The gMax MINI Kit allows the user to incubate tissue samples over-night, to make better use of valuable lab time. The gMax kit also includes Proteinase K powder that can be mixed with DI water and used during the cell lysing step for both tissue and blood samples. Chaotropic salt is used to lyse cells and degrade protein, allowing DNA to bind to the glass fiber matrix of the spin column. Contaminants are removed using a wash buffer (containing ethanol), and the purified genomic DNA is eluted by a low salt elution buffer or TE. The entire procedure can be completed in less than 30 minutes (after lysing) without phenol/chloroform extraction or alcohol precipitation, with an average DNA yield of 6µg from 200µl of of whole human blood. Purified DNA, with approximately 20-30Kb is suitable for use in PCR or other enzymatic reactions. The sample size for the gMax Kit is 50-200µl of whole blood (fresh or frozen), up to 25mg of tissue, up to 25mg of parafin-embedded tissue or buccal swab, and up to 15ml of amniotic fluid.
