G-Biosciences' FOCUS™ Protein Alkylation Kit contains an ultra pure, proteomic grade iodoacetamide reagent for blocking thiols by alkylation in order to prevent the reoxidation of proteins
G-Biosciences offers a proteomic grade Iodoacetamide in two different quantities: a 5 gram bulk supply or a OneQuant™ format (single aliquots of iodoacetamide that eliminate the need for weighing, preventing loss of reagent and saving time). For the OneQuant™ Iodoacetamide, add 150μL water to a single tube to generate a 0.5M iodoacetamide solution. To improve resolution and prevent streaking of protein spots on 2D gels, G-Biosciences offers an Agarose Sealing Solution and an Equilibration Buffer for IPG Strips. The Equilibration Buffer eliminates urea-induced protein carbamylation or waste of unused reagents, as fresh, ready-to-use reagents are prepared every time.
G-Biosciences' Proteomic Grade Water removes protein and dust contamination to improve the quality and reproducibility of 2D electrophoresis and mass spectrometry results. Offered in sets of 100 are certified protein- and dust-free Proteomic Grade Microfuge Tubes suitable for proteomic analysis and research. G-Biosciences also offers Proteomic Protein Controls for preparations specifically prepared for standardizing electrophoresis methods and protocols. Substantially free from non-protein agents such as nucleic acids, detergents, salts, lipids and other common laboratory agents, the protein preparation has low conductivity (< 50μS). Supplied as dry protein pellets, simply re-hydrate and use. Proteomic Protein Sample Controls are available for animal cells, E. coli, yeast, plant, or as a control set that contains one 2 mg vial of each.
Their FOCUS™ Protein Reductant is a water soluble, odorless, non-toxic and stable TCEP [Tris (2-carboxyethyl) phosphine] protein reductant with improved stability and efficiency compared to DTT. It reduces proteins over a wide range of pH, including lower acidic pH, and it completely reduces highly stable disulfide bonds in less than 5 minutes at room temperature. FOCUS™ Protein Reductant is compatible with the alkylation of thiol groups for 2D analysis, as it does not compete with the alkylation reagent iodoacetamide, unlike DTT and other commonly used reductants. G-Biosciences' FOCUS™ Reduction-Alkylation kit offers a simple two-step method for reduction and alkylation of protein samples for 2D gel analysis that improves resolution and prevents streaking of protein spots on 2D gels. The disulfide bonds are reduced with highly reactive and stable TCEP, followed by blocking of the thiols by alkylation with iodoacetamide reagent preparation. These kits are supplied with the proprietary buffers and reagents necessary for efficient reduction of disulfide bonds and efficient alkylation of thiols, while minimizing reoxidation of the competing thiol pairs in protein samples. G-Biosciences also offers FOCUS™ DTT, an ultra pure proteomic grade DTT, supplied in 5 vials containing dry powder to maintain the activity of the reducing agent. Add 1mL aliquot of water to give a 2M concentration, which is then added to your buffer solution to maintain protein reduction potential.
