You Searched For: Enzyme Assays

Either measuring substrate consumption or overall production, the enzyme assays are used to monitor enzymatic activity. Provided with the required supplies and reagents for any preferred method, enzyme kinetics and inhibition studies can be successfully completed. While continuous assay sampling methods give activity readings throughout development, discontinuous techniques calculate after the reaction has been stopped. The standardized enzyme assays will provide accurate identification and measurement of the protein molecules present in samples.

Catalog Number: (89156-496)
Supplier: Enzo Life Sciences
Description: It is a complete assay system designed to screen inhibitors of neutrophil elastase, providing enough material to perform at least 96 assays. Neutrophil elastase, also known as leukocyte elastase (LE), is a serine protease found in polymorphonuclear neutrophils (PMNs) that functions in inflammation. It can cleave a variety of proteins, including fibrous elastin, collagen, clotting factors, TNF-α, and α1-antichymotrypsin. It has been shown to be taken up by tumor cells and accelerate tumor growth through its degradation of insulin receptor substrate-1 (IRS-1). Because of its potential roles in cancer and inflammatory diseases such as emphysema, cystic fibrosis, and atherosclerosis, it is a target for drug discovery. The kit uses the chromogenic substrate MeOSuc-Ala-Ala-Pro-Val-pNA; cleavage of p-nitroaniline (pNA) from the substrate increases absorbance at 405nm. A neutrophil elastase inhibitor, elastatinal, is included for use as a control.

Catalog Number: (89156-526)
Supplier: Enzo Life Sciences
Description: A FLUOR DE LYS® fluorescent assay system

Supplier: Enzo Life Sciences
Description: BIOMOL Green™ provides a simple and convenient method for colorimetric phosphate quantitation (abs. 600-680 nm). Unlike other molybdate/malachite green-based assays, BIOMOL Green™ doesn’t require multiple solutions or reagents prepared fresh on the day of the assay. BIOMOL Green™ is extremely stable (>6 mos. at 4°C) and is simply mixed, at room temperature, with any enzymatic reaction that has released free phosphate. Reported applications include assays for phospholipid phosphatases, tyrosyl-tRNA synthetase (coupled with pyrophosphatase) and a viral RNA triphosphatase.BIOMOL Green™ has perhaps been most widely applied to protein phosphatase assays. Typically, a protein phosphatase (e.g. PTP1B or LAR) is simply incubated with an appropriate phosphopeptide (e.g. IR5,9,10). Addition of BIOMOL Green™ stops the reaction and begins color development, which is read 20-30 min. later.BIOMOL Green™ may be used in cuvette or microplate-based assays and is ideal for high-throughput applications. Each kit comes with a standardized phosphate solution for assay calibration.
