Description: NAGA belongs to the glycosyl hydrolase 27 family. It removes terminal alpha-N-acetylgalactosamine residues from glycolipids and glycopeptides. It is required for the breakdown of glycolipids. Biosynthetic studies performed with cultured fibroblasts indicated that the human enzyme was synthesized as a 65kDa glycosylated precursor which was processed to a mature 48-kDa lysosomal form; both the precursor and mature forms had high mannose type oligosaccharide chains, but only the precursor’s mannose residues were phosphorylated. 90-117kd is a homodimer of NAGA.

Description: Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are moved about in the cell by two distinct but interconnected cycles by means of 300 kDa cation-independent mannose 6-phospate receptors (MPR). MPR cycles and transports newly synthesized lysosomal enzymes between the TGN and late endosomes / early lysosomes. It also cycles and transports extracellular lysosomal enzymes between the plasma membrane and early endosomes via clathrin coated vesicles. The bulk of the MPR protein is in the extracellular/lumenal domain. The entire pool of MPRs cycles between these cellular compartments every 3 hours. The steady state distribution of MPR's is predominantly within late endosomes, fewer in the trans Golgi network and ~5-10 % at the cell surface. In addition to its man6P binding activity, the MPR contains a separate binding site for the type II insulin-like growth factor and is capable of binding both man6P and IGF-II simultaneously. An ~240 kDa soluble, truncated form, representing the extracellular domain of the protein, has also been found circulating in serum and is capable of binding both ligands.

Supplier: Bioss
Description: Calcium-dependent lectin displaying mannose-binding specificity. Induces the formation of Birbeck granules (BGs); is a potent regulator of membrane superimposition and zippering. Binds to sulfated as well as mannosylated glycans, keratan sulfate (KS) and beta-glucans. Facilitates uptake of antigens and is involved in the routing and/or processing of antigen for presentation to T cells. Major receptor on primary Langerhans cells for Candida species, Saccharomyces species, and Malassezia furfur. Protects against human immunodeficiency virus-1 (HIV-1) infection. Binds to high-mannose structures present on the envelope glycoprotein which is followed by subsequent targeting of the virus to the Birbeck granules leading to its rapid degradation.

Supplier: Proteintech
Description: Rab9 GTPase is required for the transport of mannose 6-phosphate receptors from endosomes to the trans-Golgi network in living cells, and in an in vitro system that reconstitutes this process. P40 is an effector of Rab9 that interacts preferentially with the active form of Rab9. p40 does not interact with Rab7 or K-Ras; it also fails to bind Rab9 when it is bound to GDI. The protein is found in cytosol, yet a significant fraction (~30%) is associated with cellular membranes. P40 is a very potent transport factor in that the pure, recombinant protein can stimulate, significantly, an in vitro transport assay that measures transport of mannose 6-phosphate receptors from endosomes to the trans-Golgi network.

Description: RPN2 is a type I integral membrane protein found only in the rough endoplasmic reticulum. The protein is part of an N-oligosaccharyl transferase complex that links high mannose oligosaccharides to asparagine residues found in the Asn-X-Ser/Thr consensus motif of nascent polypeptide chains. This gene encodes a type I integral membrane protein found only in the rough endoplasmic reticulum. The encoded protein is part of an N-oligosaccharyl transferase complex that links high mannose oligosaccharides to asparagine residues found in the Asn-X-Ser/Thr consensus motif of nascent polypeptide chains. This protein is similar in sequence to the yeast oligosaccharyl transferase subunit SWP1.

Description: CD206, also named as MMR, CLEC13D and MRC1, is a type I membrane receptor that mediates the endocytosis of glycoproteins by macrophages. CD206 has been shown to bind high-mannose structures on the surface of potentially pathogenic viruses, bacteria, and fungi so that they can be neutralized by phagocytic engulfment. CD206 is a 170 kDa transmembrane protein which contains 5 domains: an amino-terminal cysteine-rich region, a fibronectin type II repeat, a series of eight tandem lectin-like carbohydrate recognition domains (responsible for the recognition of mannose and fucose), a transmembrane domain, and an intracellular carboxy-terminal tail. It is expressed on most tissue macrophages, in vitro derived dendritic cells, lymphatic and sinusoidal endothelia. This antibody recognizes the intracellular carboxy-terminal part of CD206 and MRC1L1.

Description: Collectin-11 is a secreted protein that belongs to the COLEC10/COLEC11 family. Collectin-11 contains one C-type lectin domain and one collagen-like domain. Collectins play important roles in the innate immune system by binding to carbohydrate antigens on microorganisms, facilitating their recognition and removal. Collectin-11 binds to various sugars including fucose and mannose, but does not bind to glucose, N-acetylglucosamine and N-acetylgalactosamine. It has a higher affinity for fucose compared to mannose. Collectin-11 binds lipopolysaccharides (LPS). It also involved in fundamental development serving as a guidance cue for neural crest cell migration. Defects in Collectin-11 are the cause of 3MC syndrome type 2 (3MC2).

Description: Mannose-specific lectin. May recognize sugar residues of glycoproteins, glycolipids, or glycosylphosphatidyl inositol anchors and may be involved in the sorting or recycling of proteins, lipids, or both. The LMAN1-MCFD2 complex forms a specific cargo receptor for the ER-to-Golgi transport of selected proteins.
