CDNF/ARMETL1 is a evolutionary conserved protein which can protect and restore the function of dopaminergic neurons in the rat model of Parkinson's disease, suggesting that CDNF might be beneficial for the treatment of Parkinson's disease. CDNF is widely expressed in neurons in several brain regions including cerebral cortex, hippocampus, substantia nigra, striatum and cerebellum. Human CDNF is glycosylated and secreted from transiently transfected cells. Description: MANF Antibody: MANF, also known as ARMET, was initially identified as a protein containing an arginine-rich region that was highly mutated in a variety of tumors. More recently it was identified as a mesencephalic astrocyte-derived neurotrophic factor with selectivity for dopaminergic neurons, similar to glial cell line-derived neurotrophic factor (GDNF) and CDNF. In rat brain slices, MANF enhanced nigral gamma-aminobutyric acid release. Like GDNF and CDNF, MANF has selective neuroprotective activity for dopaminergic neurons suggesting that it may be indicated for the treatment of Parkinson's disease. Expression of MANF has also been shown to be induced during ER stress, suggesting that it may play a role in protein quality control during ER stress.

Description: Mesencephalic astrocyte-derived neurotrophic factor(MANF) is a secreted protein which belongs to the ARMET family. MANF selectively promotes the survival of dopaminergic neurons of the ventral mid-brain. It modulates GABAergic transmission to the dopaminergic neurons of the substantia nigra. MANF enhances spontaneous, as well as evoked, GABAergic inhibitory postsynaptic currents in dopaminergic neurons. MANF inhibits cell proliferation and endoplasmic reticulum (ER) stress-induced cell death. The N-terminal region of ARMET may be responsible for neurotrophic activity while the C-terminal region may play a role in the ER stress response. MANF reduces endoplasmic reticulum (ER) stress and has neurotrophic effects on dopaminergic neurons. Intracortical delivery of recombinant MANF protein protects tissue from ischemic brain injury. MANF has been described as a survival factor for dopaminergic neurons. MANF and a homologous protein, the conserved dopamine neurotrophic factor (CDNF), form a novel evolutionary conserved family of neurotrophic factors. MANF expression was widespread in the nervous system and non-neuronal tissues.

Description: This antibody is predicted to react with canine, mouse and rat based on sequence homology. The protein encoded by this gene is localized in the endoplasmic reticulum and golgi, and is also secreted. Reducing expression of this gene increases susceptibility to ER stress-induced death and promotes cell proliferation. The protein was initially thought to be longer at the N-terminus and to contain an arginine-rich region but transcribed evidence indicates a smaller open reading frame that does not encode the arginine tract. The presence of polymorphisms in the arginine-rich region, including a specific mutation that changes the previously numbered codon 50 from ATG to AGG, have been reported in a variety of solid tumors; however, these polymorphisms were later shown to exist in normal tissues and are thus not tumor-related.
