Description: The ion channels activated by glutamate are typically divided into two classes. Those that are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors (NMDAR) while those activated by beta-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid (AMPA) are known as AMPA receptors (AMPAR). The AMPAR are comprised of four distinct subunits GluR1-4 and they play key roles in virtually all excitatory neurotransmission in the brain. The GluR1 subunit is widely expressed throughout the nervous system. GluR1 is also potentiated by phosphorylation at Ser831. In addition, phosphorylation of this site has been linked to synaptic plasticity as well and learning and memory.

Description: AMPK is a heterotrimeric complex comprising a catalytic a subunit and regulatory b and g subunits. It protects cells from stresses that cause ATP depletion by switching off ATP-consuming biosynthetic pathways. AMPK is activated by high AMP and low ATP through a mechanism involving allosteric regulation, promotion of phosphorylation by an upstream protein kinase known as AMPK kinase and inhibition of dephosphorylation. Activated AMPK can phosphorylate and regulate in vivo hydroxy-methylglutaryl-CoA reductase and acetyl-CoA carboxylase, which are key regulatory enzymes of sterol synthesis and fatty acid synthesis, respectively. The human AMPKa1 and AMPKa2 genes encode 548 amino acid and 552 amino acid proteins, respectively. Human AMPKb1 encodes a 271 amino acid protein and human AMPKb2 encodes a 272 amino acid protein. The human AMPKg1 gene encodes a 331 amino acid protein. Human AMPKg2 and AMPKg3, which are 569 and 492 amino acid proteins, respectively, contain unique N-terminal domains and may participate directly in the binding of AMP within the AMPK complex.

Description: The CNS is enriched in polyunsaturated fatty acids and is therefore particularly vulnerable to lipid peroxidation. 4-hydroxy-2-noneal (HNE) is one of the major lipid peroxidation products with cytotoxic and mutagenic activity. It reacts with protein residues such as histidine to generate stable Michael adducts. HNE has been shown to inhibit neurite outgrowth and disrupt microtubules and tubulin modification, suggesting it may play a role in alzheimer′s disease.

Description: Blocks cell cycle at the G1 and G2/M phase [1] by inhibiting G1 phase cyclin dependent kinases [2] downregulating cyclin D1 [3]. Modulates all three aspects of the glutathione-mediated biotransformation system, i.e. GSH levels, glutathione S-transferase activity and transport of GSH conjugates [4]. Antitumor activity. Induces γ-glutamylcysteine synthetase and apoptosis in L-1210 cells.
