Description: GDNF is a glycosylated, disulfide-bonded homodimer molecule. It was first discovered as a potent survival factor for midbrain dopaminergic neurons and was then shown to rescue these neurons in animal models of Parkinson's disease. GDNF is about 100 times more efficient survival factor for spinal motor neurons than the neurotrophins. FUNCTION: Neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake. SUBUNIT: Homodimer; disulfide-linked. SUBCELLULAR LOCATION: Secreted protein. ALTERNATIVE PRODUCTS: 2 named isoforms produced by alternative splicing. DISEASE: Defects in GDNF may be a cause of Hirschsprung disease (HSCR). In association with mutations of RET gene, defects in GDNF may be involved in Hirschsprung disease. This genetic disorder of neural crest development is characterized by the absence of intramural ganglion cells in the hindgut, often resulting in intestinal obstruction. DISEASE: Defects in GDNF are a cause of congenital central hypoventilation syndrome (CCHS); also known as congenital failure of autonomic control or Ondine curse. CCHS is a rare disorder characterized by abnormal control of respiration in the absence of neuromuscular or lung disease, or an identifiable brain stem lesion. A deficiency in autonomic control of respiration results in inadequate or negligible ventilatory and arousal responses to hypercapnia and hypoxemia. SIMILARITY: Belongs to the TGF-beta family. GDNF subfamily.

Description: CD98 exits as a heterodimer containing a disulphide-linked glycosylated heavy chain and a non-glycosylated light chain. It is a member of the solute carrier family and encodes a cell surface, transmembrane protein. The protein exists as the heavy chain of a heterodimer, covalently bound through disulfide bonds to one of several possible light chains. The encoded transporter plays a role in regulation of intracellular calcium levels and transports L-type amino acids. Alternatively spliced transcript variants, encoding different isoforms, have been characterized.

Description: Laminin S binds to cells via a high affinity receptor, laminin is thought to mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. Laminin S is a complex glycoprotein, consisting of three different polypeptide chains (alpha, beta, gamma), which are bound to each other by disulfide bonds into a cross-shaped molecule comprising one long and three short arms with globules at each end. Beta 2 is a subunit of laminin 3 (Laminin S), laminin 4 (S merosin), and laminin 7 (KS laminin).

Description: The FN50 monoclonal antibody specifically reacts with human CD69, the 27-33 kDA type II transmembrane protein also known as the very early activation antigen (VEA) or the activation inducer molecule (AIM). It is expressed as a disulfide-linked dimer on B cells, T cells, NK cells, platelets, eosinophils, and neutrophils. It increases in expression upon cell activation and seems to serve a role as a signaling receptor.

Description: I-309 is a CC chemokine that signals through the CCR8 receptor. It is secreted by T lymphocytes, monocytes and mast cells. I-309 chemoattracts monocytes and Th2 differentiated T-cells, but not neutrophils. Human I-309 is active on murine cells. Unlike some CC chemokines, whose biologically active form in solution is a non-disulfide-linked dimer, the biologically active form of I-309 is monomeric. Recombinant Human I-309 (CCL1) is an 8.5 kDa protein consisting of 74 amino acid residues that include six cysteine residues, of which four are the highly conserved cysteine residues present in CC chemokines.

Description: Relaxin is a peptide hormone produced by the corpora lutea of ovaries during pregnancy in many mammalian species, including man. The secretion of the hormone into the blood stream just before parturition results in a marked softening and lengthening of the pubic symphysis and a softening of the cervix, which facilitates the birth process. By inhibiting uterine contractions, relaxin may influence the timing of parturition. Like insulin, relaxin consists of two peptide chains, A and B, covalently linked by disulfide bonds. By further analogy to insulin, the two peptides are synthesized as a single-chain precursor polypeptide with the B chain at the NH2-terminus.

Description: QSOX1(Sulfhydryl oxidase 1) catalyzes the oxidation of sulfhydryl groups in peptide and protein thiols to disulfides with the reduction of oxygen to hydrogen peroxide. It has two isoforms with the molecular weight of 67 kda and 83 kDa,respectively. It is detected a band of 95 Kda in HT1080 cell lines .Isoform 1 is expressed on golgi apparatus membrane and isoform 2 is secreted . The isoform1(QSOX1a) is the full length of QSOX1, approximately 78 kd; and the isoform2(QSOX1b) is the splicing form, approximately 62kd. QSOX1 is highly expressed in tumor cell lines but is not expressed in adjacent normal cells .

Description: Noggin belongs to a group of diffusible proteins that bind to ligands of the TGF-beta family, and regulate their activity by inhibiting their access to signaling receptors. The interplay between TGF-beta ligands and their natural antagonists has major biological significance during development processes, in which cellular response can vary considerably depending upon the local concentration of the signaling molecule. Noggin was originally identified as a BMP-4 antagonist whose action was critical for proper formation of the head and other dorsal structures. Consequently, noggin has been shown to modulate the activities of other BMPs including BMP-2,-7,-13, and -14. Targeted deletion of noggin in mice results in prenatal death, and a recessive phenotype displaying a severely malformed skeletal system. Conversely, transgenic mice over-expressing noggin in mature osteoblasts display impaired osteoblastic differentiation, reduced bone formation, and severe osteoporosis. Recombinant Human Noggin is a 46 kDa disulfide-linked homodimer consisting of two 205 amino acid polypeptide chains. Monomeric glycosylated noggin migrates at an apparent molecular weight of approximately 28.0-33.0 kDa by SDS PAGE analysis under reducing conditions.

Description: VEGF is a potent growth and angiogenic cytokine. It stimulates proliferation and survival of endothelial cells, and promotes angiogenesis and vascular permeability. Expressed in vascularized tissues, VEGF plays a prominent role in normal and pathological angiogenesis. VEGF signals through three receptors; fms-like tyrosine kinase (flt-1), KDR gene product (the murine homolog of KDR is the flk-1 gene product) and the flt4 gene product. Due to its increased acidity, VEGF121 circulates more freely than other VEGF forms, which bind more tightly with vascular heparin sulfates. Recombinant human VEGF121 is a 28.4 kDa disulfide-linked homodimeric protein consisting of two 121 amino acid polypeptide chains.
