Description: Intracellular channel that mediates calcium release from the endoplasmic reticulum following stimulation by inositol 1,4,5-trisphosphate. Plays a role in ER stress-induced apoptosis. Cytoplasmic calcium released from the ER triggers apoptosis by the activation of CaM kinase II, eventually leading to the activation of downstream apoptosis pathways (By similarity).

Description: Grancalcin (GCA) is a cytoplasmic granule membrane protein that contains 4 EF-hand domains. GCA is calcium-binding protein and particularly abundant in human neutrophils. GCA is highly expressed in bone marrow, and it can be detected in neutrophils and macrophages. Calcium-binding protein GCA cooperates with SRI and LCP1, so it may play a role in the adhesion of neutrophils to fibronectin. GCA also may play a role in the formation of focal adhesions.

Description: Upon cell stimulation, the kinase DGKA converts the second messenger diacylglycerol into phophatidate, initiating the resynthesis of phosphatidylinositols and attenuating protein kinase C activity. DGKA is stimulated by calcium and phosphatidylserine, and is phosphorylated by protein kinase C. Tissue expression is in lymphocytes and oligodengroglial cells. DGKA contains 2 zinc-dependent phorbol-ester and DAG binding domains, and 2 EF-hand calcium binding domains.

Description: PTK2B encodes a cytoplasmic protein tyrosine kinase which is involved in calcium-induced regulation of ion channels and activation of the map kinase signaling pathway. The encoded protein may represent an important signaling intermediate between neuropeptide-activated receptors or neurotransmitters that increase calcium flux and the downstream signals that regulate neuronal activity. The encoded protein undergoes rapid tyrosine phosphorylation and activation in response to increases in the intracellular calcium concentration, nicotinic acetylcholine receptor activation, membrane depolarization, or protein kinase C activation. This protein has been shown to bind CRK-associated substrate, nephrocystin, GTPase regulator associated with FAK, and the SH2 domain of GRB2. The encoded protein is a member of the FAK subfamily of protein tyrosine kinases but lacks significant sequence similarity to kinases from other subfamilies.

Description: Calcium-binding chaperone that promotes folding, oligomeric assembly and quality control in the endoplasmic reticulum (ER) via the calreticulin/calnexin cycle. This lectin interacts transiently with almost all of the monoglucosylated glycoproteins that are synthesized in the ER. Interacts with the DNA-binding domain of NR3C1 and mediates its nuclear export. Involved in maternal gene expression regulation. May participate in oocyte maturation via the regulation of calcium homeostasis (By similarity).

Description: The EF-hand domain is a twelve amino acid loop motif that is commonly found in proteins that participate in calcium-binding events within the cell. EF-hand domains generally exist in a pair that, together, form a stable four-helix bundle that enables the binding of calcium ions. Swiprosin-2, also known as EFHD1 (EF-hand domain-containing protein D1), SWS2, PP3051 or MST133, is a 239 amino acid protein that contains two EF-hand domains and is expressed in a wide variety of tissues, including brain, liver, heart, kidney, testis, ovaries and spleen. Expression of Swiprosin-2, a possible calcium-binding protein, is upregulated during neuronal differentiation, suggesting a role for Swiprosin-2 in brain development and maturation.

Description: SS18-like 1(SS18L1) is a transcriptional activator that is required for calcium-dependent dendritic growth and branching in cortical neurons. It's also a nuclear protein interacts with CREB-binding protein and expressed in the developing brain. It helps regulate neuronal morphogenesis in calcuim-dependent manner. The N-terminal domain of SS18L1 is required for suppressing transactivation in the basal state, while the C-terminal domain is required for calcium-induced transactivation. It's part of the CREST-BRG1 complex, a multiprotein complex that regulates promoter activation by orchestrating a calcium-dependent release of a repressor complex and a recruitment of an activator complex. This complex also binds to the NR2B promoter, and activity-dependent induction of NR2B expression involves a release of HDAC1 and recruitment of CREBBP

Description: CAPN6 Antibody: Calpains make up a ubiquitously expressed, well-conserved family of calcium-dependent cysteine proteases. The calpain proteins are heterodimers consisting of an invariant small subunit and variable large subunits. This large subunit possesses a cysteine protease domain, and both subunits possess calcium-binding domains. Calpains have been implicated in neurodegenerative processes as their activation can be triggered by calcium influx and oxidative stress. Calpain 6 (CAPN6) is most similar to Calpain 5; the C-terminal region of CAPN6 lacks homology to the calmodulin-like domain of other vertebrate calpains. CAPN6 is thought to be involved in the regulation of microtubule dynamics and cytoskeletal organization. CAPN6 has also been recently identified as an HIV dependency factor (HDF), suggesting that CAPN6 may be an important drug target in HIV treatment.
