Description: The Dumpy-30 (DPY-30) protein was first described in C. elegans, in which it is involved in dosage compensation of sex chromosomes. Conserved from yeast to humans, the DPY-30 family is involved in gene expression and chromatin modification, specifically histone methylation. DPY-30 and closely related proteins contain a short motif that is related to the dimerization motif in the regulatory subunit of protein kinase A (PKA), which consists of two ?helices that form a four-helix bundle during dimerization. As a member of the DPY-30 family, DYDC1 (DPY30 domain-containing protein 1), also known as DPY30D1 and RSD9, is a 177 amino acid protein that binds to Endophilin III and plays a crucial role during acrosome biogenesis. DYDC1 is specifically expressed in brain and testis and accumulates in the acrosome area during spermatogenesis. Knockdown of DYDC1 mRNA results in disruption of acrosome formation and spermatid differentiation.

Description: SETDB1, also named as ESET, KIAA0067 and KMT1E, belongs to the histone-lysine methyltransferase family. It is a SET domain protein with histone H3-K9-specific methyltransferase activity. H3 'Lys-9' trimethylation is coordinated with DNA methylation and represents a specific tag for epigenetic transcriptional repression by recruiting HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones. SETDB1 mainly functions in euchromatin regions, thereby playing a central role in the silencing of euchromatic genes. It probably forms a complex with MBD1 and ATF7IP that represses transcription and couples DNA methylation and histone 'Lys-9' trimethylation. Its activity is dependent on MBD1 and is heritably maintained through DNA replication by being recruited by CAF-1. SETDB1 regulates histone methylation, gene silencing, and transcriptional repression. It has been identified as a target for treatment in Huntington Disease, given that gene silencing and transcription dysfunction likely play a role in the disease pathogenesis. This antibody is a rabbit polyclonal antibody raised against residues near the N terminus of human SETDB1.

Description: Dact3 Antibody: The Wnt signaling cascade is a conserved process in multicellular animals that plays important roles during development and can contribute to cancer and other diseases. Many members of this pathway are also expressed in the postnatal tissues such as brain. One such protein is Dact3, a member of the Dact protein family that was initially identified through binding to Disheveled (Dvl), a cytoplasmic protein essential to Wnt signaling. Dact3 is expressed in the ventral region of maturing somites, limb bud and branchial arch mesenchyme, embryonic CNS, and the adult brain. Recent evidence shows that Dact3 acts as a negative regulator Wnt/beta-catenin signaling that is repressed at the transcriptional level in colorectal cancer and this repression is associated with bivalent histone modifications. This repression can be reversed by pharmacological agents that targets both histone methylation and deacetylation, suggesting that Dact3 may be a potential target for therapeutic treatment of this cancer. At least three isoforms of Dact3 are known to exist.

Description: Cyclic nucleotide phosphodiesterases (PDEs) catalyze the hydrolysis of the 5-prime,3-prime-cyclic nucleotides cAMP and cGMP to the corresponding nucleoside 5-prime-monophosphates. Mammalian PDEs have been grouped into several families based on substrate affinities, inhibitor sensitivities, mode of regulation, and amino acid sequence homologies. The PDE8 family contains high-affinity cAMP-specific, IBMX (3-isobutyl-1-methyl-xanthine)-insensitive PDEs. All PDEs share a conserved C-terminal catalytic region and a variable N-terminal domain that presumably accounts for the distinctive regulatory properties unique to the individual families. PDE8B exhibits cAMP PDE activity which was not inhibited by various PDE inhibitors, including IBMX. The cAMP hydrolytic activity was unaffected by cGMP, and no cGMP PDE hydrolysis has been detected. Northern blot analysis indicated that the 4.2-kb PDE8B mRNA was expressed at the highest levels in thyroid, and at lower levels in brain, spinal cord, and placenta.

Description: ZBTB3 Antibody: The ZBTB family of proteins is comprised of diverse zinc finger proteins that also contain a BTB (BR-C, ttk and bab) domain. While little is known about ZBTB3, the related protein ZBTB2 is thought to be phosphorylated in response to the DNA damage, probably by either ATM or ATR. Other ZBTB proteins, such as ZBTB4 and ZBTB38 bind methylated DNA and repress transcription, suggesting that ZBTB3 may also act as a transcription repressor.

Description: mTOR controls cell growth, in part by regulating p70 S6 kinase alpha (p70alpha) and eukaryotic initiation factor 4E binding protein 1 (4EBP1). Raptor is a 150 kDa mTOR binding protein that also binds 4EBP1 and p70alpha. The binding of Raptor to mTOR is necessary for the mTOR-catalyzed phosphorylation of 4EBP1 in vitro, and it strongly enhances the mTOR kinase activity toward p70alpha. Rapamycin or amino acid withdrawal increases, whereas insulin strongly inhibits, the recovery of 4EBP1 and raptor on 7-methyl-GTP Sepharose. Partial inhibition of raptor expression by RNA interference (RNAi) reduces mTOR-catalyzed 4EBP1 phosphorylation in vitro. RNAi of C. elegans raptor yields an array of phenotypes that closely resemble those produced by inactivation of Ce-TOR. Thus, raptor is an essential scaffold for the mTOR-catalyzed phosphorylation of 4EBP1 and mediates TOR action in vivo.

Description: The WDR5 antibody targets the WD repeat protein domain 5 (WDR5), a member of the WD40 repeat protein family in humans. The WDR5 protein is a core member of the mammalian trithorax complex and, like the rest of the WD40 repeat protein family, is primarily involved in diverse cellular processes. WDR5 is identified as a methyl Lys-4 H3-specific-binding protein, however it is known to interact with the Mixed Lineage Leukemia (MLL) protein at the same site for histone H3 tri-methylation. Recently, the WDR5 protein has been shown to regulate embryonic stem cell self-renewal, and an increased expression of WDR5 has been found to efficiently induce the formation of pluripotent stem cells . WDR5 is also shown to accelerate osteoblast and chondrocyte differentiation.
