Description: The SensoLyte® Plus 520 MMP-2 Assay Kit is designed specifically for detecting MMP-2 activity in biological samples, such as culture medium, serum, plasma, synovial fluid, and tissue homogenate, which may contain multiple MMPs. A monoclonal anti-human-MMP-2 antibody is employed to pull down MMP-2 from the mixture first. MMP-2 activity is then quantified by a 5-FAM/QXL® 520 fluorescence resonance energy transfer (FRET) peptide. The fluorescence signal is monitored at Ex/Em=490 nm/520 nm upon MMP-2-induced cleavage of the FRET substrate. The long wavelength fluorescence of 5-FAM is less interfered by the autofluorescence of components in biological samples and test compounds. Ample materials are provided to perform 96 assays in a 96-well format.

Description: Cytokines for expansion and cell culture of human hematopoietic stem cells to progenitor cells. Hematopoietic stem cells are self renewing pluripotent cells often characterized by the expression of CD34. Studies have shown that decreased expression of CD34 tends to correlate with the decreased self renewal properties and pluripotent potential. Culturing CD34+ cells in a cytokine mixture of SCF, FLT-3 Ligand, IL-6 and IL-3 for short time periods allows rapid expansion of immature and more mature hematopoietic progenitor cells in cord blood or bone marrow preparations. If the goal is to only expand the most immature progenitor and stem cells, it is recommended that a combination of TPO, SCF and FLT-3 Ligand are used (Catalog: PB-500-03). Either cytokine combination can also be supplemented with other cytokines to promote specific lineages.

Description: VAP-1 is a type II membrane cell adhesion protein belonging to the copper/topaquinone oxidase family. It is primarily expressed on the high endothelial venules of peripheral lymph nodes and on hepatic endothelia. VAP-1 can catalyze the oxidative deamination of low molecular weight amines, and plays an important role in the migration of lymphocytes to inflamed tissue. Inhibition of VAP-1 can protect against inflammation-related damage to certain injured tissues. Additionally, VAP-1 can function as a significant prognostic marker for certain cancers and cardiovascular diseases. Recombinant Human VAP-1 is a mixture of monomeric and disulfide-linked homodimeric forms of a 737 amino acid polypeptide, corresponding to amino acids 27 to 763 of the VAP-1 precursor. The calculated molecular weight of Recombinant Human VAP-1 is 81.8 kDa.

Description: The GL-1 monoclonal antibody reacts specifically with the mouse B7-2 (also known as CD86), a costimulatory molecule expressed by B and T lymphocytes, macrophages (thioglycollate-induced from peritoneum), astrocytes and dendritic cells. The memory CD4+ T lymphocytes express CD86 (as mRNA and protein). CD86, and the B7-1 (CD 80) molecule, are ligands for CD152 and CD28, and influence the costimulatory interactions between lymphocytes B and T. B7-2 is also involved in the mouse natural killer cell-mediated cytotoxicity. The GL-1 antibody blocks the mixed lymphocyte reaction (MLR) and inhibits the T-cells stimulation by antigen-presenting cells. Mixtures of anti-B7-1 antibody and GL-1 were reported to inhibit the interaction between CD125 and its ligand with the in vivo priming of cytotoxic T lymphocytes.

Studies have shown that decreased expression of CD34 tends to correlate with decreased self renewal properties and pluripotent potential. Culturing CD34+ cells in a cytokine mixture of TPO, SCF, and FLT-3 Ligand for short time periods allows CD34+ cells to proliferate without decreasing CD34 expression, thus retaining their stem cell properties. The combination of these cytokines allows for the ex vivo expansion of the most immature hematopoietic stem cells in cord blood or bone marrow preparations, but can be supplemented with other cytokines to promote specific lineages. For example, GM-CSF and/or G-CSF can be added to promote monocytes and granulocytes while EPO can be added to promote erthroid cells.

Description: The genes encoding type I interferons (IFNs), which include 14 IFN-alphagenes (two of which are IFN-alpha1 and IFN-alpha13), one IFN-∫ gene, one IFN-w (also known as IFN-alpha II1) gene and a number of IFN-w pseudogenes, are clustered on human chromosome 9. IFN-alpha and -beta are cytokines that are widely known to induce potent antiviral activity. They exert a variety of other biological effects, including antitumor and immunomodulatory activities and are increasingly used clinically to treat a range of malignancies, myelodysplasias and autoimmune diseases. IFN-w is antigenically different from human IFN-alpha, IFN-beta or IFN-gamma, but is a component of natural mixtures of IFN species produced by virus-induced leukocytes or Burkitt's lymphoma cells. The type I interferon receptor (IFN-alphaR) interacts with IFN-alpha, IFN-beta and IFN-w, and seems to be a multisubunit receptor.
