Description: Quenched fluorogenic substrate for cathepsin D (kcat/Km=1.09 x 10 M-1s-1) and cathepsin E (kcat/Km=1.56 x 10 M-1s-1). Mca fluorescence is quenched by the Dnp group until cleavage separates them. Activity of the two enzymes on this substrate can be distinguished by the use of Ascaris pepsin inhibitor, which inhibits cathepsin E, but not cathepsin D. This substrate is also cleaved by MMP-2, MMP-3, MMP-9, but not MMP-1 nor MMP-13. This substrate is useful for inhibitor screening and kinetic analysis. Ex.: 320-340 nm, Em.: 393-420 nm. Mca-Pro-Leu-OH (BML-P127) can be used to calibrate for Mca fluorescence.

Description: Tissue Inhibitor of Metalloproteinases 1 (TIMP-1) complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Also mediates erythropoiesis in vitro; but, unlike IL-3, it is species-specific, stimulating the growth and differentiation of only human and murine erythroid progenitors. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-11, MMP-12, MMP-13, and MMP-16. TIMP-1 does not act on MMP-14.

Description: TIMP-2 is an extracellular inhibitor of MMPs, including MMP-1, -2, -3, -7, -8, -9, -10, -12, -13, -14, -15, -16 and -19. It belongs to the I35 (TIMP) family of irreversible protease inhibitors that function as key modulators of extracellular matrix degradation during tissue development and remodeling. TIMP-2 can also act through a MMP-independent mechanism inhibiting endothelial cell proliferation in vitro and demonstrates anti-angiogenic activities Recombinant Human TIMP-2 is a 21.8 kDa protein containing 194 amino acid residues.

Description: Matrix Metalloproteinases (MMPs) are a large family of endopeptidases. Collectively, MMPs can degrade all kinds of extracellular matrix proteins, and can also process a number of bioactive molecules. They are known to be involved in the cleavage of cell surface receptors, the release of apoptotic ligands, and chemokine/cytokine inactivation. MMPs are also thought to play a major role in cell behaviors such as cell proliferation, migration (adhesion/dispersion), differentiation, angiogenesis, apoptosis, and host defense.
This substrate is readily hydrolyzed by MMP-8, 9 and collagenase-3 (MMP-13), Abs/Em = 325/393 nm. The hydrolysis is inhibited in a 1:1 stoichiometric fashion by the tissue inhibitors of metalloproteinases, TIMP-1, TIMP-2, and TIMP-3.
