Description: The three mammalian isoforms of TGF-β, TGF-β1, β2, and β3, signal through the same receptor and elicit similar biological responses. They are multifunctional cytokines that regulate cell proliferation, growth, differentiation and motility, as well as synthesis and deposition of the extracellular matrix. They are involved in various physiological processes, including embryogenesis, tissue remodeling and wound healing. They are secreted predominantly as latent complexes, which are stored at the cell surface and in the extracellular matrix. The release of the biologically active TGF-β isoform from a latent complex involves proteolytic processing of the complex and /or induction of conformational changes by proteins such as thrombospondin-1. TGF-β1 is the most abundant isoform secreted by almost every cell type. It was originally identified for its ability to induce phenotypic transformation of fibroblasts, and recently it has been implicated in the formation of skin tumors. Recombinant Human TGF-β1 is a 25.0 kDa protein composed of two identical 112 amino acid polypeptide chains linked by a single disulfide bond.

Description: Myostatin is a TGF-β family member that acts as an inhibitor of skeletal muscle growth. This muscle-specific cytokine interacts with Activin type I and type II receptors, and suppresses myoblast proliferation by arresting cell-cycle in the G1 phase. Suppression of myostatin activity facilitates muscle formation, and may be useful in reducing and/or preventing adiposity and type-2 diabetes. Myostatin activity can be blocked by the activin-binding protein follistatin, and by the propeptide of myostatin. The amino acid sequence of mature myostatin is extremely conserved across species, and is the same in murine, rat, chicken, turkey, porcine, and human. Myostatin is expressed as the C-terminal part of a precursor polypeptide, which also contains a short N-terminal signal sequence for secretion, and a propeptide of 243 amino acids. After dimerization of this precursor, the covalent bonds between the propeptide and the mature ligand are cleaved by furin-type proteases. However, the resulting two proteins remain associated through non-covalent interactions, and are secreted as a latent complex. Recombinant Human/Murine/Rat Myostatin is a 25.0 kDa protein consisting of two identical 109 amino acid polypeptides linked by a single disulfide bond.

Supplier: Prosci
Description: PDIA1 (protein disulfide isomerase family A member 1) is the beta subunit of prolyl 4-hydroxylase, a highly abundant multifunctional enzyme that belongs to the protein disulfide isomerase family. When present as a tetramer consisting of two alpha and two beta subunits, this enzyme is involved in hydroxylation of prolyl residues in preprocollagen. PDIA1 is also a disulfide isomerase containing two thioredoxin domains that catalyze the formation, breakage and rearrangement of disulfide bonds. Other known functions include its ability to act as a chaperone that inhibits aggregation of misfolded proteins in a concentration-dependent manner, its ability to bind thyroid hormone, its role in both the influx and efflux of S-nitrosothiol-bound nitric oxide, and its function as a subunit of the microsomal triglyceride transfer protein complex.

Description: Sarafotoxins have been purified and sequenced from the venom of the Israeli snake Atractaspis engaddensis. The sarafotoxins, which cause severe coronary vasospasm in snakebite victims, exhibit a remarkable structural and functional homology to the endothelin family. They bind with high affinity to rat atrial and brain membranes and induce the hydrolysis of phosphoinositides in these tissues.

Description: Bone morphogenetic proteins (BMPs) constitute a subfamily within the TGF-β superfamily of structurally related signaling proteins. Members of this superfamily are widely distributed throughout the body and are involved in diverse physiological processes during both pre- and postnatal life.  BMP-10 plays a crucial role in the development of the embryonic heart by acting to stimulate and maintain cardiomyocyte proliferation.  It can signal through various receptor complexes usually containing BMPR-1A, BMPR-1B, ALK1, ALK3, or ALK6. The interaction of BMP-10 with its specific receptors can induce signaling initiated by the phosphorylation of SMAD transcription factors, including SMAD1, SMAD5, or SMAD8, but can also induce SMAD independent processes.  BMP-10 is structurally related to BMP-9, and both can inhibit endothelial cell proliferation and migration. Recombinant Human BMP-10 is a 24.4 kDa homodimeric disulfide-linked protein consisting of two 108 amino acid subunits, which correspond to amino acid residues 317 to 424 of the full-length BMP-10 precursor.
