Description: Functions in the early steps of protein synthesis by forming a ternary complex with GTP and initiator tRNA. This complex binds to a 4S ribosomal subunit, followed by mRNA binding to form a 43S preinitiation complex. Junction of the 6S ribosomal subunit to form the 8S initiation complex is preceded by hydrolysis of the GTP bound to eIF-2 and release of an eIF-2-GDP binary complex. In order for eIF-2 to recycle and catalyze another round of initiation, the GDP bound to eIF-2 must exchange with GTP by way of a reaction catalyzed by eIF-2B.

Description: RBMY1A1 is a protein containing an RNA-binding motif in the N-terminus and four SRGY (serine, arginine, glycine, tyrosine) boxes in the C-terminus. The gene that encodes RBMY1A1 is Y-linked. RBMY1A1 may be involved in spermatogenesis. It is required for sperm development, possibly by participating in pre-mRNA splicing in the testis.

Description: The activation of RaP1 by cAMP is independent of PKA and is mediated by recently discovered family of guanine nucleotide exchange factors (GEFs) called cAMP-GEFs or Epacs. The Epac signaling therefore represents a novel mechanism for cAMP signaling with in the cAMP cascade. There are 2 members of the Epac family, Epac1 and Epac 2. Both proteins are multidomain proteins containing an autoinhibitory cAMP-binding domain that inhibits the catalytic region and a DEP domain (dishevelled, Egl-10 and pleckstrin homology domain) targeting the membrane anchors. EPAC2 has an additional cAMP-binding site in its N-terminus that binds cAMP with low affinity. EPAC1 mRNA is broadly expressed, with particularly high levels occurring in the thyroid, ovary, kidney and certain brain regions, whereas expression of EPAC2 mRNA appears to be restricted to the brain and adrenal glands. Epac 1 and Epac 2 also interact with light chain 2 (LC2) or MAP1A that serves as a scaffolding structure to stabilize the signal transduction complex. The Epac 1-selective were generated against unique antigenic sequences form near N-terminus and between RasGEFN and Ras GEF domains. The to Epac 1are affinity purified over immobilized antigen based chromatography.

Description: Involved in the alternative regulation of pre-mRNA splicing; its RNA helicase activity is necessary for increasing tau exon 10 inclusion and occurs in a RBM4-dependent manner. Binds to the tau pre-mRNA in the stem-loop region downstream of exon 10. The rate of ATP hydrolysis is highly stimulated by single-stranded RNA. Involved in transcriptional regulation; the function is independent of the RNA helicase activity. Transcriptional coactivator for estrogen receptor ESR1 and androgen receptor AR. Increases ESR1 AF-1 domain-mediated transactivation and ESR1 AF-1 and AF-2 domains transcriptional synergistic activity. Synergizes with DDX17 and SRA1 RNA to activate MYOD1 transcriptional activity and involved in skeletal muscle differentiation. Transcriptional coactivator for p53/TP53 and involved in p53/TP53 transcriptional response to DNA damage and p53/TP53-dependent apoptosis. Transcriptional coactivator for RUNX2 and involved in regulation of osteoblast differentiation. Acts as transcriptional repressor in a promoter-specicic manner; the function probbaly involves association with histone deacetylases, such as HDAC1. As component of a large PER complex is involved in the inhibition of 3' transcriptional termination of circadian target genes such as PER1 and NR1D1 and the control of the circadian rhythms.

Description: Biological Significance: Glyceraldehyde 3-Phosphate Dehydrogenase (GAPDH) is a metabolic enzyme responsible for catalyzing one step in the glycolytic pathway, the reversible oxidative phosphorylation of glyceraldehyde 3-phosphate. Because GAPDH is a protein expressed in large amounts and which is required at all times for important "house keeping" functions, levels of GAPDH mRNA are often measured and used as standards in studies of mRNA expression. Increasingly, scientists are making use of specific antibodies to GAPDH in comparable studies of levels of protein expression. This antibody can be used as a loading control for western blotting experiments, allowing comparison between the level of this protein and others in a cell or tissue. Apart from a role in glycolysis, GAPDH may have other roles such as in the activation of transcription. GAPDH is reported to bind to a variety of other proteins, including the amyloid precursor protein, mutations in which cause some forms of Alzheimer's disease, and the polyglutamine tracts of Huntingtin, the protein product aberrant forms of which are causative of Huntington's disease. Associations with actin and tubulin have also been reported. The protein may also have a role in the regulation of apoptosis, and interestingly migrates from the cytoplasm into the nucleus when cells become apoptotic.

Description: The 4 major small nuclear ribonucleoprotein particles (snRNPs), U1, U2, U4/U6, and U5, are essential components of the eukaryotic pre-mRNA splicing machinery. These snRNPs share 8 proteins which form the snRNP structural core: B/B-prime (SNRPB), D1 (SNRPD1), D2 (SNRPD2), D3 (SNRPD3), E (SNRPE), F (SNRPF), and G (SNRPG). These proteins have a role in the biogenesis of the snRNPs.

Description: Eukaryotic initiation factor 4A (EIF4A) has an essential role in the binding of mRNA to the 43S preinitiation complex when protein synthesis begins, as EIF4A unwinds RNA secondary structures in the 5'-UTR of mRNAs which is necessary to allow efficient binding of the small ribosomal subunit, and subsequent scanning for the initiator codon. EIF4A2 is a ATP-dependent RNA helicase, which is a subunit of the eIF4F complex involved in cap recognition and is required for mRNA binding to ribosome.

Description: RIOK3(Serine/threonine-protein kinase RIO3) is initially characterized as a homolog of Aspergillus nidulans sudD and showed down-regulation at the invasive front of malignant melanomas. On Western blots, human RIOK3 migrated as a 60-kD protein. Northern blot analysis indicated that the approximately 4-kb RIOK3 mRNA is expressed widely in human tissues, with additional smaller transcripts present in testis.

Description: Sm-like proteins, such as LSM4, were identified in a variety of organisms based on sequence homology with the Sm protein family. Sm-like proteins contain the Sm sequence motif, which consists of 2 regions separated by a linker of variable length that folds as a loop. Sm-like proteins are thought to form a stable heteromer present in tri-snRNP particles, which are important for pre-mRNA splicing

Description: PRPF4, also named as U4/U6 small nuclear ribonucleoprotein Prp4, is a 522 amino acid protein, which contains seven WD repeats. PRPF4 localizes in the Nucleus speckle. PRPF4 participates in pre-mRNA splicing and is a part of the U4/U5/U6 tri-snRNP complex, one of the building blocks of the spliceosome.PRPF4-mediated phosphorylation of KLF13 plays a role in the regulation of CCL5 expression in T lymphocytes
