Description: The Hit3a monoclonal antibody specifically reacts with the ε chain of the CD3/T lymphocyte antigen receptor complex. The CD3 complex is part of the TCR complex, expressed by all mature T lymphocytes and by the thymocyte lineage. CD3 enhances the antigen recognition by signal transduction. The HIT3a antibody cross-links with the TCR complex, initiating the cellular activation and proliferation, but it cannot be used for intracellular CD3 staining, as it is able to stain only the surface CD3 complex.

Description: This antibody recognizes a glycoprotein of 110 kDa, which is identified as CD68. It is important for identifying macrophages in tissue sections. It stains macrophages in a wide variety of human tissues, including Kupffer cells and macrophages in the red pulp of the spleen, in lamina propria of the gut, in lung alveoli, and in bone marrow. It reacts with myeloid precursors and peripheral blood granulocytes. It also reacts with plasmacytoid T cells, which are supposed to be of monocyte/macrophage origin. It shows strong granular cytoplasmic staining of chronic and acute myeloid leukemia and also reacts with rare cases of true histiocytic neoplasia. Lymphomas are negative or show few granules.

Description: Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

Description: Actin is a major component of the cytoskeleton and is present in most cell types. It is highly specific to actin from smooth muscles. This MAb does not stain cardiac or skeletal muscle; however, it does stain myofibroblasts and myoepithelial cells. This antibody could be used together with anti-muscle specific actin and myogenin in making a diagnosis of smooth muscle and skeletal muscle tumors. In most cases of rhabdomyosarcoma, this antibody yields negative results whereas anti-muscle specific actin and myogenin are positive. Leiomyosarcomas are positive only with anti-muscle specific actin and anti-smooth muscle actin and are negative with anti-myogenin.

Description: Allophycocyanin (APC), highly fluorescent phycobiliprotein, is made up of alpha and beta subunits and is present as a trimer ( αβ)3, which is unstable and susceptible to dissociation at low concentrations. The monomer, αβ, has a lower fluorescence quantum yield compared to the trimer and the maximal absorption is also shifted to 620 nm. The chemically cross-linked APC trimer is much more stable than the native APC trimer, but still retains the same spectroscopic properties as the native APC trimer. APC labeled streptavidin, primary and secondary antibodies have been widely used in applications such as flow cytometry, live cell staining, and immunofluorescent staining.
