Description: Flow cytometric analysis of MA1-144-D650 shows positive staining of SSEA-5 in human embryonal carcinoma NCCIT cells. Stage-Specific Embryonic Antigen-5 (SSEA-5) is an oligosaccharide that can be part of glycosphingolipids and glycoproteins. It contains the motif Fuc1-2Gal-beta1-3GlcNAc-beta, which is characteristic of H-1 antigens. SSEA-5 is found in the inner cell mass of the human blastocyst during embryogenesis, on undifferentiated human embryonic stem cells and on the surface of induced pluripotent cells. SSEA-5 was found to proceed the loss of other known stem cell markers and the combination of SSEA-5 with either CD9 and CD90 or CD50 and CD200 was superior in identifying and eliminating rare undifferentiated human ES cells from differentiated cell populations than the use of other established stem cell markers.

Description: This antibody is predicted to react with bovine, mouse and zebrafish based on sequence homology. The TEK receptor tyrosine kinase is expressed almost exclusively in endothelial cells in mice, rats, and humans. This receptor possesses a unique extracellular domain containing 2 immunoglobulin-like loops separated by 3 epidermal growth factor-like repeats that are connected to 3 fibronectin type III-like repeats. The ligand for the receptor is angiopoietin-1. Defects in TEK are associated with inherited venous malformations; the TEK signaling pathway appears to be critical for endothelial cell-smooth muscle cell communication in venous morphogenesis. TEK is closely related to the TIE receptor tyrosine kinase.

Description: A suggested positive control is mouse small intestine tissue lysate. PA5-20455 can be used with blocking peptide PEP-0572. Z-DNA binding protein 1 (ZBP1) belongs to a family of proteins that contain the Zalpha domain which binds specifically to left-handed Z-DNA and Z-RNA. ZBP1 was initially identified as a novel gene that was up-regulated in activated macrophages in mice bearing ascites tumors, suggesting that is may play a role in processes such as host response in neoplasia. More recent reports indicate that the cytosolic ZBP1 has can act like the toll-like receptor TLR9 by detecting cytosolic double-stranded (ds) DNA and trigger induction of type I interferon and other innate immune responses. It is thought that the binding of ZBP1 to dsDNA enhances its association with innate immune response proteins such as the IRF3 transcription factor and the serine/threonine kinase TBK1 (also known as NAK). Multiple isoforms of ZBP1 are known to exist.

Description: It has been successfully used in flow cytometry applications on human samples. TRA-1-81 is a cell surface antigen expressed along with SSEA-3, SSEA-4 and TRA-1-60 in human embryonic stem cells, embryonal carcinoma cells and induced pluripotent stem cells (iPS). These surface markers are down-regulated during the differentiation process. In contrast, SSEA-1 is absent in undifferentiated human stem cells but is present on the cell surface after retinoic acid mediated differentiation.

Description: CYP2C19 is a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and is known to metabolize many xenobiotics, including the anticonvulsive drug mephenytoin, omeprazole, diazepam and some barbiturates. Polymorphism within its gene is associated with variable ability to metabolize mephenytoin, known as the poor metabolizer and extensive metabolizer phenotypes.

Description: This gene encodes a protein belonging to the syntrophin family. Syntrophins are cytoplasmic peripheral membrane proteins that bind to components of mechanosenstive sodium channels and the extreme carboxy-terminal domain of dystrophin and dystrophin-related proteins. The PDZ domain of this protein product interacts with a protein component of a mechanosensitive sodium channel that affects channel gating. Absence or reduction of this protein product has been associated with Duchenne muscular dystrophy. There is evidence of alternative splicing yet the full-length nature of these variants has not been described.

Description: This antibody is predicted to react with bovine and rat based on sequence homology. Parkinson's disease (PD) is a multifactorial disease that appears to arise from the effects of both genetic and environmental influences. The known genetic factors include multiple genes that have been identified in related parkinsonian syndromes, as well as alpha-synuclein. Genes associated with either PD or Parkinson-related disorders include parkin, DJ-1, ubiquitin C-terminal hydrolase isozyme L1 (UCH-L1), nuclear receptor-related factor 1 (NURR1), and alpha-synuclein. Nurr1 is a transcription factor that is expressed in the embryonic ventral midbrain and is critical for the development of dopamine (DA) neurons. It belongs to the conserved family of nuclear receptors but lacks an identified ligand and is therefore referred to as an orphan receptor. RXR ligands can promote the survival of DA neurons via a process that depends on Nurr1-RXR heterodimers. In developing DA cells, Nurr1 is required for the expression of several genes important for DA synthesis and function. Nurr1 is also important for the maintenance of adult DA neurons.

Description: Various biochemical, physiological and behavioural processes display circadian rhythms controlled by an internal biological clock. The central ""gears"" driving this clock appear to be composed of an autoregulatory transcription/posttranslation-based feedback loop. Cryptochrome 1 (CRY1) and 2 (CRY2) are DNA-binding flavoproteins that bear some homology to blue-light receptors and photolyases. In Drosophila, CRY is a photoreceptor for the circadian clock where it binds to the clock component TIM in a light-dependent fashion and blocks its function. Mammalian CRY1 and CRY2 function via light-independent interactions with circadian genes CLOCK and BMAL1, as well as with PER1, PER2, and TIM. They seem to act as light-independent components of the circadian clock and likely regulate Per1 transcriptional cycling via interactions with both the activator and its feedback inhibitors. Mutant mice not expressing the Cry1 or Cry2 protein display accelerated and delayed periodicity of locomotor activity, respectively. It appears that the combination of both proteins working together is essential to synchronize the organism to circadian phases. A critical balance between Cry1 and Cry2 is required for proper clock function; in complete darkness, double-mutant mice present with instantaneous arrhythmicity, indicating the absence of an internal circadian clock.

Description: MAGEB2 is a member of the MAGEB gene family. The members of this family have their entire coding sequences located in the last exon, and the encoded proteins show 50 to 68% sequence identity to each other. The promoters and first exons of the MAGEB genes show considerable variability, suggesting that the existence of this gene family enables the same function to be expressed under different transcriptional controls. This gene is localized in the DSS (dosage-sensitive sex reversal) critical region. It is expressed in testis and placenta, and in a significant fraction of tumors of various histological types. The MAGEB genes are clustered on chromosome Xp22-p21.

Description: The protein encoded by this gene is involved in the formation of the pre-replication complex that is necessary for DNA replication. The encoded protein can bind geminin, which prevents replication and may function to prevent this protein from initiating replication at inappropriate origins. Phosphorylation of this protein by cyclin A-dependent kinases results in degradation of the protein.

Description: This antibody is predicted to react with canine, human and rat based on sequence homology. This gene is a member of the Src family of protein tyrosine kinases . The encoded protein contains N-terminal sites for myristylation and palmitylation, a PTK domain, and SH2 and SH3 domains which are involved in mediating protein-protein interactions with phosphotyrosine-containing and proline-rich motifs, respectively. The protein localizes to plasma membrane ruffles, and functions as a negative regulator of cell migration and adhesion triggered by the beta-2 integrin signal transduction pathway. Infection with Epstein-Barr virus results in the overexpression of this gene. Multiple alternatively spliced variants, encoding the same protein, have been identified.
