Description: Nop1p was originally identified as a nucleolar protein of bakers yeast, Saccharomyces cerevisiae. The Nop1p protein is 327 amino acids in size (34.5 kDa), is essential for yeast viability, and is localized in the nucleoli. The systematic name for S. cerevisiae Nop1 is YDL014W, and it is now known to be part of the small subunit processome complex, involved in the processing of pre-18S ribosomal RNA. Nop1p is the yeast homologue of a protein found in all eukaryotes and archea generally called fibrillarin. Fibrillarin/Nop1p is extraordinarily conserved, so that the yeast and human proteins are 67% identical, and the human protein can functionally replace the yeast protein. Patients with the autoimmune disease scleroderma often have strong circulating autoantibodies to a ~34 kDa protein which was subsequently found to be fibrillarin. Recent studies show that knock-out of the fibrillarin gene in mice results in embryonic lethality, although mice with only one functional fibrillarin/Nop1p gene were viable. This antibody is becoming widely used as a convenient marker for nucleoli in a wide variety of species (e.g. 4-6).

Description: The G protein–coupled receptor kinases (GRKs) are a versatile family of kinases that play a critical role in cancer metastasis through their regulation of G-protein coupled receptors (GPCRs) involved in growth factor mediated cell migration (1,2). Phosphorylation of receptors by GRKs appears to be strictly dependent on the receptor being in its agonist-activated state. GRK6 is one of 7 members of the GRK serine/threonine kinase subfamily, which has been shown to modulate the Wnt signaling pathway via phosphorylation of LRP6 (3,4), and the insulin-like growth factor signaling pathway. GRK6 may also play a role in immune system function.

Description: ANTS (8-aminonaphthalene-1,3,6-trisulfonic acid, disodium salt) is a highly negatively charged dye with an amino group that can be coupled to an aldehyde or ketone group to form an unstable Schiff base. The Schiff base is usually chemically reduced by sodium borohydride (NaBH4) or sodium cyanoborohydride (NaB(CN)H3) to form a stable linkage. This labeling technique has been widely used for the labeling and subsequent sequencing of oligosaccharides and glycoproteins. The negative charges of the dye facilitate the electrophoretic separation of the degradation products of carbohydrate polymers.

Description: SCUBE3 Antibody: SCUBE3 is a member of a family of secreted glycoproteins that contain N-terminal EGF-like repeats and C-terminal cysteine-rich motifs and CUB domain and is highly expressed in primary osteoblasts and bones, and to a lesser extent in heart (1,2). Other studies have shown that overexpression of SCUBE3 in mice induced cardiac hypertrophy, suggesting that it may also play a role in the regulation of cardiac growth. SCUBE3 has been shown to be an endogenous TGF-beta receptor ligand (3,4) and is thought to promote lung cancer cell mobility and invasiveness. In lung cancer cells, the secreted SCUBE3 protein was cleaved by MMP2 and MMP9, allowing the activation of the TGF-beta receptor, the increase of Smad2/3 transcriptional activity and the upregulation of expression of proteins such as TGF-beta1, VEGF, Snail, and Slug.

Description: β-NADP is a coenzyme necessary for the alcoholic fermentation of glucose and the oxidative dehydrogenation of other substances. It occurs widely in living tissue, especially in the liver. Nicotinic acid can be converted to nicotinamide in the body and, in this form, is found as a component of two oxidation-reduction coenzymes: nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP). The nicotinamide portion of the coenzyme transfers hydrogens by alternating between oxidized quaternary nitrogen and a reduced tertiary nitrogen. NADP is an essential coenzyme for glucose-6-phosphate dehydrogenase which catalyzes the oxidation of glucose-6-phosphate to 6-phosphogluconic acid. This reaction initiates metabolism of glucose by a pathway other than the citric acid cycle. This route is known as the hexose phosphate shunt or phosphogluconate pathway. Other enzymes which utilize NADP as a coenzyme are: Alcohol dehydrogenase:NADP dependent; Aromatic ADH:NADP dependent; Ferredoxin-NADP reductase; L-Fucose dehydrogenase; Gabase; Galactose-1-phosphate uridyl transferase; Glucose dehydrogenase; L-Glutamic dehydrogenase; Glycerol dehydrogenase:NADP specific; Isocitric dehydrogenase; Malic enzymes; 5,10-Methylenetetrahydrofolate dehydrogenase; 6-Phosphogluconate dehydrogenase and Succinic semialdehyde dehydrogenase.

Description: HAAO Antibody: HAAO (3-Hydroxyanthranilate 3, 4-dioxygenase) is a monomeric cytosolic protein of the family of intramolecular dioxygenases containing non-heme ferrous iron. It is widely distributed in peripheral organs, such as liver and kidney, and is present in low amounts in the central nervous system. This enzyme participates in tryptophan metabolism. It employs one cofactor, iron. HAAO catalyzes the synthesis of quinolinic acid (QUIN) from 3-hydroxyanthranilic acid. QUIN is an excitotoxin whose toxicity is mediated by its ability to activate glutamate N-methyl-D-aspartate receptors. Increased cerebral levels of QUIN may participate in the pathogenesis of neurological and inflammatory disorders. HAAO has been suggested to play a role in disorders associated with altered tissue levels of QUIN. Furthermore, recent study shows that HAAO are excellent candidate biomarkers for detecting ovarian cancer.

