Description: Protein tags are protein or peptide sequences located either on the C- or N- terminal of the target protein, which facilitates one or several of the following characteristics: solubility, detection, purification, localization and expression. His tag is often used for affinity purification and binding assays. Expressed His-tagged proteins can be purified and detected easily because the string of histidine residues binds to several types of immobilized metal ions, including nickel, cobalt and copper, under specific buffer conditions. The His tag antibody is a useful tool for monitoring of the His-tagged proteins, and recognizes His-tags placed at N-terminal, C-terminal, and internal regions of fusion proteins expressed in bacteria, insect, and mammalian cells. This antibody is conjugated with HRP.
Note: Do not add Azium (Sodium Azide or Smite) into the dilution buffer. Azium is the HRP inhibitor which decreases the enzymatic activity of HRP.

Description: Focal adhesions are actin-rich structures that enable cells to adhere to the extracellular matrix and at which protein complexes involved in signal transduction assemble. Zyxin (ZYX) is a zinc-binding phosphoprotein that concentrates at focal adhesions and along the actin cytoskeleton. Zyxin has an N-terminal proline-rich domain and three LIM domains in its C-terminal half. The proline-rich domain may interact with SH3 domains of proteins involved in signal transduction pathways while the LIM domains are likely involved in protein-protein binding. Zyxin may function as a messenger in the signal transduction pathway that mediates adhesion-stimulated changes in gene expression and may modulate the cytoskeletal organization of actin bundles.

Description: SETDB1, also named as ESET, KIAA0067 and KMT1E, belongs to the histone-lysine methyltransferase family. It is a SET domain protein with histone H3-K9-specific methyltransferase activity. H3 'Lys-9' trimethylation is coordinated with DNA methylation and represents a specific tag for epigenetic transcriptional repression by recruiting HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones. SETDB1 mainly functions in euchromatin regions, thereby playing a central role in the silencing of euchromatic genes. It probably forms a complex with MBD1 and ATF7IP that represses transcription and couples DNA methylation and histone 'Lys-9' trimethylation. Its activity is dependent on MBD1 and is heritably maintained through DNA replication by being recruited by CAF-1. SETDB1 regulates histone methylation, gene silencing, and transcriptional repression. It has been identified as a target for treatment in Huntington Disease, given that gene silencing and transcription dysfunction likely play a role in the disease pathogenesis. This antibody is a rabbit polyclonal antibody raised against residues near the N terminus of human SETDB1.

Description: SIRT1, also named as SIR2L1, contains a deacetylase sirtuin-type domain and belongs to the sirtuin family. The post-translation modified SIRT1 is a 110 kDa protein, which contains one deacetylase sirtuin-type domain. The 75 kda SirT1 fragment was detected to lack the carboxy-terminus . SIRT1 may be found in nucleolus, nuclear euchromatin, heterochromatin and inner membrane. It can shuttles between nucleus and cytoplasm. SIRT1 regulates processes such as apoptosis and muscle differentiation by deacetylating key proteins. SIRT1 in particular initiates several signaling events relevant to cardioprotection, including: activation of endothelial nitric oxide synthase, insulin receptor signaling, and autophagy. In addition SIRT1 activation elicits resistance to oxidative stress via regulation of transcription factors and co-activators such as FOXO, Hif-2a, and NF-kB. SIRT1 regulates the p53-dependent DNA damage response pathway by binding to and deacetylating p53, specifically at Lysine 382. This antibody is a rabbit polyclonal antibody raised against residues near the N terminus of human SIRT1.
