Description: Proenkephalin-A is a secreted protein that belongs to the opioid neuropeptide precursor family. Proenkephalin-A is an endogenous opioid polypeptide hormone which, via proteolyic cleavage, produces the enkephalin peptides [Met]enkephalin, and to a lesser extent, [Leu]enkephalin. Met- and Leu-enkephalins compete with and mimic the effects of opiate drugs. They play a role in a number of physiologic functions, including pain perception and responses to stress. Proenkephalin-A (114-133) and Proenkephalin-A (237-258) increase glutamate release in the striatum. Proenkephalin-A (114-133) decreases GABA concentration in the striatum.

Description: ADAR1 converts adenosine to inosine in dsRNA, which destabilizes the dsRNA helix. This activity is important for various functions like site-specific RNA editing of transcripts of the glutamate receptors and modifying viral RNA genomes (which may be responsible for hypermutation of certain negative-stranded viruses, e.g., measles virus). ADAR1 also binds to short interfering RNAs (siRNA) without editing them and suppresses siRNA-mediated RNA interference. This protein is ubiquitously expressed, with the highest levels being found in brain and lung.

Description: The ion channels activated by glutamate are typically divided into two classes. Those that are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors (NMDAR). The NMDAR plays an essential role in memory, neuronal development and it has also been implicated in several disorders of the central nervous system including Alzheimer’s disease, epilepsy and ischemic neuronal cell death. Increased membrane surface expression of the NR1 subunit of the receptor has been associated with synaptic plasticity.

Description: The protein encoded by this gene is an auxiliary subunit of the ionotropic glutamate receptor of the AMPA subtype. AMPA receptors mediate fast synaptic neurotransmission in the central nervous system. This protein has been reported to interact with the Type I AMPA receptor regulatory protein isoform gamma-8 to control assembly of hippocampal AMPA receptor complexes, thereby modulating receptor gating and pharmacology. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Aug 2012].

Description: CSNK1G2 is a cytoplasmic protein which contains one protein kinase domain. It is amember of the CK1 Ser/Thr protein kinase family of the large casein kinase I (CKI) subfamily. CSNK1G2 participates in Wnt signaling, involves in brain development and vesicular trafficking and neurotransmitter releasing from small synaptic vesicles. It regulates fast synaptic transmission mediated by glutamate. SMAD3 phosphorylation promotes its ligand-dependent ubiquitination and subsequent proteasome degradation, thus inhibiting SMAD3-mediated TGF-beta responses.

Description: Group I of the Metabotropic Glutamate Receptors includes GRM1 and GRM5, receptors that have been shown to activate phospholipase C. Alternative splice variants of GRM5 have been described, but their full-length nature has not been determined. Inflammation results in the activation of mGluR1 and mGluR5 in dorsal horn neurons of the spinal cord, leading to extracellular signal-regulated kinase (ERK) activation, which is required for nociceptive plasticity and enhanced pain. GRM5 is involved in inflammatory mechanical hyperalgesia and potentially in schizophrenia

Description: N-methyl-D-aspartate (NMDA) receptors are a class of ionotropic glutamate-gated ion channels. These receptors have been shown to be involved in long-term potentiation, an activity-dependent increase in the efficiency of synaptic transmission thought to underlie certain kinds of memory and learning. NMDA receptor channels are heteromers composed of the key receptor subunit NMDAR1 (GRIN1) and 1 or more of the 4 NMDAR2 subunits: NMDAR2A (GRIN2A), NMDAR2B (GRIN2B), NMDAR2C (GRIN2C) and NMDAR2D (GRIN2D). Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2008]