Description: MYOZ2 gene encodes for the calcineurin-binding protein myozenin-2 (or Calsarcin-1) belonging to myozenin family. Defects in MYOZ2 gene are the cause of familial hypertrophic cardiomyopathy type 16 (CMH16), a hereditary heart disorder characterized by ventricular hypertrophy. The disorder has inter- and intrafamilial variability ranging from benign to malignant forms with high risk of cardiac failure and sudden cardiac death . Myozenins may serve as intracellular binding proteins involved in linking Z line proteins such as alpha-actinin, and gamma-filamin. Important role in the modulation of calcineurin signaling and myofibrillogenesis of myozenin-2 have been elucidated . Proteintech’s Myozenin-2 antibody 11450-1-AP was raised against full-length protein of human origin and is able to detect band of approximate 34 kDa in Western blotting.

Description: ACRV1 is a testis-specific, differentiation antigen, acrosomal vesicle protein 1, that arises within the acrosomal vesicle during spermatogenesis, and is associated with the acrosomal membranes and matrix of mature sperm. The acrosomal vesicle protein 1 may be involved in sperm-zona binding or penetration, and it is a potential contraceptive vaccine immunogen for humans. This gene encodes a testis-specific, differentiation antigen, acrosomal vesicle protein 1, that arises within the acrosomal vesicle during spermatogenesis, and is associated with the acrosomal membranes and matrix of mature sperm. This gene consists of 4 exons and its alternative splicing generates multiple distinct transcripts, which encode protein isoforms ranging from 81 to 265 amino acids. The longest transcript is the most abundant, comprising 53-72% of the total acrosomal vesicle protein 1 messages; the second largest transcript comprises 15-32%; the third and the fourth largest transcripts account for 3.4-8.3% and 8.7-12.5%, respectively; and the remaining transcripts combined account for < 1% of the total acrosomal vesicle protein 1 message. It is suggested that phenomena of cryptic splicing and exon skipping occur within this gene. The acrosomal vesicle protein 1 may be involved in sperm-zona binding or penetration, and it is a potential contraceptive vaccine immunogen for humans.

Description: The TATA binding protein (TBP) is a transcription factor that binds specifically to a DNA sequence TATA box. This DNA sequence is found about 25-30 base pairs upstream of the transcription start site in some eukaryotic gene promoters. TBP, along with a variety of TBP-associated factors, make up the TFIID, a general transcription factor that in turn makes up part of the RNA polymerase II preinitiation complex. As one of the few proteins in the preinitation complex that binds DNA in a sequence-specific manner, it helps position RNA polymerase II over the transcription start site of the gene. However, it is estimated that only 10-20% of human promoters have TATA boxes. Therefore, TBP is probably not the only protein involved in positioning RNA polymerase II. This antibody recognize human TBP with MW 40 kDa and mouse/rat Tbp with MW 34 kDa.

Description: These dipeptide building blocks containing Ser- or Thr-derived oxazolidines (pseudoprolines) proved to be versatile tools for overcoming some intrinsic problems in the field of peptide chemistry. The presence of pseudoprolines within a peptide sequence results in the disruption of β-sheet structures considered as a source of intermolecular aggregation during chain elongation, thus increasing solvation and coupling kinetics in peptide assembly. Therefore, use of pseudoprolines offer new possibilities for accessing large peptides by convergent strategies and chemoselective ligation techniques. Moreover, incorporation of a pseudoproline unit facilitates cyclization of peptides.

Description: Programmed cell death 1 ligand 2 (PD-L2), also known as butyrophilin B7-DC or PDCD1 ligand 2, belongs to the member of B7 family which can regulate the activation and tolerance of T cells. PD-L2 is one ligand for Programmed cell death 1(PD-1), and the other is PD-L1. These two ligands shares 34% aa sequence identity. Mouse PD-L2 gene encodes a 273 amino acids (aa) protein with a putative 19 aa signal peptide, a 201 aa extracellular region , a 21 aa transmembrane domain and a 32 aa cytoplasmic region. The mouse PD-L2 gene is highly expressed in heart, placenta, pancreas, lung and liver while expressed weakly in spleen, lymph nodes and thymus. Besides, the expression of PD-L2 gene can be induced on dendritic cells grown from peripheral blood mononuclear cells under CSF2 and IL4/interleukin-4 treatment, and up-regulated by IFNG/IFN-gamma stimulation in monocytes. PD-L2 usually functions in a PDCD1-independent manner and is involved in regulating costimulatory signal which is essential for T-cell proliferation and IFNG production. Recent studies demonstrate that the expression of PD-L2 on the tumor cells promotes CD8 T cell–mediated rejection of tumor cells, at both the induction and effector phase of antitumor immunity. Moreover, PD-L2 binds to PD-1 cells and enhances T cell killing in a PD-1–independent mechanism.
