Description: LFNG belongs to evolutionarily conserved glycosyltransferases that act in the Notch signaling pathway to define boundaries during embryonic development. While their genomic structure is distinct from other glycosyltransferases, fringe proteins have a fucose-specific beta-1,3-N-acetylglucosaminyltransferase activity that leads to elongation of O-linked fucose residues on Notch, which alters Notch signaling. This protein is predicted to be a single-pass type II Golgi membrane protein but it may also be secreted and proteolytically processed like the related proteins in mouse and Drosophila (PMID: 9187150).

Description: CD30 ligand (CD30L) is a type-II membrane-associated glycoprotein protein belonging to the TNF superfamily and is expressed primarily on certain B cells, T cells, and monocytes. CD30L binds specifically to CD30 (receptor), which is expressed on activated, but not resting, B and T cells, in lymphomas and various chronically inflamed tissues. CD30L/CD30 interactions initiate a signaling cascade that can ultimately lead to the activation of NF-KappaB. CD30L/CD30 signaling exerts pleiotropic effects on normal cells, including cell death, differentiation, or cell division. Certain diseases, including Hodgkins’s lymphoma, allergic inflammation, diabetes (in NOD mice), and mycobacterial infection can also be affected by CD30L/CD30 signaling. The CD30L gene encodes for a 234 amino acid type II transmembrane protein, which contains a 37 amino acid cytoplasmic sequence, a 25 amino acid transmembrane domain and a 172 amino acid extracellular domain. Recombinant human soluble CD30L (sCD30L) is a 188 amino acid polypeptide corresponding to the extracellular domain, and contains an 8 residue N-terminal His -Tag.

Description: PIAS1 Antibody: The PIAS proteins (protein inhibitor of activated STAT) play a crucial role as transcriptional coregulators in various cellular pathways, including the STAT, p53 and the steroid hormone signaling pathway. The PIAS protein family includes at least five evolutionarily conserved genes, including PIAS1. The major function of the PIAS proteins is the control of gene transcription and can also act as small ubiquitin-like-modifier (SUMO) E3 ligases. PIAS1 binds specifically to STAT1, inhibiting STAT1-mediated gene activation and also binds to the Gu/RNA helicase II enzyme, leading to the proteolytic cleavage of Gu/RH-II. PIAS1 is a potent co-activator for CP2c-mediated alpha-globin expression in erythroid cells.

Description: Translation initiation in eukaryotes necessitates the assembly of an 80S ribosomal complex. Eukaryotic initiation factors (eIFs) are utilized in a sequence of reactions that leads to 80S ribosomal assembly and initiation of translation. Mammalian eukaryotic translation initiation factor 4F (eIF4F) is a protein complex that contains eIF4A, eIF4E and eIF4G, binds mRNA at a 5-cap motif and recruits the 43S ribosomal preinitiation complex to the eligible transcript. Along with eIF4B, the eIF4F complex mediates the unwinding of mRNA secondary structure to facilitate ribosome association. eIF4E specifically interacts with the 5 cap, eIF4A(I,II) are bidirectional RNA helicases, and eIF4G(I,II) are scaffolding proteins which coordinate eIF4E, eIF4A, eIF3 and the 40S ribosome. Human eIF4AI (eIF4A, DDX2A) is a 406 amino acid protein that is 92.7% homologous to mouse eIF4AI. The promoter region of human eIF4A1 contains TATA and CAAT motifs and consensus binding sites to Sp1 and AP2.

Description: MARCH8 Antibody: MARCH8 (c-MIR) is a novel E3 ubiquitin ligase designated as the modulator of immune recognition (MIR) family, whose catalytic domain is a variant RING domain (RING-CH domain). MARCH8 was found as a functional and structural homolog of KSHV MIR1 and MIR2. MARCH8 targets B7-2 to lysosomal degradation and down-regulates the B7-2 surface expression through ubiquitination of its cytoplasmic tail. Furthermore, MARCH8 has been shown to down-regulate the expression of transferrin receptor and Fas, an important molecule for the induction of apoptosis. MARCH8 is the first example of an E3 ubiquitin ligase that is capable of inhibiting MHC II expression. Recent findings suggest that MARCH8 may regulate immune responses by promoting ubiquitination of MHC-II and CD86, leading to their subsequent endocytosis and lysosomal degradation.

Description: Spo11 is a type II topoisomerase that is thought to generate the chromosome breaks that initiate meiotic recombination. The Spo11 protein initiates meiotic recombination by generating DNA double-strand breaks (DSBs) and is required for meiotic synapsis in S. cerevisiae. The DSBs are located mostly in promoter regions, where the chromatin is in an open configuration, and cluster in domains along the chromosome. Expression of the Spo11 is detected mainly in the testis, in agreement with its predicted function in the initiation of meiotic recombination. Disruption of Spo11 leads to severe gonadal abnormalities from defective meiosis and results in infertility.

Description: CTSG(Cathepsin G) is also named as CTLA, CG, MGC23078, Catg and belongs to the peptidase S1 family. It may participate in the killing and digestion of engulfed pathogens, and in connective tissue remodeling at sites of inflammation. CTSG may also play a role in the progression to heart failure by activating angiotensin II, leading to detrimental effects on the heart. The full length protein contains 255 amino acid and has a singal peptide, a propeptide and a glycosylation site.

Description: HEAB is a 425 amino acid nuclear protein that belongs to the Clp1 family. Utilizing magnesium, manganese or nickel as cofactors, HEAB participates in the phosphorylation of the 5'-hydroxyl groups of double- and single- stranded RNA and DNA. HEAB is a member of the tRNA splicing endonuclease complex, in conjunction with TSEN2, TSEN15, TSEN34 and TSEN54, and is also a member of the pre-mRNA cleavage complex II. The gene encoding HEAB maps to human chromosome 11q12.1 and mouse chromosome 2 D; mutations to this gene may lead to a reduced pre-mRNA cleavage activity. HEAB exists as two isoforms due to alternative splicing events.

Description: OX40 ligand (OX40L), also called CD252, is a single-pass type II membrane protein of the TNF/TNF receptor superfamily. OX40L is expressed by DCs, macrophages and B cells and signals via its cognate receptor OX40 which is mainly expressed on APCs. OX40L/OX40 interactions are important in T-cell activation and survival and for the generation of memory T cells from activated effector T cells. OX40L–OX40 co-stimulation leads to activation of TNF receptor associated factor (TRAF) 2, 3 and 5. This pathway has been shown to prolong the survival of effector CD4+Th cells as well as contributes to generation of memory T cells.

Description: Cytokeratin 12 is a member of the intermediate filament family of proteins and is a heterotetramer of two type I and two type II keratins. Keratin 3 is specifically expressed in the corneal epithelium with family member KRT12. Cytokeratin 12 encodes the type I intermediate filament chain keratin 12, expressed in corneal epithelia. Defects in KRT3 and KRT12 are a cause of Meesmann corneal dystrophy (MCD), an autosomal dominant disease that causes fragility of the anterior corneal epithelium. Symptoms occur in adulthood and include rupture of the corneal microcysts that may lead to photophobia, contact lens intolerance and intermittent diminution of visual acuity. Defects in KRT12 are a cause of juvenile epithelial corneal dystrophy of Meesmann (MCD).
