Description: RTP801 Antibody: RTP801 was initially identified as a gene induced by DNA damage, and later found to also be regulated by other cellular stresses such as hypoxia and glucocorticoid treatment. Recently, RTP801 has been shown to act as a mediator of tuberous sclerosis complex (TSC)-dependent regulation of the mammalian Target of Rapamycin (mTOR), an evolutionarily conserved serine/threonine kinase that regulates cell growth and cell cycle. In response to energy stress, RTP801 inhibits mTOR function, resulting in dephosphorylation of downstream targets such as ribosomal protein S6 kinase 1 and 4EBP1 and decreasing cell growth. Disregulation of RTP801 may thus contribute to human tumorigenesis.

Description: CDNF Antibody: The conserved dopamine neurotrophic factor (CDNF) is a neurotrophic factor for dopaminergic neurons and highly homologous to the mesencephalic-astrocyte-derived neuro-trophic factor. Somewhat surprisingly, CDCF is expressed at higher levels in tissues such as heart, skeletal muscle and testes than in brain. Similar to the glial cell line-derived neurotrophic factor (GDNF), CDNF can prevent the 6-hydroxylamine (6-OHDA)-induced degeneration of dopaminergic neurons in a rat model of Parkinson's disease. Furthermore, CDNF was able to restore the dopaminergic function and prevent the degeneration of dopaminergic neurons in the substantia nigra, suggesting that CDNF might be suitable for the treatment of Parkinson's disease. At least two isoforms of CDNF are known to exist.

Description: Siglec11 Antibody: Siglecs are sialic acid-binding lectins of the immunoglobulin superfamily that are mainly expressed in cells of the hematopoietic system. Siglec11 is unlike other siglecs in that it binds specifically to alpha2-8-linked sialic acids and is not found in peripheral blood leukocytes but instead on macrophages of various tissues. Siglec11 is highly homologous to Siglec10 over their extracellular domain but not over their cytoplasmic domain, suggesting that Siglec11 arose through gene duplication followed by a recombination event involving another ancestral siglec gene. Following treatment of Siglec11-transfected cells with pervanadate, Siglec11 becomes tyrosine-phosphorylated and strongly associates with SHP-1 and SHP-2. At least five isoforms of Siglec11 are known to exist. Siglec11 antibody will not cross-react with Siglec10.

Description: MINA Antibody: MINA is nuclear localized, myc-inducible protein that is thought to play a role in mammalian cell proliferation. Treatment of cancer cells lines such as the colon cancer cell line SW680 with siRNA against MINA inhibits cell growth, demonstrating that MINA may be a potential therapeutic target. MINA regulates several genes related to cell adhesion and metabolism that have also been shown to be regulated by c-Myc, but also regulates other genes whose expression are not modulated by c-Myc such as EGFR, IL-6 and HGF. MINA has also been found to act as a repressor to IL-4 expression in T cells, indicating that it may also play a role in T cell differentiation and genetic variation in T helper type 2 bias.

Description: PAR4 is a member of a unique family of GPCRs, the proteinase-activated receptors (PARs), that are activated by proteolytic cleavage of the N-terminal domain of the receptor to reveal a tethered ligand. The PAR family consists of 4 receptors; PAR1 and PAR3 are activated by thrombin, and PAR2 and PAR4 are activated by several serine proteases (Macfarlane et al., 2001). PAR4 is expressed along with PAR1 on human platelets, and each plays an important role in thrombin-induced platelet aggregation (Holinstat et al., 2006). However, platelets from mice express PAR4 but not PAR1, and platelets from mice lacking PAR4 fail to aggregate in response to thrombin (Sambrano et al., 2001). Mice lacking PAR4 are protected from mesenteric arteriole thrombosis, indicating that PAR4 is a potential target for treatment of thrombosis in humans.

Description: CNOT2 (CCR4-NOT transcription complex subunit 2) is a ubiquitous protein encoded by the human gene CNOT2. CNOT2 belongs to the CNOT2/3/5 family and is part of the CCR4-NOT complex. The CCR4-NOT complex is an evolutionarily conserved, multi-component complex known to be involved in transcription as well as mRNA degradation. Various subunits (e.g. CNOT1, CNOT2) are involved in influencing nuclear hormone receptor activities. The CCR4-NOT complex is also involved in the regulation of Histone H3 lysine 4 methylation through a ubiquitin-dependent pathway that likely involves the proteasome. Increased expression of the CNOT2 subunit acts to strongly repress transcription by RNA polymerase II. This repressive effect is mediated by a conserved NOT-Box, which is located at the C-terminus of CNOT2 proteins. Repression by the NOT-Box is sensitive to treatment with the histone deacetylase (HDAC) inhibitor trichostatin A.

