Description: Blimp-1 Antibody: Blimp-1 was initially identified as a zinc finger-containing protein that drives the maturation of B lymphocytes into immunoglobulin-secreting cells. Together with X-box-binding protein 1 (XBP1), Blimp-1 is induced upon terminal differentiation of plasma cells. The transcriptional repressor activity of Blimp-1 has also been found to regulate T cell homeostasis and function, possibly by suppressing the expression of the cytokines IL-2 and interferon-gamma during T cell development. More recent experiments have suggested that Blimp-1 also plays a major role in the formation of primordial germ cells (PGC) in developing mammalian embryos. In these experiments, Blimp-1-deficient mutant mouse embryos form a cluster of PGC-like cells which fail to show the expected migration, proliferation, and repression of homeobox genes that normally accompany specification of primordial germ cells. Blimp-1 exists as at least two different isoforms.

Description: DDX3 Antibody: DDX3 contains all of the motifs of the DEAD-box family of RNA helicases, including the Asp-Glu-Ala-Asp sequence that gives the protein family its name and distinguishes it from other RNA helicases. DDX3 is localized to the X chromosome and has a highly conserved functional homolog (DBY) on the Y chromosome. DDX3 is thought to be involved in RNA splicing, RNA transport, and translation initiation. It has also been shown to be involved in cell growth control and is deregulated in hepatitis virus-associated hepatocellular carcinoma. Recent experiments suppressing DDX3 expression blocked HIV-1 RNA export from the nucleus, suggesting that DDX3 functions as a shuttling protein that transports the HIV-1 protein Rev and its cofactor CRM1 from the nucleus to the cytoplasm.

Description: ZBRK1 Antibody: ZBRK1 is a member of the Kruppel-associated box-zinc finger protein (KRAB-ZFP) protein family of transcriptional repressors. ZBRK1 has been shown to not only be up-regulated in senescent fibroblasts but also interacts with BRCA1, a transcriptional regulator with tumor suppressor activity. ZBRK1 binds to a specific sequence within the GADD45 gene, supporting the assembly of a nuclear complex containing BRCA1 and repressing its transcription. Recent experiments demonstrate that ZBRK1 can homo-oligomerize in vitro and in vivo through a C-terminal transcriptional repression domain (CTRD), and this oligomerization facilitates the ZBRK1-directed transcriptional repression through ZBRK1 response elements. These results suggest that higher order ZBRK1 oligomers may assemble through both protein-DNA and CTRD-dependent protein-protein interactions.

Description: TRIM30 Antibody: TRIM30 belongs to a family of the tripartite motif (TRIM) proteins involved in the regulation of cell proliferation, differentiation, development, oncogenesis, apoptosis and antiviral responses. The TRIM protein family is an expanding family of RING ('really interesting new gene') proteins, also known as RBCC proteins as they contain an RBCC motif, which comprises a RING domain, one or two B-boxes and a predicted coiled-coil region. Studies have shown that some TRIM family members are critical to innate immunity; TRIM5, TRIM19 and TRIM25, for example, have been shown to restrict viral infection. A recent study shows that TRIM30 functions as a negative modulator of the TLR signaling pathway, by targeting TAB2 and TAB3, and contributes to the inhibition of TLR-mediated NF-kappa B activation. The importance of TRIM30 in the attenuation or termination of NF-kappa B activation suggests that targeting of TAB2 and TAB3 by TRIM30alpha may be a mechanism for modulating many types of immune responses.

Description: Gle1 Antibody: The proper expression of gene products in eukaryotic cells relies on efficient transport of mRNA molecules out of the nucleus. Gle1 is an essential mRNA export factor in both human and yeast cells. It associates with the nuclear pore complex (NPC) through hCG1 and NUP155 in mammalian cells and in conjunction with inositol hexakisphosphate (IP6), stimulates Dbp5, a member of the DEAD-box helicase family, triggering mRNP remodeling and facilitating RNA export from the nucleus. Recent evidence suggests that mutations in Gle1 causing defects in mRNA export can result in human disease. At least three isoforms of Gle1 are known to exist.

Description: Tripartite motif-containing protein 3 (TRIM3), also known as RING finger protein 22 (RNF22), RING finger protein 97 (RNF97) or brain-expressed RING finger protein (BERP), is a 744 amino acid member of the TRIM family, also known as the RING-B-box coiled-coil (RBCC) family. Members of the RBCC family have an N-terminal RING finger, followed by one or two zinc-binding domains (B-box domains), a leucine coiled-coil region and a variable C-terminal domain. Localized to cytoplasmic filaments, TRIM3 has been shown to interact with å-actinin-4 and myosin V, two proteins associated with the actin cytoskeleton. Specifically, å-actinin-4 interacts with the RBCC domain of TRIM3, and the C-terminal tail of Myosin V interacts with with the unique C-terminal ∫-propeller domain of TRIM3. These associations suggest that TRIM3 may play a role in cell motility and cargo transport. Three named isoforms of TRIM3 exist as a result of alternative splicing events.

Description: Stk39 Antibody: The serine/threonine kinase Stk39 belongs to the STE20 family, a group of kinases that are known to interact with inflammation-related kinases (such as p38, JNK, NKCC1, PKC-theta, WNK and MLCK), and with transcription factor AP-1. The STE 20 family is involved in diverse biological phenomena, including cell differentiation, cell transformation/ proliferation, cytoskeleton rearrangement, and the regulation of ion transporters. STK39 contains an N-terminal series of proline and alanine repeats (PAPA box), followed by a serine/threonine kinase catalytic domain and is abundantly expressed in the brain. STK39 is activated in response to hypotonic stress, leading to phosphorylation of several cation-chloride-coupled co-transporters. The catalytically active kinase specifically activates the p38 MAP kinase pathway, and its interaction with p38 decreases upon cellular stress, suggesting that this kinase may serve as an intermediate in the response to cellular stress. Recent studies show that STK39 tend to be a novel candidate gene for autism and hypertension.

Description: This antibody is predicted to react with mouse based on sequence homology. This gene encodes a member of the cupin superfamily. The encoded protein is an Fe-containing nuclear protein expressed in all tissues of the body and concentrated within dot-like subnuclear structures. Interactions with nuclear factor I/CCAAT box transcription factor as well as B cell lymphoma 3-encoded oncoprotein suggest the encoded protein may act as a transcriptional cofactor and be involved in the regulation of DNA transcription and replication. Alternatively spliced transcript variants have been described.

Description: SOX2 Antibody: SOX2 is a member of the SRY-related HMG-box (SOX) family of transcription factors involved in the regulation of embryonic development and in the determination of cell fate. SOX2 is required for stem-cell maintenance in the central nervous system, and also regulates gene expression in the stomach. Mutations in this gene have been associated with optic nerve hypoplasia and with syndromic microphthalmia, a severe form of structural eye malformation. The role of SOX2 in embryonic development suggested that it might be useful in the creation of stem cells that might be useful in cell replacement therapies in the treatment of degenerative diseases. Artificial stem cells, termed induced pluripotent stem (iPS) cells, can be created by expressing SOX2 and the transcription factors POU5F1, Klf4 and Lin28 along with c-Myc in mouse fibroblasts. Other experiments have shown that iPS cells could be generated using expression plasmids expressing POU5F1, SOX2, KlfF4 and c-Myc, eliminating the need for virus introduction.
