Biotium's apoptosis/necrosis quantitation kits pair green fluorescent FITC-Annexin V conjugate with the membrane impermeant red fluorescent nucleic acid stain ethidium homodimer III


Green fluorescent CF™488A is brighter and more photostable than FITC, and unlike FITC, CF™488 fluorescence is not pH sensitive. Biotium offers CF™488A Annexin V conjugate in combination with propidium iodide or 7-AAD, membrane impermeant nucleic acid vital dyes that stain dead cell with red fluorescence. Propidium iodide is recommended for flow cytometry, while 7-AAD is recommended for fluorescence microscopy due to minimal fluorescence spill-over of 7-AAD in the green channel.

In the Apoptosis and Necrosis Quantitation Kit, FITC-Annexin V stains phosphatidylserine on the surface of apoptotic cells, while ethidium homodimer III stains necrotic and late apoptotic cells that have compromised plasma membrane integrity. Ethidium homodimer III also allows visualization of dead cell nuclear morphology to distinguish late apoptotic cells from necrotic cells. The Apoptotic, Necrotic and Healthy cells Quantitation Kit includes FITC-Annexin V, ethidium homodimer III, and Hoechst 33342 DNA dye, which stain the nuclei of all cells with blue fluorescence. The kits can be used for detection by microplate reader, flow cytometry, or fluorescence microscopy.
