides bound to MHC molecules on antig en-presenting cells (APC). T-cell activation is generally self-limited as activated T cells express receptors such as PD-1 (also known as PDCD-1) that mediate inhibitory signals from the APC. PD-1 can bind two different but related ligands, PDL-1 and PDL-2. Upon binding to either of these ligands, signals generated by PD-1 inhibit the activation of the immune response in the absence of "danger signals" such as LPS or other molecules associated with bacteria or other pathogens. Evidence for this is seen in PD1-null mice who exhibit hyperactivated immune systems and autoimmune diseases.

Description: Nuclear factor kappa B (NFkB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NFkB mediates the expression of a great variety of genes in response to extracellular stimuli including IL1, TNF alpha, and bacterial product LPS. NFkB is associated with IkB proteins in the cell cytoplasm, which inhibit NFkB activity. IKK is a serine protein kinase, and the IKK complex contains alpha and beta subunits (IKK alpha and IKK beta). IKK alpha and IKK beta interact with each other and both are essential for NFkB activation. IKK alpha specifically phosphorylates IkBa. IKKa is expressed in variety of human tissues.

Description: IKK epsilon Antibody: Nuclear factor kappa B (NF-kappa B) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kappa B mediates the expression of a great variety of genes in response to extracellular stimuli. NF-kappa B is associated with Ikappa B proteins in the cell cytoplasm, which inhibit NF-kappa B activity. Ikappa B is phosphorylated by Ikappa B kinase (IKK) complex that contains IKK alpha , IKK beta, and IKKgamma;. A novel molecule in the IKK complex was recently identified and designated IKKepsilon /IKK-i. IKK epsilon is required for the activation of NF-kappa B by PMA and T cell receptors but not by TNF alpha and IL-1. IKKepsilon /IKK-i message is expressed in a variety of tissues and is inducible by TNFalpha , IL-1, and LPS.

Description: TIRAP Antibody: Toll-like receptors (TLRs) are signaling molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. These proteins act through adaptor molecules such as TIRAP and MyD88 to activate various kinases and transcription factors. In TIRAP-deficient mice, TLR signaling in response to TLR2 ligands (using either TLR1 and TLR6 as co-receptors) is totally abolished, suggesting that MyD88 and TIRAP work together and are both required for TLR2 signaling. Furthermore, these mice are also resistant to the toxic effects of LPS and show defects in NF-kappaB and MAP kinase activation, suggesting that TIRAP is also involed in TLR4 signaling.

Description: CARMA1 Antibody: CARMA proteins belong to the membrane-associated guanylate kinase-like (MAGUK) family of proteins that can function as molecular scaffolds that assist assembly of signal transduction molecules. CARMA1, CARMA2, and CARMA3 share high degrees of sequence and functional homology, but their tissue-specific distribution suggests that they serve distinct biological functions in different cell types. Both CARMA1 and CARMA3 associate with NEMO, the regulatory subunit of the I kappa K complex, thereby regulating activation of the NF-kappa B transcription factor. Also, gene inactivation studies showed a complete block in T and B cell immunity as well as an impaired response to LPS, indicating that CARMA1 is a critical regulator in both the adaptive and innate immune systems.

Description: Caspase-5 Antibody: Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens. Members of this subfamily include caspase-1, -4, -5, and -12. Activation of these caspases results in the cleavage and activation of proinflammatory cytokines such as IL-1 beta and IL-18. Caspase-5 can interact with caspase-1; both are constituents of the NALP1 inflammasome, a complex that can trigger the cleavage of pro-IL-1 beta. Expression of caspase-5 can be regulated by lipopolysaccharide (LPS) and IFN-gamma.
