Transfection Reagents

Electroporation-based methods have so far required cells to be in suspension for transfection. However, the 4D-Nucleofector® Y Unit allows direct Nucleofection of cells in adherence. Cells which typically grow in adherence in cell culture, can be kept and transfected by Nucleofection in their physiological state.


  • Nucleofection of neuronal cells at any time point during their culture period, i.e. at a later developmental stages
  • Nucleofection of cells in adherence without the need for transfer in suspension
  • Transfection efficiencies up to 70% combined with high viabilities


The AD1 primary cell 4D-Nucleofector® Y Kit is one of our two kits that are suited for electroporation of primary cells in adherence when working with the 4D-Nucleofector® Y Unit. The cells are cultured in the 24-well culture plates provided with the kit* and for the transfection step a 24-well dipping electrode array is inserted after replacing culture medium by the specific Nucleofector® Solution mixed with the DNA or RNA substrate.


This kit has been determined to be optimal for transfecting following cell types:
AoSMC (rat)
Dorsal root ganglia DRG (rat)
HUVEC (endothelial cells, umbilical vein, human)
Neurons (mouse), cortex
Neurons (rat), cortex
Neurons (rat), hippocampal
Osteoblast (rat)
PAEC (endothelial cells, pulmonary artery, human)
Retinal cells (rat)


Applications:
Electroporation of cells in adherence.
For transfection of 2.5×10⁴ to 1×10⁵ cells.

2×4.5 ml AD1 4D-Nucleofector® Y Solution.
2×1 ml Supplement 1.
3×150 μg pmaxGFP® Vector (1 μg/μl in 10 mM Tris pH 8.0).
1×24-well dipping electrode array.
1×Nunclon® Δ surface 24-well plate (Nunc).
