Description: Persephin is a disulfide-linked, homodimeric, neurotrophic factor structurally related to GDNF, artemin, and neurturin. These proteins belong to the cysteine knot family of growth factors that assume stable dimeric structures. Persephin signals through a multicomponent receptor system, composed of RET and one of four GFR α (α1-α4) receptors. The GFRα4 was first identified in chicken, and was later shown to be the preferential binding subunit for persephin. Persephin promotes the survival of ventral midbrain dopaminergic neurons and motor neurons after sciatic nerve oxotomy, and, like GDNF, promotes ureteric bud branching. However, in contrast to GDNF and neurturin, persephin does not support the survival of peripheral neurons. Recombinant Murine Persephin is a disulfide-linked homodimer, composed of two 10.3 kDa polypeptide chains (192 total amino acid residues). Each chain contains seven conserved cysteine residues, one of which (Cys 63) is used for inter-chain disulfide bridging, and the others are involved in the intramolecular ring formation known as the cysteine knot configuration.

Description: Catalyzes the last step in the transsulfuration pathway from methionine to cysteine. Has broad substrate specificity. Converts cystathionine to cysteine, ammonia and 2-oxobutanoate. Converts two cysteine molecules to lanthionine and hydrogen sulfide. Can also accept homocysteine as substrate. Specificity depends on the levels of the endogenous substrates. Generates the endogenous signaling molecule hydrogen sulfide (H2S), and so contributes to the regulation of blood pressure.Defects in CTH are the cause of cystathioninuria (CSTNU). It is an autosomal recessive phenotype characterized by abnormal accumulation of plasma cystathionine, leading to increased urinary excretion.

Description: E64d (EST), a membrane permeable analog of E64c, was expected to be useful as an oral drug for muscular dystrophy due to its potent inhibitory activities against the cysteine proteinases involved in the myofibrillar protein degradation that occurs in this disease. E64d attenuates the effects of parathyroid hormone on osteoblasts in vitro. By inhibiting cathepsin B, a cysteine protease showing β-secretase activity, E64d could reduce brain Aβ and improve memory deficits in Alzheimer's disease animal models.

Description: Peroxiredoxin (Prx) is an antioxidant enzyme detoxifying reactive oxygen species and has a cysteine at the active site. Prx enzymes modulate various receptor signaling pathways and protect cells from oxidatively induced death. Peroxiredoxin 1 to 4 have two conserved Cys residues corresponding to Cys51 and Cys172 of mammalian Peroxiredoxin 1. The active site cysteine(Cys51) is oxidized to cysteine sulfenic acid(Cys51-SOH) when a peroxide is reduced. Because Cys51-SOH is unstable, it forms a disulfide with Cys172-SH which comes from the other subunit of the homodimer. The disulfide is then reduced back to the Prx active thiol form by the thioredoxin-thioredoxin reductase system. However, the formation of the disulfide is a slow process. Thus under oxidative stress conditions, the sulfenic intermediate(Cys51-SOH) can be easily over oxidized to cysteine sulfinic acid(Cys-SO2H) or cysteine sulfonic acid(Cys-SO3H) before it is able to form a disulfide. Recent studies suggest that over oxidized Prx can be reduced back to the active form during recovery after oxidative stress.

Description: The cysteine-rich, adipose tissue-specific, secretory factor resistin (resistance to insulin, also known as ADSF) is a secreted hormone that potentially links obesity to diabetes. Resistin is rich in serine and cysteine residues and contains a unique cysteine repeat motif. Resistin and the resistin-like molecules share the characteristic cysteine composition and other signature features. Resistin-like a is a secreted protein that has restricted tissue distribution and is most highly expressed in adipose tissue. Another family member, Resistin-like b, is a secreted protein expressed only in the gastrointestinal tract, particularly in the colon, in both mouse and human. Resistin-like b expression is highest in proliferative epithelial cells and is markedly increased in tumors, suggesting a role in intestinal proliferation.

Description: I-309 is a CC chemokine that signals through the CCR8 receptor. It is secreted by T lymphocytes, monocytes and mast cells. I-309 chemoattracts monocytes and Th2 differentiated T-cells, but not neutrophils. Human I-309 is active on murine cells. Unlike some CC chemokines, whose biologically active form in solution is a non-disulfide-linked dimer, the biologically active form of I-309 is monomeric. Recombinant Human I-309 (CCL1) is an 8.5 kDa protein consisting of 74 amino acid residues that include six cysteine residues, of which four are the highly conserved cysteine residues present in CC chemokines.