Description: This antibody is predicted to react with canine, mouse and rat based on sequence homology. Catechol-O-methyltransferase catalyzes the transfer of a methyl group from S-adenosylmethionine to catecholamines, including the neurotransmitters dopamine, epinephrine, and norepinephrine. This O-methylation results in one of the major degradative pathways of the catecholamine transmitters. In addition to its role in the metabolism of endogenous substances, COMT is important in the metabolism of catechol drugs used in the treatment of hypertension, asthma, and Parkinson disease. COMT is found in two forms in tissues, a soluble form and a membrane-bound form . The differences between S-COMT and MB-COMT reside within the N-termini. Several transcript variants are formed through the use of alternative translation initiation sites and promoters.

Description: This antibody recognizes an N-glycosylated glycoprotein of 120 kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3. CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This MAb is excellent for staining of formalin/paraffin tissues.

CF® dyes are Biotium's next-generation fluorescent dyes. CF®647 is a far-red fluorescent dye (Ex/Em 650/665 nm) with excellent brightness. It also is compatible with super-resolution imaging by STORM.

Description: ZCRB1 Antibody: ZCRB1 is a nuclear protein first identified in a differential display screen involving morphine-dependence-related genes. Its expression is increased following morphine treatment and has been observed to be elevated in HepG2 cells. ZCRB1 contains a CCHC-type zing finger RNA-binding motif and can interact with the DBA-binding domain of the stem cell regulator C/EBP transcription factors. ZCRB1 was identified as one of the protein components of U11/U12 snRNPs, which are components of U12-type spliceosome and function as a molecular bridge connecting both ends of the intron, suggesting ZCRB1 may play a key role in U12-type splicing.

Description: Indoleamine 2,3-dioxygenase (IDO) is a heme enzyme that initiates the oxidative degradation of the least abundant, essential amino acid, l-tryptophan, along the kynurenine pathway. This protein is normally expressed in the dendritic cells, macrophages, microglia, eosinophils, fibroblasts, endothelial cells, and most tumor cells. IDO activity is associated with immunosuppression and immune attenuation. Several studies showed that IDO can contribute to immune escape when expressed directly in tumor cells or when expressed in immunosuppressive antigen presenting cells such as tolerogenic dendritic cells or tumor associated macrophages. IDO also is a promising therapeutic target for the treatment of cancer, chronic viral infections, and other diseases characterized by pathological immune suppression.

Description: MARVELD1 Antibody: MARVELD1, (MARVEL domain-containing 1), is one of the MARVEL (MAL and related proteins for vesicle trafficking and membrane link) domain-containing proteins. It is a nuclear-localized protein that is widely expressed in normal human tissues. While it has been reported to be overexpressed in colon cancer, MARVELD1 protein levels are generally lower in several primary tumors including ovary, breast, kidney, bladder and liver. This lower level of MARVELD1 expression in tumors is thought to be due to a higher methylation status of the MARVELD1 promoter as treatment of MDA-MB-361 with 5-aza-2'-deoxycytidine restores MARVELD1 expression, suggesting that loss of MARVELD1 expression may contribute to tumorgenicity.

Description: p44/42 MAP Kinase(Thr202); ERK (extracellular signal regulated kinase), also known as MAPK (mitogen activated protein kinase) has two closely related isoforms of 44 kDa and 42 kDa, respectively. These kinases belong to a family of serine/threonine kinases that are activated upon treatment of cells with a large variety of stimuli including mitogens, hormones, growth factors, cytokines, and bioactive peptides. Cell stimulation induces the activation of a signaling cascade, the downstream effects of which have been linked to the regulation of cell growth and differentiation as well as the cytoskeleton. ERK1 and ERK2 are phosphorylated within the activation loop on both a Threonine and a Tyrosine residue (within a Thr-Glu-Tyr motif) by MEKs (MAPK/ERK kinases), thereby greatly elevating the activity of ERK1&2.

Description: Bcl-xL Antibody: Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. Bcl-xL is a member of the Bcl-2 family of proteins that are critical regulators of apoptosis. These can be divided into two classes: those that inhibit apoptosis and those that promote cell death. Bcl-xL is an anti-apoptotic mitochondrial protein related to Bcl-w and the major transcript of the bcl-x gene. Its high expression in tumors is correlated with advanced disease and poor prognosis. Bcl-xL expression level increases in response to several stimuli such as ionizing radiation and treatment with chemotherapeutic agents.

Description: p40 (p63 delta) is a marker recently determined to be highly specific for squamous basal cells in the immunohistochemistry (IHC) application. The current more routinely recommended marker, p63, appears to have less specificity compared to p40, especially on squamous cell tumors. The ability to differentiate between lung adenocarcinoma vs. squamous cell carcinoma is difficult and has bearing on the different therapeutic avenues for each subtype treatment. p63 antibody's ability to distinguish between the tumor types appears to be inferior when compared to p40. The ability to utilize an antibody probe for p40 as a squamous cell marker bolsters its use for future sub-classification of lung cancers, especially by immunohistochemical techniques.
